Design and performance evaluation of a high power density EMI filter for PWM inverter-fed induction motor drives

2015

This paper presents the design of an electromagnetic interference (EMI) filter for a low voltage high current induction motor drives supplied by DC power grids, such as those used in several vehicle applications. In order to effectively design the EMI filter a suitable Common Mode/Differential Mode (CM/DM) separation technique has been used. In consideration of the high operating currents, a software based separation technique using time domain measurements has been applied. The proposed technique allows the CM and DM sections of the EMI filter to be properly selected in a more economical way, i.e. without the need of dedicated hardware or costly radio frequency (RF) instrumentation. The de…

Combined inhibition of Bcl-2 and NFκB synergistically induces cell death in cutaneous T-cell lymphoma.

2019

Abstract Therapeutic options for cutaneous T-cell lymphoma (CTCL) are limited and curative treatment regimens are not available. Thus, new targeted and well-tolerated therapeutic approaches are urgently needed. In this respect, we have recently shown that dimethyl fumerate (DMF) inhibits NF-κB acting as a survival factor in CTCL. Similarly, inhibition of the antiapoptotic protein B-cell lymphoma 2 (Bcl-2) has been shown to induce cell death in CTCL especially when combined with histone deacetylase inhibitors. Therefore, we hypothesized that inhibition of Bcl-2 should potentiate NF-κB inhibition in a novel combination treatment of CTCL. We show that, in vitro, the Bcl-2 inhibitors ABT-199 an…

Prohibitin, an essential protein for Colorado potato beetle larval viability, is relevant to Bacillus thuringiensis Cry3Aa toxicity

2013

Bacillus thuringienesis (Bt) Cry toxins constitute the most extensively used environmentally safe biopesticide and their mode of action relies on the interaction of the toxins with membrane proteins in the midgut of susceptible insects that mediate toxicity and insect specificity. Therefore, identification of Bt Cry toxin interacting proteins in the midgut of target insects and understanding their role in toxicity is of great interest to exploit their insecticidal action. Using ligand blot, we demonstrated that Bt Cry3Aa toxin bound to a 30kDa protein in Colorado potato beetle (CPB) larval midgut membrane, identified by sequence homology as prohibitin-1 protein. Prohibitins comprise a highl…

Integrative genomic and proteomic analyses identify targets for Lkb1 deficient metastatic lung tumors

2010

SummaryIn mice, Lkb1 deletion and activation of KrasG12D results in lung tumors with a high penetrance of lymph node and distant metastases. We analyzed these primary and metastatic de novo lung cancers with integrated genomic and proteomic profiles, and have identified gene and phosphoprotein signatures associated with Lkb1 loss and progression to invasive and metastatic lung tumors. These studies revealed that SRC is activated in Lkb1-deficient primary and metastatic lung tumors, and that the combined inhibition of SRC, PI3K, and MEK1/2 resulted in synergistic tumor regression. These studies demonstrate that integrated genomic and proteomic analyses can be used to identify signaling pathw…

SIK2 orchestrates actin-dependent host response upon Salmonella infection

2021

Significance Through conducting quantitative proteomics upon Salmonella infection, we identified a SIK2 signaling network, implementing the kinase into a so far concealed biological function. Our data exposed SIK2 as a central orchestrator of an actin regulatory network, coordinating the stability of Salmonella-containing vacuole (SCV) and cellular actin assembly, in order to limit the acute phase of the infection. Most strikingly, SIK2 is not exclusively acting locally on actin assembly associated with the SCV but impacts the actin cytoskeleton architecture in its entirety upon Salmonella infection. Our work provides a mechanistic framework for how the actin cytoskeleton is regulated and h…

CiliaCarta: An integrated and validated compendium of ciliary genes

2019

The cilium is an essential organelle at the surface of mammalian cells whose dysfunction causes a wide range of genetic diseases collectively called ciliopathies. The current rate at which new ciliopathy genes are identified suggests that many ciliary components remain undiscovered. We generated and rigorously analyzed genomic, proteomic, transcriptomic and evolutionary data and systematically integrated these using Bayesian statistics into a predictive score for ciliary function. This resulted in 285 candidate ciliary genes. We generated independent experimental evidence of ciliary associations for 24 out of 36 analyzed candidate proteins using multiple cell and animal model systems (mouse…

Judging the contact-times of multiple objects: Evidence for asymmetric interference.

2009

The accuracy of time-to-contact (TTC) judgments for single approaching objects is well researched, however, close to nothing is known about our ability to make simultaneous TTC judgments for two or more objects. Such complex judgments are required in many everyday situations, for instance when crossing a multi-lane street or when engaged in multi-player ball games. We used a prediction-motion paradigm in which participants simultaneously estimated the absolute TTC of two objects, and compared the performance to a standard single-object condition. Results showed that the order of arrival of the two objects determined the accuracy of the TTC estimates: Estimation of the first-arriving object …

Pterostilbene-induced tumor cytotoxicity: a lysosomal membrane permeabilization-dependent mechanism.

2012

The phenolic phytoalexin resveratrol is well known for its health-promoting and anticancer properties. Its potential benefits are, however, limited due to its low bioavailability. Pterostilbene, a natural dimethoxylated analog of resveratrol, presents higher anticancer activity than resveratrol. The mechanisms by which this polyphenol acts against cancer cells are, however, unclear. Here, we show that pterostilbene effectively inhibits cancer cell growth and stimulates apoptosis and autophagosome accumulation in cancer cells of various origins. However, these mechanisms are not determinant in cell demise. Pterostilbene promotes cancer cell death via a mechanism involving lysosomal membrane …

Optimization of fluorescence enhancement for silicon-based microarrays

2008

An optical technique for the enhancement of fluorescence detection sensitivity on planar samples is presented. Such a technique is based on the simultaneous optimization of excitation and light collection by properly combining interference and reflectance from the sample holder. Comparative tests have been performed in microarray applications, by evaluating the proposed solution against commercial glass-based devices, using popular labeling dyes, such as Cy3 and Cy5. The proposed technique is implemented on a substrate built with standard silicon technology and is therefore well suited for integrated micro total analysis systems (microTAS) applications.
