Insights on the origin of catalysis on glycine N-methyltransferase from computational modeling.

The origin of enzyme catalysis remains a question of debate despite much intense study. We report a QM/MM theoretical study of the SN2 methyl transfer reaction catalyzed by a glycine N-methyltransferase (GNMT) and three mutants to test whether recent experimental observations of rate-constant reductions and variations in inverse secondary α-3H kinetic isotope effects (KIEs) should be attributed to changes in the methyl donor−acceptor distance (DAD): is catalysis due to a compression effect? Semiempirical (AM1) and DFT (M06-2X) methods were used to describe the QM subset of atoms, while OPLS-AA and TIP3P classical force fields were used for the protein and water molecules, respectively. The lithium isotope response to the variable weathering of soils in Iceland

Abstract This study has analysed Li isotopes ratios from well-studied soil and pore water profiles from Iceland that have the same parent material but have experienced different degrees of chemical weathering. Thus, from least to most weathered, we have analysed vitrosols (V), gleyic andosols (GA), brown andosols (BA), Histosols (H) and Histic Andosols (HA). Although the most weathered H and HA soils have the highest content in clay-sized material, they have the least fractionated δ7Lipore water values. High resolution Fourier-transform spectroscopy data of term values in the spin-orbit (SO) coupled first excited A 1Σ+ and b 3Π states in KCs were obtained from (4)1Σ+ → A 1Σ+ − b 3Π, A 1Σ+ − b 3Π → X1Σ+, and (1)3Δ1→bΠ0±3 spectra of laser-induced fluorescence (LIF). About 3000 new rovibronic term values of the A 1Σ+ and b3ΠΩ states were obtained with an uncertainty about 0.01 cm−1 and added to the previously obtained 3439 term values in Kruzins et al. [Phys. The equilibrium structure of trans-glyoxal from experimental rotational constants and calculated vibration–rotation interaction constants

A total of six high-resolution FT-IR spectra for trans-glyoxal-d2, trans-glyoxal-d1 and trans-glyoxal-13C2 were recorded with a resolution ranging from 0.003 to 0.004 cm−1. By means of a simultaneous ground state combination difference analysis for each of these isotopologues using the Watson Hamiltonian in A-reduction and Ir-representation the ground state rotational constants are obtained. An empirical equilibrium structure is determined for trans-glyoxal using these experimental ground state rotational constants and vibration–rotation interaction constants calculated at the CCSD(T)/cc-pVTZ level of theory. Deuterium isotope effect on the induction period of the cerium catalyzed Belousov-Zhabotinsky reaction

Abstract In this work we present results about the deuterium isotopic effect on the global kinetics of a cerium catalyzed Belousov–Zhabotinsky reaction. A nonlinear dependence of the induction period upon the percentage of deuterated reactants was found in batch conditions. In order to understand this result, we investigated two reaction pathways responsible for the length of the induction period, namely: (a) the reaction between the enolic form of the malonic acid with molecular bromine and (b) the oxidation of malonic acid by the Ce(IV) ion. The conductance of lithium-7 fluoride in dioxane-water mixtures at 25�C

Conductance data for lithium-7 fluoride in dioxane-water mixtures covering the range 78.35>D>36 in dielectric constant are presented. These data and other previous data on lithium-7 chloride and lithium-7 iodide were analyzed by the Fuoss 1980 conductance equation in order to find the limiting conductance Δo, the pairing distance R and the conductometric association constant Kλ. Setting Ka=Kλ/VM (where VM is the molar volume of the solvent), the thermodynamic pairing constant and the corresponding change of the free energy Δg were calculated. Correlation among the values found for R and Δg=Δh−TΔs and the properties characteristic of the ions and solvents are discussed.

Protonation of Cp2TaH(CO) (Cp  =  C5H5, 1a; C5H4But, 1b) by HBF4·Et2O at −78 °C in CH2Cl2 affords [Cp2TaH2(CO)]BF4 (2, 3) as mixtures of 2 isomers. The minor ones (2a, 2b) contain the known trans-dihydride [Cp2TaH2(CO)]+ cations whereas the major ones (3a, 3b) are [Cp2Ta(η2-H2)(CO)]BF4, the first group 5 dihydrogen complexes. The crystal structure of the analogous complex 3a·BArf4 recorded at 120 K confirms the presence of the coordinated dihydrogen ligand, which displays an H–H separation of 1.09(2) A in agreement with distances calculated from NMR data. Protonation of Cp2TaH2(SiMe2Ph) by (Et2O)2 ·HBArf4 does not lead to an analogous silane derivative but to the new dinuclear complex [(Cp2…

Extending Limits of Chlorine Kinetic Isotope Effects

Chlorine kinetic isotope effects exceeding semiclassical limits were observed in enzyme-catalyzed reactions, but their source has not been yet identified. Herein we show that unusually large chlorine kinetic isotope effects are associated with reactions in which chlorine is the central atom that is being passed between two heavy atoms. The origin of these large values is the ratio of imaginary frequencies for light-to-heavy species (the so-called temperature-independent factor).

Assessment of a single-acquisition imaging sequence for oxygen-sensitive (3)He-MRI.

MRI of the lungs using hyperpolarized helium-3 (3He) allows the determination of intrapulmonary oxygen partial pressures (pO2). The need to separate competing processes of signal loss has hitherto required two different imaging series during two different breathing maneuvers. In this work, a new imaging strategy to measure pO2 by a single series of consecutive scans is presented. The feasibility of the method is demonstrated in three healthy human volunteers. Maps and histograms of intrapulmonary pO2 are calculated. Changes in the oxygen concentration of the inhaled gas mixture are well reproduced in the histograms.
