Thioredoxin-related protein of 14 kDa may directly reduce protein cysteinylation motifs
2018
Disulfide stress has been associated with inflammation and characterized by an increase in cystine levels and protein cysteinylation. Furthermore, it was recently discovered that thioredoxin-related protein of 14 kDa (TRP14, encoded by TXNDC17) exhibits efficient cystine reductase activity. The aim of our research was to elucidate if TRP14 is also able to reduce cysteinylated proteins in mammalian cells. Thus, protein cysteinylation was assessed in control and TRP14 knockdown cells in vitro through their pre-treatment with 25 µg/ml cycloheximide for 30 min and incubation with 250 µM biotinylated cysteine for 1 h. Moreover, such TRP14 knockdown cell lysates were tested as cysteinylated subst…
Properties of new asymmetrically quaternized dicationic ammonium based room-temperature ionic liquids with ether functionality
2013
Eleven asymmetrically quaternized dicationic ammonium-based room-temperature ionic liquids (DRTILs) with bis(trifluoromethanesulfonyl)imide (TFSI) were synthesized and characterized, along with 11 analogous dibromide precursors. Two-step synthesis was used to diquaternize tetramethyl-1,3-propanediamine and 2-(dimethylamino)-ethyl ether amines with a variety of alkyl and ether functionalized side chain groups (R1 ≠ R2). Each salt contain 1 to 3 ether groups located either in a linkage or in a side chain moieties. Structural and thermoanalytical properties, water content, and viscosity have been characterized using, for example, NMR, mass spectrometry (MS), X-ray diffraction, and thermal anal…
Synthesis of peptides with α,β‐dehydroamino acids, V. coupling experiments with C‐terminal dehydrophenylalanine and dehydroalanine residues
1986
N-Protected model dipeptides of C-terminal (Z)-ΔPhe and ΔAla couple with glycine esters after activation with either diphenyl phosphorazidate (DPPA), N-cyclohexyl-N′-[2-(N-methylmorpholinio)ethyl]carbodiimide p-toluenesulfonate (WSC), N,N′-dicyclohexylcarbodiimide + 1-hydroxybenzotriazole (DCC + HOBt), or the mixed anhydride (MA) with isobutyl chlorocarbonate to give in good or mostly moderate yields the (Z)-ΔPhe-containing tripeptides 1 – 9 and ΔAla-containing tripeptides 11 – 19, respectively. In the MA and DPPA methods, further acylation products 20a – d are formed to a great extent, expecially during the ΔAla peptide syntheses. (E)-TFA-Gly-ΔPhe and Gly-OtBu, irrespective of the activati…
Synthesis of 1,3,5-Trisubstituted Hydantoins by Regiospecific Domino Condensation/Aza-Michael/O→N Acyl Migration of Carbodiimides with Activated α,β-…
2005
[reaction: see text] Carbodiimides and suitably activated alpha,beta-unsaturated carboxylic acids react effectively to afford a vast array of 1,3,5-trisubstituted hydantoins by means of a regiospecific domino condensation/aza-Michael/N--O acyl migration. The reaction works well in very mild conditions (20 degrees C, dichloromethane) with fumaric acid derivatives bearing an electron-withdrawing group in the beta position. Good results have been obtained also with less activated substrates bearing only one electron-withdrawing group in the beta position, using more polar solvents (acetonitrile, DMF), and in the presence of a base (2,4,6-trimethylpyridine). Reactions with asymmetric carbodiimi…
The use of metallocenic esters of n-hydroxysuccinimide for metallohapten synthesis
1991
Different organometallic markers have been described in a new technique for the labelling of many drugs. Thus metallocenic esters of [M = (;CO)3CrC6H5; (;CO)3CrC6H5(;CH2)3; η-C5H5FeC5H4; (;CO)3MnηC5H4; (;CO)3MnηC5H4COCH2CH2; ηC5H4(;ηC5H5)Co+PF−6] react with primary or secondary amine drugs [DRUGNHR] for a psychostimulant drug: amphetamine; tricyclic antidepressants—desipramine and nortriptyline; a vasodilator—histamine; an adrenergic substance—norfenefrine; and for a central stimulant—meth-amphetamine, to give the metallohaptens MCON(;R)—DRUG. All these compounds have been fully characterized by different analytical methods and have potentialities for biological assays. This synthetic route…
Synthetic Antitumor Vaccines Through Coupling of Mucin Glycopeptide Antigens to Proteins
2017
The requirements for coupling reactions of carbohydrate molecules very much depend upon the biological recognition processes that should be investigated and upon the target structures of the desired carbohydrate ligand. If the carbohydrate conjugate itself is the recognized ligand, as for example, the binding site of a P-selectin ligand comprising sialyl-LewisX and a specific peptide sequence, the natural glycoside bond must be installed. A stereoselective and regioselective block glycosylation between a sialyl-LewisX trichloroacetimidate and a partially deprotected Thomsen–Friedenreich antigen derivative was developed to achieve this aim. In contrast, the coupling reactions by which glycop…
Fluorescent 3-amino-1,8-naphthalimide Tröger’s bases (3-amino-TBNaps) incorporating protected α-amino acids
2021
Abstract The synthesis of 3-amino-1,8-naphthalimide Troger’s base’s (TBNaps) 2–4, possessing several amino acids at the imide terminus, from the corresponding 3-amino-1,8-naphthalimide (Naps) units is described. The solid-state X-ray crystal structure of the related 3-amino-TBNaps piperidino-ethyl derivative 1, and its supramolecular packing is also outlined. In these molecules (as demonstrated from the X-ray structure of 1), the two Naps units have an orthogonal arrangement that makes them potentially attractive β-turn mimics for incorporation into peptide and polypeptide structures. The ground and the excited state properties of these systems were evaluated in a range of solvents that var…
Der Phenylacetyl-(PhAc-)Rest als enzymatisch ablösbare Schutzgruppe für Peptide und Kohlenhydrate: Selektive Schutzgruppenabspaltungen mit Penicillin…
1988

Aggregation processes of perylene bisimide diimidazolium salts
2015
The supramolecular aggregation of three diimidazolium- functionalized perylene bisimides, differing in the alkyl chain length was investigated. These salts form aggregates in solvents like chloroform, dichloromethane, and glycerol. Solvent-, concentration-, and temperature-dependent spectroscopic studies were carried out, evidencing the occurrence of an isodesmic, enthalpy-driven aggregation process, underpinned by p–p stacking and hydrogen bonding. Moreover, dynamic light scattering (DLS) measurements and SEM images revealed that these salts aggregate in chloroform into elongated structures.
