New Biologically Active Triterpene-Saponins fromRandia dumetorum

Two new triterpene-saponins, 3-O-[O-beta-D-glucopyranosyl-(1----4)-O-beta-D-glucopyranosyl-(1----3)-( beta- D-glucuronopyranosyl)]oleanolic acid (1), 3-O-[O-beta-D-glucopyranosyl-(1----6)-O-beta-D-glucopyranosyl- (1----3)-(beta-D-glucuronopyranosyl)]oleanolic acid (2) together with five known saponins (3-7) were isolated from the methanolic extract of the fruits of Randia dumetorum (Retz) Lam. (Rubiaceae). Their structures were established on the basis of chemical and spectral data. The compounds 1, 3, 4, 5 were found to enhance significantly the proliferation of human lymphocytes in vitro. The crude saponin fraction showed haemolytic, molluscicidal, and immunostimulating activities.

The structure and interactions of proteins play a critical role in determining the quality attributes of many foods, beverages, and pharmaceutical products. Incorporating a multiscale understanding of the structure–function relationships of proteins can provide greater insight into, and control of, the relevant processes at play. Mutants of Saccharomyces cerevisiae cell division cycle defective in cytokinesis. Biosynthesis of the cell wall and morphology

The four temperature-sensitive mutants of Saccharomyces cerevisiae in the cell division cycle defective in cytokinesis (cdc, 3, 10, 11 and 12), have been analyzed with respect to the biosynthesis of the cell wall polymers. After 3 hours of incubation at the non-permissive temperature (37 degrees C) these strains stop growing. The synthesis of glucan, mannan and chitin (wall polymers) level off in a similar time, but glucan, mannan and chitin synthases remained active for at least 4 hours. If the mutants are analyzed by transmission and scanning electron microscopy different pictures emerge. SIRT1 prevents genotoxic stress-induced p53 activation in acute myeloid leukemia

SIRT1 is an important regulator of cellular stress response and genomic integrity. Its role in tumorigenesis is controversial. Whereas sirtuin 1 (SIRT1) can act as a tumor suppressor in some solid tumors, increased expression has been demonstrated in many cancers, including hematologic malignancies. In chronic myeloid leukemia, SIRT1 promoted leukemia development, and targeting SIRT1 sensitized chronic myeloid leukemia progenitors to tyrosine kinase inhibitor treatment. Abstract Methanol dried over powdered 4 A molecular sieves can be used for a selective mono-de-O-acetylation of the phenolic acetyl group of the per-O-acetyl protected brasilicardin A carbohydrate side chain. This reaction opens a practical procedure for a synthetic access to derivates of the immunosuppressive and cytotoxic natural product brasilicardin A.

The expression, inducibility, and regulation of four different cytochrome (cyt.) P-450 isoenzymes (PB1, PB2, MC1, and MC2) NADPH-cytochrome P-450 reductase, the glutathione transferases (GSTs) B and C and microsomal epoxide hydrolase (mEHb) have been studied during nitrosamine-induced hepatocarcinogenesis using immunohistochemical techniques. The investigations revealed basic differences in the expression of the individual drug metabolizing enzymes in the course of neoplastic development. Modulation of mutagenicity by phosphorylation of mutagen-metabolizing enzymes.

In this Minireview, we discuss our findings on phosphorylation of cytochromes P450 (CYP) and influence of this modification on metabolic toxification and/or detoxification of a variety of mutagens. We show that phosphorylation drastically interferes with the mutagenicity of several classes of compounds which are of high human relevance (cytostatic drugs of the cyclophosphamide type, aromatic amines/amides, and nitrosamines).
