FISH and CHIPs: Colorful Clues to Radiation-Induced Chromosomal Instability

Radiation produces a variety of clonal and non-clonal chromosome aberrations that can be characterized by fluorescence in situ hybridization (FISH). Epigenetic changes affecting the expression of an essential DNA repair gene(s) may be an importantant mechanism for radiation-induced chromosomal instability. Expression profiling with specialized cDNA chips promises to identify candidate genes for the delayed effects of radiation and to provide new insights into the manifold and complex cellular responses to DNA damage. Much progress can be made by using FISH and CHIPs to study the mechanisms and biological consequences of ionizing radiation.
Assessing Chronological Aging in Saccharomyces cerevisiae

Saccharomyces cerevisiae is one of the most studied model organisms for the identification of genes and mechanisms that affect aging. The chronological lifespan (CLS) assay, which monitors the survival of a non-dividing population, is one of the two methods to study aging in yeast. To eliminate potential artifacts and identify genes and signaling pathways that may also affect aging in higher eukaryotes, it is important to determine CLS by multiple methods. Here, we describe these methods as well as the assays to study macromolecular damage during aging in yeast, with a focus on genomic instability.

Most human tumors are characterized by aneuploidy that is believed to be the consequence of chromosomal instability (CIN). The mechanism(s) leading to aneuploidy and the pathways that allow its tolerance are not completely understood. The Spindle Assembly Checkpoint (SAC) is a cellular surveillance mechanism working during mitosis, and alterations of genes that encode components of the SAC weakening the mitotic checkpoint, induce aneuploidy by chromosome mis-segregation. Theoretical study on travelling web dynamics and instability under non-homogeneous tension

Problems of dynamics and stability of a moving web, travelling between two rollers at a constant velocity, are studied using analytical approaches. Transverse vibrations of the web are described by a partial differential equation that includes the centrifugal force, in-plane tension, elastic reaction and nonstationary inertial terms. The model of a thin elastic plate subjected to bending and non-homogeneous tension is used to describe the bending moment and the distribution of membrane forces. Static instability analysis for travelling membranes and plates interacting with axially moving ideal fluid

The out-of-plane instability of a moving plate, travelling between two rollers with constant velocity, is studied, taking into account the mutual interaction between the buckled plate and the surrounding, axially flowing ideal fluid. Transverse displacement of the buckled plate (assumed cylindrical) is described by an integro-differential equation that includes the centrifugal force, the aerodynamic reaction of the external medium, the vertical projection of membrane tension, and the bending force. Abstract Instability of the Au/Ni(111) surface alloy is studied in different CO gas pressure, p , and temperature limits using kinetic Monte Carlo simulations. We analyze the reaction front dynamics and formation of Au clusters using the model which takes into account surface adatom pair and three-body interactions, CO adsorption and desorption, catalytic carbonyl formation reaction, Au and Ni adatom diffusion and their concerted exchange. Pyrogallarene shows concentration-dependent instability in dilute solutions resulting in elimination of two ketene molecules and formation of pyrogallarene lactones. This unexpected phenomenon, which is not observed with resorcinarenes, highlights the significance of the four hydroxyl groups at 2-position for the molecular characteristics of pyrogallarenes.
Stability of melt flow due to a traveling magnetic field in a closed ampoule

The linear three-dimensional instability of the flow due to a low frequency traveling magnetic field in a regular cylinder is studied numerically for height-to-diameter ratios in the range [0.5:2.5]. The first instability has the form of an azimuthal wave with wave numbers between 1 and 6 depending on the aspect ratio. It is shown that the flow is stable if the Reynolds number is below 290. Even stronger stationary flows may be obtained in a flattened cylinder.

Ion exchange resins (IERs) are widely used by the nuclear industry to decontaminate radioactive effluents. After use they are usually encapsulated in cementitious materials.
