Inhibition of gluconeogenesis by extracellular ATP in isolated rat hepatocytes.

1991

The aim of this study was to determine the effect of externally added ATP on gluconeogenesis by isolated hepatocytes from starved rats. High concentrations of extracellular ATP inhibited gluconeogenesis from lactate and pyruvate but not from glycerol or fructose. This inhibition was associated with an increase in intracellular adenosine contents. ADP, AMP, or adenosine but not guanosine 5'triphosphate, inosine 5' triphosphate, or adenine also inhibited gluconeogenesis. alpha, beta-Methylene-ATP, a nonmetabolizable structural analogue of ATP, did not affect the rate of gluconeogenesis. Intracellular ATP levels were increased by externally added ATP or adenosine, but ATP-to-ADP ratios in the…

Lipolysis in isolated epididymal adipocytes from genetically obese Zucker rat treated with 3,5,3'-L-triiodothyronine.

1990

Abstract. Glycerol released by isolated epididymal adipocytes was measured in 6-8 weeks old genetically obese (fa/fa) and lean (C.c.a/-) male Zucker rats, ip treated or not treated with T3 (0.46, 2.3 and 4.6 nmol/100 g in a single injection or 4.6 nmol · (100 g)−1 · day−1 for 5 days). In the non-treated rats, lipolysis was less stimulated by adrenalin or isoproterenol in obese than in lean rats, whereas it was stimulated by theophylline up to the same level in both kinds of rats. A single injection of the T3 (highest dose) increased isoproterenol-induced lipolysis in the lean but not in the obese rats. The 5-day T3 treatment caused a stimulation of the adrenalin- or isoproterenol-induced li…

Comparative study of the lubricant performance of Compritol® HD5 ATO and Compritol® 888 ATO: effect of polyethylene glycol behenate on lubricant capa…

2003

The aim of this paper is to study the lubricant capacity of Compritol HD5 ATO, a glyceryl and polyethylene glycol dibehenate, obtained by atomization. This material is compared to Compritol 888 ATO, constituted only by glyceryl dibehenate. First, this study verifies that Compritol HD5 ATO and Compritol 888 ATO present the same granular characteristics and that their mixes with Lactopress present no structural differences. Secondly, in term of compressibility and cohesiveness, the use of Compritol 888 ATO or Compritol HD5 ATO with Lactopress does not involve any significant modification. Finally, the minor difference of lubricant capacity between Compritol HD5 ATO and Compritol 888 ATO has n…

Hybrid Organic-Inorganic Nanostructures Fabricated from Layer-by-Layer Self-Assembled Multilayers of Hyperbranched Polyglycerols and Phosphorus Dendr…

2007

Multilayer thin films of cationic phosphorous dendrimers and anionic hyperbranched polyglycerols were fabricated by electrostatic layer-by-layer (LbL) self-assembly (SA). The film formation was monitored by surface plasmon resonance (SPR) spectroscopy and UV-visible spectroscopy, and it was found that the stepwise, alternating deposition results in a linear growth up to four bilayers. Hybrid organic-TiO2 nanostructures were generated by exposing the supramolecular multilayers to TiCl4 precursors. The amounts of TiO2 incorporated inside the scaffolds could be tuned by controlling the porosity of the multilayers with the addition of a small amount of salts. The resulting hybrid films exhibit …

Stable, hydroxyl functional polycarbonates with glycerol side chains synthesized from CO(2) and isopropylidene(glyceryl glycidyl ether).

2012

A series of functional polycarbonates, poly((isopropylidene glyceryl glycidyl ether)-co-(glycidyl methyl ether) carbonate) (P((IGG-co-GME) C)) random copolymers with different fractions of 1,2-isopropylidene glyceryl glycidyl ether (IGG) units, is synthesized. After acidic hydrolysis of the acetal protecting groups, a new type of functional polycarbonate prepared directly from CO(2) and glycerol is obtained, namely poly((glyceryl glycerol)-co-(glycidyl methyl ether) carbonate) (P((GG-co-GME) C)). All hydroxyl functional samples exhibit monomodal molecular weight distributions with PDIs between 2.5 and 3.3 and M(n) between 12 000 and 25 000 g mol(-1) . Thermal properties reflect the amorphou…

Effect of mode of operation, substrate and final electron acceptor on single-chamber membraneless microbial fuel cell operating with a mixed community

2018

Abstract Waste minimization and circular thinking are to be achieved in order to cope with the limited amount of resources of our planet. In this perspective, bio-electrochemical systems (BESs) can contribute to the global balance with their ability to extract chemical residual energy from wastewater and transform it directly into electrical current. BESs development has been limited by the cost connected to reactor design, in which membranes and cathode catalyst constituted a major drawback. In this paper we report the optimization process of a simple reactor without membranes or precious catalyst that produced 47.1 mW m−2, which is more than what achieved with configurations including mem…

Characterization of a novel open reading frame, urf a, in the mitochondrial genome of fission yeast: correlation of urf a mutations with a mitochondr…

1991

Between the genes for tRNA(gin) and tRNA(ile) an open reading frame of 227 amino acids has been identified which is unique among known mitochondrial genomes and which has been termed urf a (Lang et al. 1983; Kornrumpf et al. 1984). It uses the "mitochondrial" genetic code, i.e., it contains a TGA codon, whereas all other protein-encoding genes, and all but one intronic open reading frame, use the "standard" genetic code (UGG for tryptophan). A previous paper has demonstrated that "mutator" strains show an increased formation of mitochondrial drug-resistant and respiration-deficient mutants (including deletions). In this paper we show that the mutator activity is correlated with mutations in…

Model-based biotechnological potential analysis of Kluyveromyces marxianus central metabolism

2016

Abstract The non-conventional yeast Kluyveromyces marxianus is an emerging industrial producer for many biotechnological processes. Here, we show the application of a biomass-linked stoichiometric model of central metabolism that is experimentally validated, and mass and charge balanced for assessing the carbon conversion efficiency of wild type and modified K. marxianus. Pairs of substrates (lactose, glucose, inulin, xylose) and products (ethanol, acetate, lactate, glycerol, ethyl acetate, succinate, glutamate, phenylethanol and phenylalanine) are examined by various modelling and optimisation methods. Our model reveals the organism’s potential for industrial application and metabolic engi…

The GlpF residue Trp219 is part of an amino-acid cluster crucial for aquaglyceroporin oligomerization and function

2018

The vestibule loop regions of aquaglyceroporins are involved in accumulation of glycerol inside the channel pore. Even though most loop regions do not show high sequence similarity among aquaglyceroporins, loop E is highly conserved in aquaglyceroporins, but not in members of the homologous aquaporins. Specifically, a tryptophan residue is extremely conserved within this loop. We have investigated the role of this residue (Trp219) that deeply protrudes into the protein and potentially interacts with adjacent loops, using the E. coli aqualgyeroporin GlpF as a model. Replacement of Trp219 affects the activity of GlpF and impairs the stability of the tetrameric protein. Furthermore, we have id…

Squaric Acid Mediated Synthesis and Biological Activity of a Library of Linear and Hyperbranched Poly(Glycerol)-Protein Conjugates

2012

Polymer-protein conjugates generated from side chain functional synthetic polymers are attractive because they can be easily further modified with, for example, labeling groups or targeting ligands. The residue specific modification of proteins with side chain functional synthetic polymers using the traditional coupling strategies may be compromised due to the nonorthogonality of the side-chain and chain-end functional groups of the synthetic polymer, which may lead to side reactions. This study explores the feasibility of the squaric acid diethyl ester mediated coupling as an amine selective, hydroxyl tolerant, and hydrolysis insensitive route for the preparation of side-chain functional, …
