Stromal Interaction Molecule 1 (STIM1) Is Involved in the Regulation of Mitochondrial Shape and Bioenergetics and Plays a Role in Oxidative Stress
2012
Calcium ions are involved in a plethora of cellular functions including cell death and mitochondrial energy metabolism. Store-operated Ca(2+) entry over the plasma membrane is activated by depletion of intracellular Ca(2+) stores and is mediated by the sensor STIM1 and the channel ORAI1. We compared cell death susceptibility to oxidative stress in STIM1 knock-out and ORAI1 knockdown mouse embryonic fibroblasts and in knock-out cells with reconstituted wild type and dominant active STIM1. We show that STIM1 and ORAI1 deficiency renders cells more susceptible to oxidative stress, which can be rescued by STIM1 and ORAI1 overexpression. STIM1 knock-out mitochondria are tubular, have a higher Ca…
Molecular basis of early epithelial response to streptococcal exotoxin: role of STIM1 and Orai1 proteins
2011
Streptolysin O (SLO) is a cholesterol-dependent cytolysin (CDC) from Streptococcus pyogenes. SLO induces diverse types of Ca(2+) signalling in host cells which play a key role in membrane repair and cell fate determination. The mechanisms behind SLO-induced Ca(2+) signalling remain poorly understood. Here, we show that in NCI-H441 cells, wild-type SLO as well as non-pore-forming mutant induces long-lasting intracellular Ca(2+) oscillations via IP(3) -mediated depletion of intracellular stores and activation of store-operated Ca(2+) (SOC) entry. SLO-induced activation of SOC entry was confirmed by Ca(2+) add-back experiments, pharmacologically and by overexpression as well as silencing of ST…
Multidimensional Health Modelling: Association between Socioeconomic Factors and Health in Latvia
2012
This paper proposes new approach for modelling self-assessed health. We find that the concept of health is too complicated to measure effects of health determinants using a one-dimensional econometric model. We apply two-dimensional stereotype logistic model that allows capturing nonmonotonicity in effects of factors and revealing significant effects that remain unrevealed if single dimension models, such as ordered logit or ordered probit, are used. Modelling self-assessed health using multi-dimensional stereotype logit provides higher model goodness of fit and quality measures in comparison to ordered probit model. Multi-dimensional stereotype logit is applied to estimate association betw…
Association Between Self-Assessed Health and Attitude Towards Own Health
2014
This paper explores association between health and attitude towards own health in two dimensions – taking care of own health and lifestyle. We apply two-dimensional stereotype logit model to estimate association between self-assessed health and attitude towards health, after accounting for socioeconomic factors. We find evidence of strong positive association between health status and (perceived) taking care of own health and lifestyle. Analysis of perception of the two concepts – "taking care of own health" and "healthy lifestyle" – provides insights into possible reasons of not very good indicators of health behaviour among Latvian population.
IFI16 expression is related to selected transcription factors during B-cell differentiation
2015
The interferon-inducible DNA sensor IFI16 is involved in the modulation of cellular survival, proliferation, and differentiation. In the hematopoietic system, IFI16 is consistently expressed in the CD34+ stem cells and in peripheral blood lymphocytes; however, little is known regarding its regulation during maturation of B- and T-cells. We explored the role of IFI16 in normal B-cell subsets by analysing its expression and relationship with the major transcription factors involved in germinal center (GC) development and plasma-cell (PC) maturation.IFI16mRNA was differentially expressed in B-cell subsets with significant decrease inIFI16mRNA in GC and PCs with respect to naïve and memory subs…
School Tracking and Mental Health
2019
We examine the effects of a comprehensive school reform on mental health. The reform postponed the tracking of students into vocational and academic schools from age 11 to age 16. The reform was implemented gradually across Finnish municipalities between 1972 and 1977. We use difference-in-differences variation and administrative data. Our results show that there is no discernible effect on mental health related hospitalizations on average even though the effect is precisely estimated. Heterogeneity analysis shows that, after the reform, females from highly-educated families were more likely to be hospitalized for depression. nonPeerReviewed
