Infliximab to treat Crohn's disease: an update
2011
Abstract: Crohn’s disease (CD) is a chronic inflammatory disease of the gastrointestinal tract characterized by recurring flares followed by periods of inactive disease and remission. The etiology is unknown, although the common opinion is that the disease arises from a disordered immune response to the gut contents in genetically predisposed individuals. Infliximab (IFX), a chimeric immunoglobulin G1 monoclonal antibody to tumor necrosis factor, has dramatically changed the approach to managing patients with CD and improving their treatment, by achieving treatment goals, such as mucosal healing, and decreasing the need for hospitalizations and surgeries. This review provides an update on e…
Adalimumab vs Azathioprine in the Prevention of Postoperative Crohn's Disease Recurrence. A GETECCU Randomised Trial.
2017
Background and aims Postoperative recurrence of Crohn's disease [POR-CD] is almost certain if no prophylaxis is administered. Evidence for optimal treatment is lacking. Our aim was to compare the efficacy of adalimumab [ADA] and azathioprine [AZA] in this setting. Methods We performed a phase 3, 52-week, multicentre, randomised, superiority study [APPRECIA], in which patients with ileocolonic resection were randomised either to ADA 160-80-40 mg subcutaneously [SC] or AZA 2.5 mg/kg/day, both associated with metronidazole. The primary endpoint was endoscopic recurrence at 1 year [Rutgeerts i2b, i3, i4], as evaluated by a blinded central reader. Results We recruited 91 patients [median age 35.…
Anti-IgE antibodies for the treatment of asthma
2005
Purpose of review Allergic asthma is a hypersensitivity reaction initiated by immunologic mechanisms mediated by IgE antibodies. IgE plays a central role in the initiation and propagation of the inflammatory cascade and thus the allergic response. Targeting factors involved in the allergic response, such as IgE, is a novel strategy for new therapies. Attenuating allergic disease by specifically inhibiting IgE and the development of the monoclonal anti-IgE antibody, omalizumab, were major breakthroughs in asthma management. Recent findings Several studies have shown that omalizumab has significant anti-inflammatory effects and that it may act on multiple components of the inflammatory cascad…
Test system for trifunctional antibodies in 3D MCTS culture.
2009
The aim of the present study was to assess the feasibility of a 3D tumor cell culture model, that is, multicellular tumor spheroids (MCTSs) as an adequate model for micrometastases and therefore as a pharmacological model for efficacy testing of trifunctional therapeutic antibodies. Unlike conventional monolayer cultures, spheroids allow researchers to study parameters, such as 3D cell shape, 3D cell arrangement and microenvironment, and penetration efficiency of defense cells that may largely influence the efficacy of antibody treatment in vivo. The authors established a long-term coculture of human MCTSs with peripheral blood mononuclear cells (PBMCs) to test the anticancer effect of the …
Can the response to Omalizumab be influenced by treatment duration? A real-life study
2017
Objective It is unknown whether Omalizumab effectiveness changes over the course of time. Our retrospective real-life study tried to analyze whether Omalizumab response may be influenced by treatment duration. Methods 340 severe asthmatics treated with Omalizumab for different periods of time were recruited. They were subdivided into 4 groups according to the Omalizumab treatment length: 60 months. Omalizumab treatment results (FEV1, exacerbations, ACT, SABA use, asthma control levels, medications used e and ICS doses) were compared. Results ACT, exacerbations, GINA control levels, ICS doses and SABA use were similar in all groups with different Omalizumab treatment durations. Using a linea…

2018
This mini-review will summarize the present state of development of the CD38 antibody daratumumab for the treatment of multiple myeloma.
A Selective Change Driven System for High-Speed Motion Analysis.
2016
Vision-based sensing algorithms are computationally-demanding tasks due to the large amount of data acquired and processed. Visual sensors deliver much information, even if data are redundant, and do not give any additional information. A Selective Change Driven (SCD) sensing system is based on a sensor that delivers, ordered by the magnitude of its change, only those pixels that have changed most since the last read-out. This allows the information stream to be adjusted to the computation capabilities. Following this strategy, a new SCD processing architecture for high-speed motion analysis, based on processing pixels instead of full frames, has been developed and implemented into a Field …

Performance of the Fully Digital FPGA-Based Front-End Electronics for the GALILEO Array
2014
In this work we present the architecture and results of a fully digital Front End Electronics (FEE) read out system developed for the GALILEO array. The FEE system, developed in collaboration with the Advanced Gamma Tracking Array (AGATA) collaboration, is composed of three main blocks: preamplifiers, digitizers and preprocessing electronics. The slow control system contains a custom Linux driver, a dynamic library and a server implementing network services. The digital processing of the data from the GALILEO germanium detectors has demonstrated the capability to achieve an energy resolution of 1.53 per mil at an energy of 1.33 MeV.
Uniform rectifiability implies Varopoulos extensions
2020
We construct extensions of Varopolous type for functions $f \in \text{BMO}(E)$, for any uniformly rectifiable set $E$ of codimension one. More precisely, let $\Omega \subset \mathbb{R}^{n+1}$ be an open set satisfying the corkscrew condition, with an $n$-dimensional uniformly rectifiable boundary $\partial \Omega$, and let $\sigma := \mathcal{H}^n\lfloor_{\partial \Omega}$ denote the surface measure on $\partial \Omega$. We show that if $f \in \text{BMO}(\partial \Omega,d\sigma)$ with compact support on $\partial \Omega$, then there exists a smooth function $V$ in $\Omega$ such that $|\nabla V(Y)| \, dY$ is a Carleson measure with Carleson norm controlled by the BMO norm of $f$, and such th…
