Secondary Structure-Driven Hydrogelation Using Foldable Telechelic Polymer-Peptide Conjugates.

The synthesis of ABA and ABA' triblock polyethylene glycol-and polysarcosine-peptide conjugates is reported. The A/A' peptides are based on phenylalanine(F)-histidine(H) pentapeptide sequences FHFHF, which promote pH-switchable β-sheet self-assembly into nanorods in water. Only parallel β-sheet-driven folding and intermolecular assembly using ABA triblock polymer-peptide conjugates leads to interstrand cross-linking and hydrogelation, highlighting the impact of supramolecular interactions-directed structure formation at the nano- and mesoscopic level.

Extension of hydroxytelechelic polybutadiene oligomers ( 3200 g/mol) is performed in mild conditions, in order to avoid backbone modifications, using succinic anhydride or methylenedicyclohexyl diisocyanate. The two routes present significative extension of the initial oligomers. Extension through the anhydride route, in the presence of DCC and DMAP, leads to new hydroxytelechelic oligomers ( 8000 g/mol). A modified benzyloxycarbonyl (BOC) protective group, the 3,5-di-tert-butyl substituted BOC (3,5-tBBOC) group, was introduced to the stepwise synthesis of molecularly uniform oligourethanes based on 1,5-naphthalene diisocyanate (NDI) and 1,4-butanediol (BDO) for the protection of the amino functions of the starting material 1,5-naphthalene-diamine (NDA). Using 2-hydroxyethyl α-bromoisobuty-rate as initiator, atom transfer radical polymerization (ATRP) of tert-butyl acrylate leads to poly(tert-butyl acrylate) (PtBA) with a hydroxyl group at one and a bromine atom at the other end. Esterification of the hydroxyl group of these heterotelechelic polymers with acryloyl chloride yields PtBA (M n = 3060) with a polymerizable double bond at one end and a bromine atom at the other end which can act as an initiator in ATRP (macroinimer).
