Functional C1-inhibitor diagnostics in hereditary angioedema: Assay evaluation and recommendations

2008

Hereditary angioedema (HAE) is an autosomal dominant disease characterized by recurrent episodes of potentially life-threatening angioedema. The most widespread underlying genetic deficiency is a heterozygous deficiency of the serine protease inhibitor Cl esterase inhibitor (C1-Inh). In addition to low C4 levels, the most important laboratory parameter for correct diagnosis of HAE or angioedema due to acquired C1-Inh deficiency is reduced C1-Inh function (fC1-Inh). No direct recommendations about the assays for fC1-Inh or sample handling conditions are available, although this would prove especially useful when a laboratory first starts to offer assays on fC1-Inh for HAE diagnosis. In the p…

Etude de la régulation transcriptionnelle des lymphocytes T CD4 dans un contexte de cancer : application en immunothérapie anticancéreuse

2015

Immune surveillance of tumors is based on the ability of effector cells of the immune system to detect and eliminate the cancer cells. Notwithstanding, the complete and spontaneous regression of established cancers was observed only in very few cases. The failure of cancer resolution by the immune system could result from the combination of several factors: i) inadequate immune response related to a low tumor immunogenicity, ii) incompetent immune system consecutively to induced or acquired immunodeficiencies and iii) the selection of resistant tumor variants able to thwart immune surveillance or subverting immune responses. Developing novel cancer immunotherapy strategies leading to potent…

HSP70 sequestration by free α-globin promotes ineffective erythropoiesis in β-thalassaemia

2014

International audience; β-Thalassaemia major (β-TM) is an inherited haemoglobinopathy caused by a quantitative defect in the synthesis of β-globin chains of haemoglobin, leading to the accumulation of free α-globin chains that form toxic aggregates. Despite extensive knowledge of the molecular defects causing β-TM, little is known of the mechanisms responsible for the ineffective erythropoiesis observed in the condition, which is characterized by accelerated erythroid differentiation, maturation arrest and apoptosis at the polychromatophilic stage. We have previously demonstrated that normal human erythroid maturation requires a transient activation of caspase-3 at the later stages of matur…

Epidemiological approach of type 2 myocardial infarction : Etiologies, characteristics, treatment and prognosis.

2020

Introduction: Type 2 Myocardial infarction (MI)has been recently defined as an imbalance between oxygen supply and demand, in the absence of atherothromthrombosis. This work aimed to describe the main etiolgies as well as epidemiological, clinical and prognostic characteristics.Method: Data from patients with type 2 MI were collected from the RICO cohort (Observatoire des Infarctus de Cote d'Or). In a complementary work, we analyzed the retrospective data of the emergency department of Dijon University Hospital. Results: Among 4,436 consecutive patients hospitalized for MI in Dijon emergency department over 3 years, 947 (21%) had type 2 MI (median age: 81 years). In the RICO cohort, 4,572 co…

Effect anti inflammatory and anti obesity of the polyphenolic extracts of carob leaves "Ceratonia siliqua" and cladode "Opuntia ficus-indica"

2018

In the present study, we have investigated the effects of polyphenol-rich infusions from carob leaves and OFI-cladodes on inflammation associated with obesity and dextran sulfate sodium (DSS)-induced ulcerative colitis in Swiss mice. In vitro studies revealed that aqueous extracts of carob leaves and OFI-cladodes exhibited anti-inflammatory properties marked by the inhibition of , TNF-α and nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells concomitant with NF-κβ nucleus translocation inhibition. For in vivo investigations, Swiss male mice were subjected to control or high fat diet (HFD). At the 4th week after the start of study, animals received or not 1% i…

Preventive effects of n-3 polyunsaturated fatty acids in alteration of colon mucus layer and reticulum endoplasmic stress induced by an obesogenic di…

2020

Obesity is now recognized as a real global epidemic affecting more than 650 million people worldwide. This chronic inflammatory disease is accompanied with intestinal alteration, including alteration of the colic mucus layer and an increase in stress of the endoplasmic colic reticulum, associated with microbiota dysbiosis. Polyunsaturated fatty acids in n-3 are known for their anti-inflammatory and immuno-modulatory properties and appear to be a promising lead in the fight against metabolic and intestinal alterations induced by obesity. The objectives of this work was initially to determine whether tissue enrichment in n-3, in the fat-1 mouse model, prevent alterations of colon mucus layer in…

Acyl-CoA oxidase 1 (ACOX1) deficiency is a rare and severe peroxisomal leukodystrophy associated with a very long-chain fatty acids β-oxidation defect. As peroxisomal defects in microglia appear to be a key component of physiopathogenesis in ACOX1 deficiency, we used the murine BV-2 microglial cell line as model: (i) to evaluate the antioxidant and anti-inflammatory properties of Opuntia ficus-indica cactus cladode extracts; (ii) to characterize a new BV-2 ACOX1 deficient cell line recently generated in our laboratory by CRISPR/Cas9 gene editing. In the first part of this work, the microglia activation, achieved by BV-2 cells exposition to four structurally and biologically well-defined lip…

Chirurgie cardiaque sous circulation extra-corporelle et ses biomarqueurs : rôle du Growth / Différentiation Factor 15 (GDF 15) : études cliniques

2016

Ischemic cardiac diseases are the most frequent and deleterious pathologies leading to important cardiovascular-related mortality worldwide. One of the alternative therapies consists to treat these patients using cardiac surgery. Cardiopulmonary bypass was developed to greatly improve this surgical procedure. However, some adverse effects can occur during cardiac surgery associated with cardiopulmonary bypass due to the inflammatory response. This phenomenon is the result of various mechanisms including oxidative stress and inflammatory cytokines which lead to multi-organ failure and then to myocardial and renal injuries occurring during the peri- and post-operative periods. The first part o…
