Mechanisms involved in the transmission of epigenetic deregulation : analyses of transmission in human sperm

The notion that environmental exposure can be memorized and promote epimutation (defined as defects on DNA methylation) raises the question of possible epigenetic transgenerational transmission in humans. To address whether an epimutation could be transmitted in humans, we pursued two axes. First, the evaluation of intergenerational transmission in the family of a Silver-Russell patient has shown, for the first time, the efficiency of epigenetic reprogramming in humans, specifically on imprinted regions. Protonation and methylation reactions of 2-pyridyl-palladium(II) and -platinum(II) complexes

Abstract The reactions of strong acids HX and HClO4 with the 2-pyridyl complexes [PdX(μ-C5H4N-C2,N)(PPh3)]2 (X = Cl, Br), trans-[PdCl(C5H4N-C2)(PEt3)2] and [PdCl(C5H4N-C2)(dppe)] yield the N-protonated derivatives cis-[PdX2-(C5H5N-C2)(PPh3)], trans-[PdCl(C5H5N-C2)(PEt3)2]ClO4 and [PdCl-(C5H5N-C2)(dppe)]ClO4, respectively. The terminal 2-pyridyl group of trans-[PdCl(C5H4N-C2)(PEt3)2] and [PdCl(C5H4N-C2)(dppe)] also reacts with Me2SO4/NaClO4 to give trans-[PdClC5H4(l-Me)N-C2(PEt3)2]ClO4 and [PdClC5H4(l-Me)N-C2(dppe)]ClO4. Interleukin-12 (IL-12) is a proinflammatory cytokine produced by antigen-presenting cells in response to many microbial infections. IL-12 plays an important role in the generation of T helper type-1 cells, which favor cell-mediated immune response. IL-12 is composed of two different subunits, p40 and p35, whose expression can be regulated concomitantly or differentially. Monocytic cells, the major producers of IL-12, can be primed by interferon-gamma (IFN-gamma) to produce optimal amounts of IL-12 in response to LPS stimulation as a consequence of bacterial infection. Structural determination of the lipo-chitin oligosaccharide nodulation signals produced by Rhizobium giardinii bv. giardinii H152

Abstract Rhizobium giardinii bv. giardinii is a microsymbiont of plants of the genus Phaseolus and produces extracellular signal molecules that are able to induce deformation of root hairs and nodule organogenesis. We report here the structures of seven lipochitooligosaccharide (LCO) signal molecules secreted by R. giardinii bv. giardinii H152. Six of them are pentamers of GlcNAc carrying C 16:0 , C 18:0 , C 20:0 and C 18:1 fatty acyl chains on the non-reducing terminal residue. Fatty Liver and Fibrosis in Glycine N-Methyltransferase Knockout Mice Is Prevented by Nicotinamide

Deletion of glycine N-methyltransferase (GNMT), the main gene involved in liver S-adenosylmethionine (SAM) catabolism, leads to the hepatic accumulation of this molecule and the development of fatty liver and fibrosis in mice. To demonstrate that the excess of hepatic SAM is the main agent contributing to liver disease in GNMT knockout (KO) mice, we treated 1.5-month-old GNMT-KO mice for 6 weeks with nicotinamide (NAM), a substrate of the enzyme NAM N-methyltransferase. We previously reported the characterization 5S ribosomal DNA (rDNA) clusters in the common sea urchin Paracentrotus lividus and demonstrated the presence of DNA methylation-dependent silencing of embryo specific 5S rDNA cluster in adult tissue. In this work, we show genetic and epigenetic characterization of 18S-26S rDNA clusters in this specie. The results indicate the presence of three different 18S-26S rDNA clusters with different Non-Transcribed Spacer (NTS) regions that have different chromosomal localizations.
