A Polar18F-Labeled Amino Acid Derivative for Click Labeling of Biomolecules

This work describes the synthesis and 18F-labeling of an amino acid based prosthetic group that is able to participate in copper(I)-catalyzed cycloadditions. The prosthetic group can be used for 18F labeling of biomolecules under mild conditions. The synthesis started with L-serine methyl ester, which was derivatized by introducing an alkyne moiety and a leaving group for 18F labeling. Subsequently, 18F labeling as well as deprotection conditions were screened, which resulted in an overall radiochemical yield (RCY) of around 28 %. Furthermore, the 18F-labeled prosthetic group was treated with an azido cyclic Arg-Gly-Asp (cRGD) peptide as a model system in very high RCY of 98 %.

AbstractTo localize the nucleotide binding sites of the F1ATPase (TF1) from the thermophilic bacterium PS3 we have used 14C-labeled 8-azido ATP (8-N3ATP) as photoaffmity label. 8-N3ATP is hydrolyzed by the F,ATPase in the absence of ultraviolet light. Irradiation by ultraviolet light of the enzyme in the presence of 8-N3ATP results in reduction of ATPase activity and in preferential nucleotide specific labeling of the α subunits (0.8–0.9 mol 8-N3ATP/TF1,α:β = 4:1). Inactivation and labeling do not depend on the presence of Mg2+. Both effects decrease upon addition of various nucleotide di- or triphosphates.

Abstract The positron-emitting radionuclide 44 Sc with a half-life of 3.97 h and a β + branching of 94.3% is of potential interest for clinical PET. As so far it is available from a 44 Ti/ 44 Sc generator in Mainz, where long-lived 44 Ti decays to no-carrier-added (nca) 44 Sc. The 44 Sc is a trivalent metal cation and should be suitable for complexation with many well established bifunctional chelators conjugated to peptides or other molecular targeting vectors. In the present paper one population of the “large” subtidal mollusc Cerithium vulgatum Bruguiere, 1792 (Gastropoda: Cerithiidae) from the Northwestern coast of Sicily was investigated from a karyological point of view. The chromosome complement was Giemsa stained, conventionally karyotyped in 18 homomorphic chromosome pairs (10 bi-armed and 8 mono-armed), and subsequently analysed using silver, CMA3 and DAPI staining, and fluorescent in situ hybridization (FISH) with three repetitive DNA probes [ribosomal DNA (rDNA), (TTAGGG)n and (GATA)n]. Abstract The steroid hormone dehydroepiandrosterone (DHEA) has beneficial effects on vascular function, survival of neurons, and fatty acid metabolism. However, a specific receptor for DHEA has not been identified to date. Here, we describe the synthesis of a photoreactive DHEA derivative (Photo-DHEA). In Photo-DHEA, typical characteristics of DHEA are conserved: (i) a “planar” tetracyclic ring system with a Δ 5 double bond, (ii) a 3β-hydroxyl group, and (iii) a keto group at C17. In cell-based assays, Photo-DHEA showed the same properties as DHEA. We conclude that Photo-DHEA is suitable for radioiodination to yield a tool for the identification of the elusive DHEA receptor.

The radio antipodal number is the minimum span of a radio (diam(G) − 1)-labeling of G and the radio number is the minimum span of a radio (diam(G))-labeling of G. In this paper, the radio antipodal number and the radio number of the hypercube are determined by using a generalization of binary Gray codes.

Efferent projections of the medial cortex of the lizards Podarcis hispanica and Gallotia stehlinii were studied by examining the transport of horseradish peroxidase; results were correlated with those from Timm-stained sections. Two efferent systems were found. The first reaches the distal part of the outer plexiform layer in the medial, dorsomedial, and dorsal cortices, i.e., zones that are negative to Timm staining, and possibly originates from horizontal fusiform neurons.
