Pentamidine rescues contractility and rhythmicity in a Drosophila model of myotonic dystrophy heart dysfunction

2015

Up to 80% of individuals with myotonic dystrophy type 1 (DM1) will develop cardiac abnormalities at some point during the progression of their disease, the most common of which is heart blockage of varying degrees. Such blockage is characterized by conduction defects and supraventricular and ventricular tachycardia, and carries a high risk of sudden cardiac death. Despite its importance, very few animal model studies have focused on the heart dysfunction in DM1. Here, we describe the characterization of the heart phenotype in a Drosophila model expressing pure expanded CUG repeats under the control of the cardiomyocyte-specific driver GMH5-Gal4. Morphologically, expression of 250 CUG repeat…

Effet de l'humidité sur la réponse à l'ammoniac de capteurs conductimétriques à base de matériaux moléculaires

2012

Many ammonia sensors are available but the consideration of the humidity effect is scarcely insured even by manufacturers. The aim of this thesis is to propose a conductimetric transducer capable tomeasure ammonia selectively, with enough accuracy for air quality control, in a variable humidity environment. To reach this goal, we used thin film resistors made of cobalt phthalocyanines as semiconductor. This study deals with thin films processing made by vacuum evaporation, solventcast and electrodeposition. Automated workbenches were developed to perform electrical measurements under argon and to study the response of sensors to dry or humid ammonia by meansof 1 min / 4 min-long exposure / …

A factor derived from chick embryo retina which inhibits DNA synthesis of retina itself.

1992

Chick embryo retinas contain a peptide factor that inhibits DNA synthesis in explants of chick embryo retina. The inhibitory factor, obtained by acid/ethanol extraction from 15-day-old chick embryo retinas, was partially purified by affinity chromatography on heparin-sepharose CL-6B and gel filtration on Sephadex G-100. The inhibitor reduced DNA synthesis with maximal effects observed in retinal explants from 7 to 8-day-old chick embryos. The inhibitory effect became apparent after 10 h of incubation and reached the maximum levels after 16 h. DNA-inhibiting activity was heat and acid-stable and was destroyed by trypsin and alkaline treatments. The inhibitory effect was observed in retinal e…

The subnivium, a haven for

2019

Parasite nematodes of the genus Trichinella are transmitted from one host to another through the ingestion of larvae present in striated muscles. The longer the survival of muscle larvae in host carcasses, the higher the probability of being ingested by a scavenging host. Thereby, these nematodes developed an anaerobic metabolism favouring their survival in decaying tissues. In addition, muscle larvae of three taxa, namely Trichinella nativa, Trichinella britovi and Trichinella T6, can survive freezing for several months to several years depending on the taxon. The aim of the present work was to investigate the survival time of T. britovi larvae in naturally infected host carcasses preserve…

Foldameerien isäntä-vieraskemia

2017

Tutkielman kirjallisessa osassa tarkastellaan foldameerejä isäntä-vieraskemian näkökulmasta. Foldameerit voivat sitoa rakenteeseensa erilaisia vierasmolekyylejä, jolloin muodostuu isäntä-vieraskomplekseja. Kirjallisuudesta valittiin tarkasteluun mielenkiintoisimpia anioneja, kationeja ja neutraaleja vierasmolekyylejä sitovia foldameereja ja perehdyttiin niiden vuorovaikutuksiin. Lisäksi tarkasteltiin vierasmolekyylin sitoutumisen aikaansaamia muutoksia foldameerien konformaatiossa. Kokeellisen osan tarkoituksena oli syntetisoida pitkä, 11 aromaattista rengasta sisältävä aryyliamidifoldameeri käyttäen hyväksi pääasiassa peptidikytkentäreaktioita. Koska synteesi osoittautui erittäin haasteell…

Jaunu Staphylococcus Epidermidis sortāzes A inhibitoru atlase izmantojot FRET skrīninga sistēmu

2020

Pētījuma mērķis ir atrast jaunus zāļu kandidātus, kuri spēj inhibēt Staphylococcus epidermidis sortāzi A, vienlaicīgi mazinot patogēnu multirezistēnces izplatīšanos. Šim mērķim tika izveidoti ģenētiski DNS konstrukti, kuri kodē S.epidermidis sortāzes A katalītisko domēnu, iebūvējot pET22b un pET28a vektoros srtA gēnu. Rekombinantā sortāzes proteīna sintēze tika inducēta ar IPTG, izmantojot BL21(DE3) celmu Escherichia coli ekspresijas sistēmā. Proteīns bez papildus heksahistidīna sekvences (pET22b vektors) tika attīrīts ar anjonapmaiņas hromatogrāfijas kolonnām, savukārt heksahistidīna sekvenci saturošā sortāze (pET28a vektors) tika attīrīta iz…

Cilvēka mikroflora kā potenciāls acs infekcijas avots

2015

Bakalaura darbs uzrakstīts latviešu valodā. Mērķis: novērtēt cilvēka normālās mikrofloras iespējamo lomu acu slimību etioloģijā un patoģenēzē. Uzdevumi: noteikt S.aureus nēsātāju izplatību sabiedrībā; novērtēt roku mazgāšanas un dezinfekcijas efektivitāti; noteikt no dezinficēto roku ādas izolēto KONS jutību pret antibiotikām.

PVC silver zeolite composites with antimicrobial properties

2011

Poly(vinyl chloride) (PVC) composites containing increasing amounts (2-20%, w/w) of silver zeolite (SZ) were prepared by melt mixing and characterized by thermal, mechanical and rheological analyses. The addition of large amount of SZ did not influence the processability and the formability of the composites, if compared to neat plasticized PVC. The antibacterial activity of PVC SZ composites was tested on Escherichia coli and Staphylococcus epidermidis and resulted promising both in culture broth and on agar plate and also in sterile urine seeded with these strains, for simulation purposes. In sterile urine, composites induced a significant reduction (4-6 log units) of viability of both st…
