Carbonyl reductase 1 is a predominant doxorubicin reductase in the human liver.
2008
A first step in the enzymatic disposition of the antineoplastic drug doxorubicin (DOX) is the reduction to doxorubicinol (DOX-OL). Because DOX-OL is less antineoplastic but more cardiotoxic than the parent compound, the individual rate of this reaction may affect the antitumor effect and the risk of DOX-induced heart failure. Using purified enzymes and human tissues we determined enzymes generating DOX-OL and interindividual differences in their activities. Human tissues express at least two DOX-reducing enzymes. High-clearance organs (kidney, liver, and the gastrointestinal tract) express an enzyme with an apparent Km of approximately 140 microM. Of six enzymes found to reduce DOX, Km valu…
Generation of human pulmonary microvascular endothelial cell lines.
2001
The limited lifespan of human microvascular endothelial cells in cell culture represents a major obstacle for the study of microvascular pathobiology. To date, no endothelial cell line is available that demonstrates all of the fundamental characteristics of microvascular endothelial cells. We have generated endothelial cell lines from human pulmonary microvascular endothelial cells (HPMEC) isolated from adult donors. HPMEC were cotransfected with a plasmid encoding the catalytic component of telomerase (hTERT) and a plasmid encoding the simian virus 40 (SV40) large T antigen. Cells transfected with either plasmid alone had an extended lifespan, but the cultures eventually entered crisis aft…
Implanted neonatal human dermal fibroblasts influence the recruitment of endothelial cells in mice
2012
The vascularization of new tissue within a reasonable time is a crucial prerequisite for the success of different cell- and material-based strategies. Considering that angiogenesis is a multi-step process involving humoral and cellular regulatory components, only in vivo assays provide the adequate information about vessel formation and the recruitment of endothelial cells. The present study aimed to investigate if neonatal human dermal fibroblasts could influence in vivo neovascularization. Results obtained showed that fibroblasts were able to recruit endothelial cells to vascularize the implanted matrix, which was further colonized by murine functional blood vessels after one week. The ve…

Circulating CD34+ progenitor cells () gained importance in the field of regenerative medicine due to their potential to home in on injury sites and differentiate into cells of both endothelial and osteogenic lineages. In this study, we analyzed the mobilization kinetics and the numbers of CD34+, CD31+, CD45+, and CD133+ cells in twenty polytrauma patients (n = 13 male, n = 7 female, mean age 46.5±17.2 years, mean injury severity score (ISS) 35.8±12.5 points). In addition, the endothelial differentiation capacity of enriched CD34+cells was assessed by analyzing DiI-ac-LDL/lectin uptake, the expression of endothelial markers, and the morphological characteristics of these cells in Matrigel an…
Impaired in vivo vasculogenic potential of endothelial progenitor cells in comparison to human umbilical vein endothelial cells in a spheroid-based i…
2009
Objectives: Neovascularization represents a major challenge in tissue engineering applications since implantation of voluminous grafts without sufficient vascularity results in hypoxic cell death of implanted cells. An attractive therapeutic approach to overcome this is based on co-implantation of endothelial cells to create vascular networks. We have investigated the potential of human endothelial progenitor cells (EPC) to form functional blood vessels in vivo in direct comparison to vascular-derived endothelial cells, represented by human umbilical vein endothelial cells (HUVEC). Materials and methods: EPCs were isolated from human peripheral blood, expanded in vitro and analysed in vit…
ALMA spectral survey of Supernova 1987A – molecular inventory, chemistry, dynamics and explosive nucleosynthesis
2017
We report the first molecular line survey of Supernova 1987A in the millimetre wavelength range. In the ALMA 210--300 and 340--360 GHz spectra, we detected cold (20--170 K) CO, 28SiO, HCO+ and SO, with weaker lines of 29SiO from ejecta. This is the first identification of HCO+ and SO in a young supernova remnant. We find a dip in the J=6--5 and 5--4 SiO line profiles, suggesting that the ejecta morphology is likely elongated. The difference of the CO and SiO line profiles is consistent with hydrodynamic simulations, which show that Rayleigh-Taylor instabilities cause mixing of gas, with heavier elements much more disturbed, making more elongated structure. We obtained isotopologue ratios of…
The dynamic properties of neuronal chromatin are modulated by triiodothyronine.
1992
The effect of triiodothyronine (T3) on the rate of synthesis of nuclear proteins was studied during terminal differentiation of rat cortical neurons cultured in a serum-free medium. To this aim total and acid soluble nuclear proteins were analyzed by different electrophoretic techniques. Our results show that: 1) during maturation in vitro, neuronal nuclei undergo a dramatic change in the rate at which different classes of histones and high mobility group (HMG) proteins are synthesized; the synthetic activity, measured as incorporation of radioactive precursors into nuclear proteins, slows indeed down with age: especially evident is the decrease in core histones synthesis; at day 15, on the…
A multifuctional nanoplatform for drug targeted delivery based on radiation-engineered nanogels
2020
Abstract Under a rational design, combining biologically active molecules, ligands to specific cell receptors and fluorescent, radioactive or paramagnetic labels into a single nano-object can bridge the unique properties of the individual components and improve conventional sensing, imaging and therapeutic efficacies. The validation of these functional nano-objects requires careful testing both in terms of physico-chemical properties and biological behaviour in vitro and in vivo, prior to translation into the clinic. Ionising radiation of aqueous polymer solutions is a viable strategy to produce multifunctional nanogels from aqueous solutions of hydrophilic polymers. By proper selection of …
Non-radial pulsations in be stars. Preparation of the corot space mission
2006
The general objective of the present work is to contribute to the knowledge of the physics of Be Stars. In particular, we are interested in studying and characterizing their pulsational properties. A very suitable tool to reach this goal is the study and analysis of photometric time series with the maximum time baseline, density and photometric accuracy. The space mission COROT scheduled to be launched in December 2006, will provide ultra high precision, relative stellar photometry for very long continuous observing runs. Up to ten stars will be observed in the seismology fields with a photometric accuracy of a few 10-4 and color information during 150 days. The observations of Be stars with …
