Psoriātiska artrīta pacientu terapijas izvērtējums P.Stradiņa KUS Reimatoloģijas centrā
2016
Tēmas aktualitāte: pēdējos gados zinātniskās literatūras dati norāda par pētījumiem par NLR, PLR, MPV, CRO lietošanu psoriātiskā artrīta (PsA) pacientu slimības aktivitātes un terapijas efektivitātes novērtēšanā (Kim D.S. et all 2015), (Fu H. et all 2015), (Kisacik B. et all 2008), (Yazici S. et all 2010).
Glutamatergic projection from the nucleus incertus to the septohippocampal system
2012
Abstract Recent findings support a relevant role of the nucleus incertus in the control of the hippocampal activity through the modulation of theta rhythm. Previous studies from our group have shown that this nucleus is a critical relay between reticularis pontis oralis and the medial septum/diagonal band, regarded as the main activator and the pacemaker of the hippocampal oscillations, respectively. Besides, the nucleus incertus is highly linked to activated states related to the arousal response. The neurotransmission of the nucleus incertus, however, remains uncertain. Only GABA and the neuromodulator relaxin 3 are usually considered to be involved in its contribution to the septohippoca…
Abstract A004: Systemic RNA vaccines: Connecting effective cancer immunotherapy with antiviral defense mechanisms
2016
Abstract Mechanisms of antiviral host defense are important for survival and evolutionarily optimized for high sensitivity and potency. Intending to harvest the multitude of highly specialized and intertwined pathogen immune defense programs for cancer immunotherapy, we simulated a systemic pathogen intrusion into the blood stream by intravenous injection of lipid-formulated, tumor antigen-encoding mRNA nanoparticles. These RNA-lipoplexes (RNA-LPX) were directed to various lymphoid tissues, including the spleen, lymph nodes and bone marrow, which provide the ideal microenvironment for efficient priming and amplification of T cell responses. Solely the RNA-to-lipid ratio was discovered to de…
Toll-like receptors: Expression and involvement in Multiple Myeloma
2010
Multiple Myeloma (MM) cells express and respond to a broad range of TLRs. Accumulating evidences suggest that TLRs act as double-edged sword in MM biology. Indeed, TLR9 or TLR3 ligands could enhance immunity against MM cells or directly induce cell apoptosis, whereas various TLR agonists could induce MM survival, proliferation, and immune escape. This review is focused on the heterogeneous expression and function of TLRs in MM and on the potential implication of TLR ligands of infectious or endogenous origin in MM emergence, resistance, or progression.
Analysis of differentially expressed proteins in oral squamous cell carcinoma by MALDI-TOF MS
2010
J Oral Pathol Med (2010) 40: 369–379 Purpose: To explore the presence of differentially expressed proteins in OSCC for discrimination of tumour and normal mucosa to establish potential biomarkers and therapeutic targets. Experimental Design: Paired protein samples of 12 individuals (tongue cancer and non-cancerous mucosa) were separated by two-dimensional polyacrylamid gel electrophoresis. The protein patterns were compared pairwise and protein spots were quantified. We identified about 70 regulated proteins which we subsequently identified by MALDI-TOF mass spectrometry. Results: Cancerous and non-cancerous tissues could be most precisely distinguished by a panel of proteins. They inclu…
Identification of non-canonical NLRP3 functions in Th17 cells - Therapeutic interest to promote antitumor immune response
2021
Since decades, cancers have become a major public health issue, occupying the first premature mortality cause in France. One of the hallmarks of cancer cells is their ability to escape the immune system surveillance, notably through setting up an immunosuppressive environment within the tumor. The NLRP3 protein is largely studied in the inflammasome context, which is assembled in order to promote inflammation through IL-1β secretion. However, this protein also functions as a transcription factor in CD4+ T cells. Indeed, in Th2 cells, NLRP3 cooperate together with IRF4, to form a complex that is able to activate the transcription of Il4, Il5 and Il13, which are necessary for the function of …
THE ROLE OF INFLAMMATION IN TYPE A AORTIC DISSECTION: A PILOT STUDY
2013
Type A aortic dissection (TAAD) is a severe cardiovascular disease with high mortality rates. Current evidence suggests inflammation as the main mechanism of its complex pathophysiology. Accordingly, in this study the eventual presence of inflammatory cells in aorta specimens and any contribution of these cells in both apoptosis and metalloproteinase levels were assessed. The potential relationship between plasma inflammatory molecules and TAAD was also detected. In addition, implication in TAAD susceptibility of ten common and functional single nucleotide polymorphisms (SNP)s of six candidate genes (CCR5, TLR4, ACE, eNOs, MMP-9 and −2) was determined. Thus, histo-pathological and immunois…
Pharmacogenomics: a tool to prevent and cure coronary heart disease.
2007
Inflammation and genetics play an important role in the pathogenesis of coronary heart disease (CHD). This is supported by epidemiological studies which have thoroughly investigated the association between CHD and gene polymorphisms of the inflammatory molecules. Moreover, efforts to find elective therapy have not been rewarding and, despite the increasing appreciation of the role of genetics in CHD and myocardial infarction (MI) pathogenesis, pharmacogenomic approaches to uncover drug target have not been extensively explored. A critical search of published literature has suggested few inflammatory genes directly involved in the risk to develop CHD and MI. The selected genes are, the pro- …
Physical Activity Is Related with Cartilage Quality in Women with Knee Osteoarthritis
2017
Purpose To study the relationship between 12-month leisure-time physical activity (LTPA) level and changes in estimated biochemical composition of tibiofemoral cartilage in postmenopausal women with mild knee osteoarthritis (OA). Methods Originally, 87 volunteer postmenopausal women, age 60 to 68 yr, with mild knee OA (Kellgren Lawrence I/II and knee pain) participated in a randomized controlled, 4-month aquatic training trial (RCT), after which 76 completed the 12-month postintervention follow-up period. Self-reported LTPA was collected along the 12-month period using a diary from which MET task hours per month were calculated. Participants were divided into MET task hour tertiles: 1, lowe…
