In vitro pharmacological evaluation of the dichloromethanol extract fromSchinus molle l.

The pharmacological activity of the dichloromethanol extract of Schinus molle L. (SM-DCM) was analysed in in vitro models. Preincubation of the isolated guinea-pig ileum or rat uterus preparations with the extract (100 μg/mL) abolished the contractile effects of histamine and serotonin respectively. At the same dose, the extract partially reduced the contractile effects of acetylcholine on the isolated rat duodenum. A 10 μg/mL dose showed an inhibitory effect on histamine and serotonin, but not on acetylcholine-induced contractions (NS). No significant effect was found with a 1 μg/mL dose. © 1998 John Wiley & Sons, Ltd.

The inhibitor/activator and substrate properties of enantiomers of two methylated histamines (MH) were investigated using a histamine methyltransferase preparation which was purified 1207-fold from pig fundic mucosa by ultracentrifugation, ion-exchange chromatography on DEAE-cellulose and preparative electrofocusing. In 1-100 microM concentrations, S-alpha-MH and R-alpha-MH were acceptor substrates as good as histamine itself. When substrate concentrations were increased to 1 mM these substances were methylated to an even greater extent than histamine, since they did not exert substrate inhibition on HMT. Histamine release events were shown in a prospective randomized controlled trial in patients undergoing elective general surgery with an extraordinarily high incidence: 73 per cent. Comparison of roxatidine and ranitidine in the treatment of refractory duodenal ulcer

Abstract Fourteen patients with duodenal ulcer not healed after treatment with ranitidine at the standard dosage of 300 mg/day for 12 weeks were enrolled in the study. Eight patients were treated with roxatidine acetate, 150 mg/day, for 8 weeks (group A); six patients received treatment with ranitidine, 600 mg/day, for 8 weeks (group B). Efficacy of treatment was evaluated on the basis of reduction of ulcer pain, lowering of maximal acid output (MAO) levels, and cicatrization of ulcer lesion. The patients treated with roxatidine showed a significant reduction in MAO levels after 4 weeks, whereas the patients treated with ranitidine showed a significant reduction in MAO levels after 8 weeks.…

Selective phosphodiesterase 4 (PDE4) inhibitors are of potential interest in the treatment of asthma. We examined the effects of the alkaloid S-(+)-glaucine, a PDE4 inhibitor, on human isolated bronchus and granulocyte function. Glaucine selectively inhibited PDE4 from human bronchus and polymorphonuclear leukocytes (PMN) in a non-competitive manner (Ki=3.4 μM). Glaucine displaced [3H]-rolipram from its high-affinity binding sites in rat brain cortex membranes (IC50∼100 μM). Glaucine inhibited the spontaneous and histamine-induced tone in human isolated bronchus (pD2∼4.5). Globularia alypumL. extracts reduced histamine and serotonin contractionin vitro

The present study analyses in in vitro models the pharmacological activity of methanol and dichloromethane extracts (1, 10 and 100 microg/mL) obtained from the leaves and stems of Globularia alypum L. Preincubation of the guinea-pig ileum and rat uterus with both extracts produced a dose dependent abolition of the contractile effects of histamine and serotonin, respectively. At the same doses, neither methanol nor dichloromethane extract reduced the contractile effects of acetylcholine on rat duodenum or noradrenaline on rat vas deferens.
