Inhibition of T cell activityin vivo: a test model for quantitative evaluation

A test model is presented which, in comparison with the conventional models of skin transplantation or graft-versus-host (GvH) reaction in mice, permits a more sensitive quantitative evaluation of T cell inhibition in vivo. Prospective donors (type AA) are immunized with prospective recipient material (type AB); the resulting T cell reaction of A versus B is inhibited by consecutive treatment. Extent of inhibition can be evaluated after transfer of the pretreated AA material onto AB recipients by calculation of remaining GvH reactivity, if compared to adequate control tranfers. The reactions of labile [Re(diimine)(CO)3(H2O)]+ precursors (diimine = 2,2′-bipyridine, bpy; 1,10-phenanthroline, phen) with dicyanoargentate anion produce the dirhenium cyanide-bridged compounds [{Re(diimine)(CO)3}2CN)]+ (1 and 2). Substitution of the axial carbonyl ligands in 2 for triphenylphosphine gives the derivative [{Re(phen)(CO)2(PPh3)}2CN]+ (3), while the employment of a neutral metalloligand [Au(PPh3)(CN)] affords heterobimetallic complex [{Re(phen)(CO)3}NCAu(PPh3)]+ (4). Donor lymphocyte infusions (DLI) are used to resolve mixed T-cell chimerism (TCC) after allo-SCT despite a substantial risk of GVHD. We analyzed the impact of prophylactic CD8-depleted (CD8(depl)) DLI in 20 recipients of anti-CD52 alemtuzumab in vivo T-cell-depleted allografts with declining donor TCC after day +60. A total of 13 patients received CD8(depl) DLI and 7 patients did not. All but one of the DLI patients converted to complete donor T-cell chimeras, whereas only one non-DLI patient converted spontaneously. International audience; Borexino measured with unprecedented accuracy the fluxes of solar neutrinos emitted at all the steps of the pp fusion chain. Still missing is the measurement of the flux of neutrinos produced in the CNO cycle. A positive measurement of the CNO neutrino flux is of fundamental importance for understanding the evolution of stars and addressing the unresolved controversy on the solar abundances. The analog signals generated in the read-out electronics of particle detectors are shaped prior to the digitization in order to improve the signal to noise ratio (SNR). The real amplitude of the analog signal is then obtained using digital filters, which provides information about the energy deposited in the detector. The classical digital filters have a good performance in ideal situations with Gaussian electronic noise and no pulse shape distortion. However, high-energy particle colliders, such as the Large Hadron Collider (LHC) at CERN, can produce multiple simultaneous events, which produce signal pileup. The aim of this chapter is to summarise the problems incurred during the phases of calibrating and validating the extortion racket models used by the GLODERS project. The chapter starts with the discussion of the data availability and summarises shortly the contents of Sect. 4.3. It continues with a discussion of what parameterisation, calibration, sensitivity analysis and validation have to do with each other and ends up with a discussion of the validity of the GLODERS models.

Abstract Introduction Imaging of somatostatin receptor expressing tumours has been greatly enhanced by the use of 68 Ga-DOTATOC and PET/CT. Methods In this work, a purification method for the 68 Ge/ 68 Ga generator eluate and a method to produce 68 Ga-DOTATOC suitable for clinical use were evaluated. The generator eluate was purified and concentrated on a cation-exchange cartridge in HCl/acetone media. The efficacy of this procedure in eliminating metal impurities from the 68 Ga solution was investigated by ICP-MS. The radiotracer quality was evaluated by radio-TLC, GC and γ-ray spectrometry. Prevention of desiccation is a constant challenge for terrestrial organisms. Land insects have an extracellular coat, the cuticle, that plays a major role in protection against exaggerated water loss. Here, we report that the ABC transporter Oskyddad (Osy)—a human ABCA12 paralog—contributes to the waterproof barrier function of the cuticle in the fruit fly Drosophila melanogaster. We show that the reduction or elimination of Osy function provokes rapid desiccation. Osy is also involved in defining the inward barrier against xenobiotics penetration. Consistently, the amounts of cuticular hydrocarbons that are involved in cuticle impermeability decrease markedly when Osy activity is reduced. …
