In vivoanalysis of the lumenal binding protein (BiP) reveals multiple functions of its ATPase domain

International audience; The endoplasmic reticulum (ER) chaperone binding protein (BiP) binds exposed hydrophobic regions of misfolded proteins. Cycles of ATP hydrolysis and nucleotide exchange on the ATPase domain were shown to regulate the function of the ligand-binding domain in vitro. Here we show that ATPase mutants of BiP with defective ATP-hydrolysis (T46G) or ATP-binding (G235D) caused permanent association with a model ligand, but also interfered with the production of secretory, but not cytosolic, proteins in vivo. Furthermore, the negative effect of BiP(T46G) on secretory protein synthesis was rescued by increased levels of wild-type BiP, whereas the G235D mutation was dominant. U…

Ecosystem carbon response of an Arctic peatland to simulated permafrost thaw

Permafrost peatlands are biogeochemical hot spots in the Arctic as they store vast amounts of carbon. Permafrost thaw could release part of these long-term immobile carbon stocks as the greenhouse gases (GHGs) carbon dioxide (CO 2 ) and methane (CH 4 ) to the atmosphere, but how much, at which time-span and as which gaseous carbon species is still highly uncertain. Here we assess the effect of permafrost thaw on GHG dynamics under different moisture and vegetation scenarios in a permafrost peatland. Vegetation structure and greenness in Central Africa from Modis multi-temporal data.

African forests within the Congo Basin are generally mapped at regional scale as broad-leaved evergreen forests, with a main distinction between terra-firme and swamp forests types. At the same time, commercial forest inventories, as well as national maps, have highlighted a strong spatial heterogeneity of forest types. A detailed vegetation map generated using consistent methods is needed to inform decision makers about spatial forest organisation and theirs relationships with environmental drivers in the context of global change. Novel Insights Into Gas Embolism in Sea Turtles: First Description in Three New Species

The recent finding of gas embolism and decompression sickness in loggerhead sea turtles (Caretta caretta) in the Mediterranean Sea challenged the conventional understanding of marine vertebrate diving physiology. Additionally, it brought to light a previously unknown source of mortality associated with fisheries bycatch for this vulnerable species. This paper examines international cooperation on technological development as an alternative to international cooperation on GHG emission reductions. In order to analyze the scope of cooperation, a three-stage technology agreement formation game is solved. First, countries decide whether or not to sign up to the agreement. Novel Autotrophic Organisms Contribute Significantly to the Internal Carbon Cycling Potential of a Boreal Lake

ABSTRACT Oxygen-stratified lakes are typical for the boreal zone and also a major source of greenhouse gas emissions in the region. Due to shallow light penetration, restricting the growth of phototrophic organisms, and large allochthonous organic carbon inputs from the catchment area, the lake metabolism is expected to be dominated by heterotrophic organisms. In this study, we test this assumption and show that the potential for autotrophic carbon fixation and internal carbon cycling is high throughout the water column.
