We investigate the behavior of polymer colloids at the interface between two immiscible liquids using molecular dynamics simulations. We study several colloid morphologies with various degrees of amphiphilicity, that is, purely solvophobic homogeneous and Janus particles and amphiphilic Janus and core-shell particles. Regardless of the specific morphology, the polymer colloids irreversibly anchor at the liquid-liquid interface, accompanied by a marked reduction of the interfacial tension, γ. Purely solvophobic particles lower γ because they reduce the interfacial area shared by the two immiscible liquids, whereas amphiphilic colloids have an additional enthalpic contribution. Several hundred grams of Janus nanoparticles (d ≈ 40 nm) were synthesized from triblock terpolymers as compatibilizers for blending of technologically relevant polymers, PPE and SAN, on industry-scale extruders. The Janus nanoparticles (JPs) demonstrate superior compatibilization capabilities compared to the corresponding triblock terpolymer, attributed to the combined intrinsic properties, amphiphilicity and the Pickering effect. Straightforward mixing and extrusion protocols yield multiscale blend morphologies with "raspberry-like" structures of JPs-covered PPE phases in a SAN matrix. Type I Interferonopathies comprise inherited inflammatory diseases associated with perturbation of the type I IFN response. Use of Janus kinase (JAK) inhibitors has been recently reported as possible tools for treating some of those rare diseases. We describe herein the clinical picture and treatment response to the JAK-inhibitor ruxolitinib in a 5-year-old girl affected by Aicardi-Goutières Syndrome type 6 (AGS6) due to ADAR1 mutation. The girl's interferon score (IS) was compared with that of her older brother, suffering from the same disorder, who was not treated. The use of Janus kinase (JAK) inhibitors is a new approach in the therapy of inflammatory diseases with immune base. Tofacitinib is one of these inhibitors targeting JAK1 and JAK3, and its efficacy has been demonstrated in the treatment of moderate to severe ulcerative colitis (UC). It is a small synthetic molecule administered orally, with a fast bioavailability and elimination rate, predictable pharmacokinetics and lack of immunogenicity, which are convenient characteristics for both efficacy and safety. This article reviews the pharmacological characteristics of tofacitinib and its safety profile.

Hard and soft: Binding of inorganic Pt@Fe3O4 Janus particles to WS2 nanotubes through their Pt or Fe3O4 domains is governed by the difference in Pearson hardness: the soft Pt block has a higher sulfur affinity than the harder magnetite face; thus the binding proceeds preferentially through the Pt face. This binding preference can be reversed by masking the Pt face with an organic protecting group.

The diagnosis of essential thrombocythemia and polycythemia vera is often made during a thrombotic event which can be serious. Philadelphia-negative chronic myeloproliferative neoplasia patients have an increased thrombotic risk. This is assessed using various scoring systems but these are far from ideal and individual risk. Melanoma treatment has been revolutionized by antibody-based immunotherapies. IFNγ secretion by CD8+ T cells is critical for therapy efficacy having anti-proliferative and pro-apoptotic effects on tumour cells. Our study demonstrates a genetic evolution of IFNγ resistance in different melanoma patient models. Chromosomal alterations and subsequent inactivating mutations in genes of the IFNγ signalling cascade, most often JAK1 or JAK2, protect melanoma cells from anti-tumour IFNγ activity.
