Mapping and structure of DMXL1, a human homologue of the DmX gene from Drosophila melanogaster coding for a WD repeat protein.
2000
The DmX gene was recently isolated from the X chromosome of Drosophila melanogaster. TBLASTN searches of the dbEST databases revealed sequences with a high level of similarity to DmX in a variety of different species, including insects, nematodes, and mammals showing that DmX is an evolutionarily highly conserved gene. Here we describe the cloning of the cDNA and the chromosomal localization of one of the human homologues of DmX, Dmx-like 1 (DMXL1). The human DMXL1 gene codes for a large mRNA of 11 kb with an open reading frame of 3027 amino acids. The putative protein belongs to the superfamily of WD repeat proteins, which have mostly regulatory functions. The DMXL1 protein contains an exc…
Report of a european collaborative exercise comparing DNA typing results using a single locus VNTR probe
1991
A collaborative exercise was carried out in 1989 among 12 European forensic laboratories using the single locus VNTR probe pYNH24, the restriction enzyme HinfI, the same set of human genomic DNA samples, and a standardized DNA size marker. The objectives of the exercise were: (1) to study the degree of variation within and between laboratories, (2) to obtain information on requirements for technical standardization allowing the exchange of typing results and (3) to compare different approaches for the identification of allelic DNA fragments of unknown size. Each laboratory carried out up to 10 independent typing experiments using the same DNA samples. The results were analysed independently…
Inhibition of expression of natural UAG suppressor glutamine tRNA in HIV-infected human H9 cells in vitro by Avarol.
1988
HTLV-IIIB-infected H9 cells are shown to contain a high level of the natural UAG suppressor glutamine tRNA(UmUG Gln); this tRNA has been demonstrated to be required for the synthesis of Moloney murine leukemia virus (Mo-MuLV)-encoded protease. After cultivation of HTLV-IIIB-infected H9 cells with Avarol at a concentration (1 microgram/ml), previously found to protect the cells against the cytopathic effects of HTLV-III, an almost complete inhibition of the synthesis of the tRNA(UmUG Gln) was observed. Moreover, we obtained some evidence that the processing of the HTLV-III precursor protein p53 to p24 is inhibited by Avarol in infected cells, suggesting that the compound interferes with the …
PGC-1α signaling coordinates susceptibility to metabolic and oxidative injury in the inner retina.
2013
Retinal ganglion cells (RGCs), used as a common model of central nervous system injury, are particularly vulnerable to metabolic and oxidative damage. However, molecular mechanisms underlying this sensitivity have not been determined in vivo . PGC-1α (encoded by PPARGC1A ) regulates adaptive metabolism and oxidative stress responses in a tissue- and cell-specific manner. Aberrant PGC-1α signaling is implicated in neurodegeneration, but the mechanism underlying its role in central nervous system injury remains unclear. We provide evidence from a mouse model that PGC-1α expression and activity are induced in adult retina in response to metabolic and oxidative challenge. Deletion of Ppargc1a d…
Distribution of the bilbo Non-LTR Retrotransposon in Drosophilidae and its Evolution in the Drosophila obscura Species Group
2001
The bilbo element is a non-LTR retrotransposon isolated from Drosophila subobscura. We conducted a distribution survey by Southern blot for 52 species of the family Drosophilidae, mainly from the obscura and melanogaster groups. Most of the analyzed species bear sequences homologous to bilbo from D. subobscura. In the obscura group, species from the same species subgroup also share similar Southern blot patterns. To investigate the phylogenetic relationship among these elements, we analyzed eight copies of a short sequence of the element from several species of the obscura group. The obtained phylogram agrees with the phylogeny of the species, which suggests vertical transmission of the ele…
Differential display reverse transcription PCR reveals IL-1 induced gene expression patterns in human articular chondrocytes
1995
IL-1 exerts diverse effects on the metabolism of articular chondrocytes, including inhibition of proteoglycan synthesis and stimulation of matrix metallopro teinase synthesis. Therefore it is believed that IL-1 might play an important role in cartilage degradation in osteo- and rheumatoid arthritis. To improve our understanding of IL-1 induced effects on overall gene expression patterns of human articular chondrocytes, wc used a novel mRNA fingerprinting technique: Differential Display Reverse Transcription-PCR (DDRT-PCR) (Liang and Pardee 1992). The reported high sensitivity of this powerful technique promised to enable work with human articular cartilage, a tissue from which only small am…
Interspecific hybridisation among diverse Saccharomyces species: A combined biotechnological solution for low-temperature and nitrogen-limited wine f…
2019
Lack of the prezygotic barrier in the Saccharomyces genus facilitates the construction of artificial interspecific hybrids among different Saccharomyces species. Hybrids that maintain the interesting features of parental strains have been applied in industry for many beneficial purposes. Two of the most important problems faced by wine makers is nitrogen deficiency in grape must and low-temperature fermentation. In our study, hybrids were constructed by using selected low nitrogen-demanding cryotolerant S. eubayanus, S. uvarum strains and S. cerevisiae. The fermentation capacity of the hybrid strains was tested under four conditions by combining two temperatures, 12 °C and 28 °C, and two ni…
Gone before its known? Camera-trapping shows alarming levels of putative hybrids in the wildcat (Felis silvestris) population of the Gargano National…
2022
Abstract The European wildcat is a small carnivore widespread across Europe and hybridization with the domestic cat is one of the major threats to wildcats. We estimate the population density of wild-living cats (both wildcats and putative hybrids based on pelage) in the Gargano National Park (Southern Italy). We sampled 20 stations for 540 camera days. We obtained 23 images from 17 detections at 10 cameras, and we identified 10 individuals (6 wildcats and 4 putative hybrids). Population density was estimated at 0.34 ± 0.15 SE wild-living cats/km2. The proportion of putative hybrids indicates an alarming hybridization for this population.
Physiological and genomic characterisation of Saccharomyces cerevisiae hybrids with improved fermentation performance and mannoprotein release capaci…
2015
Yeast mannoproteins contribute to several aspects of wine quality by protecting wine against protein haze, reducing astringency, retaining aroma compounds and stimulating lactic-acid bacteria growth. The selection of a yeast strain that simultaneously overproduces mannoproteins and presents good fermentative characteristics is a difficult task. In this work, a Saccharomyces cerevisiae × S. cerevisiae hybrid bearing the two oenologically relevant features was constructed. According to the genomic characterisation of the hybrids, different copy numbers of some genes probably related with these physiological features were detected. The hybrid shared not only a similar copy number of genes SPR1…
Short-term effect of ammonia concentration and salinity on activity of ammonia oxidizing bacteria.
2010
A continuously aerated SHARON (single reactor high activity ammonia removal over nitrite) system has been operated to achieve partial nitritation. Two sets of batch experiments were carried out to study the effect of ammonia concentration and salinity on the activity of ammonia-oxidizing bacteria (AOB). Activity of AOB raised as free ammonia concentration was increased reaching its maximum value at 4.5 mg NH 3 -N l −1 . The half saturation constant for free ammonia was determined (K NH 3 = 0.32 mg NH 3 -N l −1 ). Activity decreased at TAN (total ammonium–nitrogen) concentration over 2,000 mg NH 4 -N l −1 . No free ammonia inhibition was detected. The effect of salinity was studied by adding…
