Abstract This article reviews the literature published in the last decade dealing with the transformation of α-santonin into bioactive or potentially bioactive sesquiterpenes. A number of syntheses starting from 8α-hydroxysantonin (artemisin) have also been included. Special emphasis has been placed on synthesized products that show biological activity. Major advances in this field include the application of new reagents and methodologies for the structural modification of the santonin skeleton and functionality, and its transformation into other sesquiterpenes, especially sesquiterpene lactones. This article reports the transformation of O-acetylisophotosantonin, obtained by photochemical rearrangement of santonin, into plagiochiline N, an ent-2,3-secoaromadendrane isolated from Plagiochila ovalifolia. The synthesis was carried out in a sequence involving as the key steps (a) the substitution of the lactone moiety by a gem-dimethylcyclopropane ring through a synthetic intermediate having a C(6)-C(7) double bond and (b) the ozonolysis of the C(2)-C(3) bond followed by cyclization to the dihydropyran ring characteristic of plagiochiline N. Spectroscopic data of the synthetic product fully coincided with the reported data for the natural product.

Lactols are compounds of increasing interest in the synthesis of active pharmaceutical derivatives. Nevertheless, the product obtained by the reduction of the carbonyl group of coumarin has been described only twice, and without definition of its precise chemical structure. Since these studies, doubts have been raised about the existence of a monomeric or dimeric form. Our study has led us to conclude definitely that the single dimeric form exists and to precisely define the spectral properties of the two diastereoisomers.

The structure of the title compound, C7H8O3, was determined in the course of our studies of the synthesis of cyclopenta[1,2-b]furan-4-one derivatives. The molecule has four chiral C atoms. The X-ray crystal structure analysis shows the compound to possess an epoxide group with an endo orientation with respect to the lactone group.

The molecular structure of the title compound [enantiomers (VIII) and (VIIIa)], C8H10O4, was determined in the course of our studies on the synthesis of cyclopenta[1,2-b]furan-4-one derivatives. Tricyclic (VIIIa) consists of a planar bridged lactone unit and the two other ring systems in the envelope conformation. It contains five chiral C atoms and was obtained as a racemic mixture. The X-ray analysis showed the compound to possess a half-acetal unit with an endo orientation of the half-acetal ether bridge with respect to the lactone unit.
An Insight into Substrate-Dependent Fluorination of some Highly Substituted Alicyclic Scaffolds

The substrate-dependent fluorination of some highly-functionalized cyclopentane derivatives with multiple chiral centers has been investigated. The key steps of the stereocontrolled syntheses are the oxidative cleavage of the ring carbon–carbon double bond of readily available diexo or diendo norbornene β-amino acid derivatives followed by transformation of the resulted dialdehyde stereoisomers by reduction. Finally, substrate-directable chemodifferentiation of different types of hydroxy groups under fluorination procedures gave various densely functionalized alicyclic derivatives or heterocycles.

Abstract The diastereoselective synthesis of hydroxy substituted γ- and δ-lactones was accomplished following two approaches. A 5- or 6-endo cyclization promoted by electrophilic selenium reagents of α- or β-hydroxy acids and a 5- or 6-exo cyclization of hydroxy esters obtained through a diastereoselective hydroxy selenenylation reaction of α- or β-hydroxy esters. Moreover, the diastereoselective methoxy selenenylation of the above compounds was investigated showing a case in which the compound that was unreactive in the hydroxy selenenylation conditions gave, in the methoxy selenenylation conditions, the deprotected diol. The thermal degradation of poly(oxycarbonylpentamethylene), (poly-e-caprolactone), (3), was investigated at 220°C and 80 mm Hg under nitrogen. 3 was found to be thermally much more stable than poly(oxycarbonylethylene), (poly-β-propiolactone), (1), although it decomposed faster than poly(oxycarbonyl-1,1-dimethylethylene), (polypivalolactone), (2). The reaction was of the first order with a rate constant of k=6,2.10−4 min−1 and proceeded via a “zipper mechanism” to yield monomeric e-caprolactone. A Carbohydrate-Based Julia–Kocienski Reagent for Syntheses of Chain-Extended and C-Linked Saccharides

The carbohydrate-derived Julia–Kocienski reagent 2-{[(3aS,4S,6R,6aR)-6-methoxy-2,2-dimethyltetrahydrofuro[3,4-d][1,3]dioxol-4-yl]methylsulfonyl}-1,3-benzothiazole (6) was prepared from d -ribose and investigated in the eponymous olefination. The base-promoted generation of the Julia anion induced a rearrangement to the corresponding l -lyxose epimer 2-{[(3aS,4R,6R,6aR)-6-methoxy-2,2-dimethyltetrahydrofuro[3,4-d][1,3]dioxol-4-yl]methylsulfonyl}-1,3-benzothiazole (12), which reacted readily with aldehydes and with a gluconolactone. The latter reaction furnished an exo-glycal-linked C-diglycoside.
