Cryo-EM structure of ssDNA bacteriophage ΦCjT23 provides insight into early virus evolution.

2022

AbstractThe origin of viruses remains an open question. While lack of detectable sequence similarity hampers the analysis of distantly related viruses, structural biology investigations of conserved capsid protein structures facilitate the study of distant evolutionary relationships. Here we characterize the lipid-containing ssDNA temperate bacteriophage ΦCjT23, which infects Flavobacterium sp. (Bacteroidetes). We report ΦCjT23-like sequences in the genome of strains belonging to several Flavobacterium species. The virion structure determined by cryogenic electron microscopy reveals similarities to members of the viral kingdom Bamfordvirae that currently consists solely of dsDNA viruses wit…

A Model for High-Cycle Fatigue in Polycrystals

2018

A grain-scale formulation for high-cycle fatigue inter-granular degradation in polycrystalline aggregates is presented. The aggregate is represented through Voronoi tessellations and the mechanics of individual bulk grains is modelled using a boundary integral formulation. The inter-granular interfaces degrade under the action of cyclic tractions and they are represented using cohesive laws embodying a local irreversible damage parameter that evolves according to high-cycle continuum damage laws. The consistence between cyclic and static damage, which plays an important role in the redistribution of inter-granular tractions upon cyclic degradation, is assessed at each fatigue solution jump,…

Virtual Element Method: Micro-Mechanics Applications

2019

In this contribution we present an application of the lowest order Virtual Element Method (VEM) to the problem of material computational homogenization. Material homogenization allows retrieving material properties through suitable volume averaging procedures, starting from a detailed representation of the micro-constituents of the considered material. The representation of such microstructure constitutes a remarkable effort in terms of data/mesh preparation, especially when there is not evident microstructural regularity. For such a reason, computational micromechanics may represent a challenging benchmark for showing the potential of VEM. In this contribution, polycrystalline materials ar…

A Model for Low-Cycle Fatigue in Micro-Structured Materials

2019

A microscale formulation for low-cycle fatigue degradation in heterogeneous materials is presented. The interface traction-separation law is modelled by a cohesive zone model for low-cycle fatigue analysis, which is developed in a consistent thermodynamic framework of elastic-plastic-damage mechanics with internal variables. A specific fatigue activation condition allows to model the material degradation related to the elastic-plastic cyclic loading conditions, with tractions levels lower than the static failure condition. A moving endurance surface, in the classic framework of kinematic hardening, enables a pure elastic behaviour without any fatigue degradation for low levels of cyclic tra…

Structural characterization and electrochemical hydrogen storage properties of Ti2LxZrxNi (x [ 0, 0.1, 0.2) alloys prepared by mechanical alloying

2013

International audience; Nominal Ti2Ni was synthesized under argon atmosphere at room temperature using a planetary high-energy ball mill. The effect of milling time and Zr substitution for Ti on the microstructure was characterized by XRD, SEM and TEM, and the discharge capacities of Ti2xZrxNi (x 1/4 0, 0.1, 0.2) were examined by electrochemical measurements at galvanostatic conditions. XRD analysis shows that amorphous phase of Ti2Ni can be elaborated by 60 h of milling, whereas Zr substitution hinders amorphization process of the system. The products of ball milling nominal Ti2xZrxNi (x 1/4 0.1, 0.2) were austenitic (Ti, Zr)Ni and partly TiO, despite the fact that the operation was carrie…

Magnetic domain structure of La0.7Sr0.3MnO 3 thin-films probed at variable temperature with scanning electron microscopy with polarization analysis

2013

The domain configuration of 50 nm thick La0.7Sr0.3MnO3 films has been directly investigated using scanning electron microscopy with polarization analysis (SEMPA), with magnetic contrast obtained without the requirement for prior surface preparation. The large scale domain structure reflects a primarily four-fold anisotropy, with a small uniaxial component, consistent with magneto-optic Kerr effect measurements. We also determine the domain transition profile and find it to be in agreement with previous estimates of the domain wall width in this material. The temperature dependence of the image contrast is investigated and compared to superconducting-quantum interference device magnetometry …

Dielectric response of BaTiO3 electronic states under AC fields via microsecond time-resolved X-ray absorption spectroscopy

2021

Abstract For the first time, the dielectric response of a BaTiO 3 thin film under an AC electric field is investigated using microsecond time-resolved X-ray absorption spectroscopy at the Ti K-edge in order to clarify correlated contributions of each constituent atom on the electronic states. Intensities of the pre-edge e g peak and shoulder structure just below the main edge increase with an increase in the amplitude of the applied electric field, whereas that of the main peak decreases in an opposite manner. Based on the multiple scattering theory, the increase and decrease of the e g and main peaks are simulated for different Ti off-center displacements. Our results indicate that these s…

Conic optical fiber probe for generation and characterization of microbubbles in liquids

2021

Abstract A novel optical fiber probe has been developed to provide mechanical stability to microbubbles generated in fluids, the tip of the fiber is etched with hydrofluoric acid to pierce a truncated horn that fastens the microbubbles to the fiber tip and prevents misalignment or detachment caused by convection currents, vibrations or shocks in the liquid. Microbubbles are photo-thermally generated on the etched fiber and used as Fabry-Perot cavity sensor. Two methods were used to interrogate the probe: the first one, in the wavelength domain, is suitable for calibration in static or quasi static situations; the second one, in the time domain, can be used in dynamic environments. Experimen…

Low energy nano diffraction (LEND) – A versatile diffraction technique in SEM

2019

Abstract Electron diffraction is a powerful characterization method that is used across different fields and in different instruments. In particular, the power of transmission electron microscopy (TEM) largely relies on the capability to switch between imaging and diffraction mode enabling identification of crystalline phases and in-depth studies of crystal defects, to name only examples. In contrast, while diffraction techniques have found their way into the realm of scanning electron microscopy (SEM) in the form of electron backscatter diffraction and related techniques, on-axis transmission diffraction is still in its infancy. Here we present a simple but versatile setup that enables a ‘…
