Docking and synthesis of pyrrolopyrimidodiazepinone derivatives (PPDs) and their precursors: New scaffolds for DNA-interacting agents

New classes of pyrrolopyrimidodiazepinone derivatives (PPDs) and their precursors were studied in silico for their ability to form stable complex with DNA fragment. In the docking studies two binding modes can be envisaged: groove mode and intercalating mode. In the case of the best ligands the docking studies revealed a common binding mode with the chromophore intercalated between GC base pairs whereas the side chain lies close to the minor groove. Synthetic approach to the PPD ring systems is discussed.

Synthesis and photophysical properties of dansyl-based polyamine ligands and their Zn(II) complexes

The synthesis, potentiometric studies and photophysical properties of two new polyamine ligands (L1 and L2) possessing the dansyl chromophore were studied in aqueous 0.15 M NaCl. The compounds show the absorption and emissions bands characteristic of the dansylamide fluorophore and both present intramolecular excited state proton transfer at intermediate pH ranges. One of the ligands (L2) strongly coordinates Zn(II) leading to fluorescence quenching. Optically detected magnetic resonance of triplet states populated by photoexcitation in water-soluble chlorophyll proteins (WSCPs) from Lepidium virginicum has been performed using both absorption and fluorescence detection. Well resolved triplet-singlet (T-S) spectra have been obtained and interpreted in terms of electronic interactions among the four chlorophylls (Chls), forming two dimers in the WSCP tetramer. Localization of the triplet state on a single Chl leads to a redistribution of the oscillator strength in the remaining three Chls of the complex. Determination of diphenhydramine hydrochloride by flow injection with Bromophenol Blue and turbidimetric measurement

The study of a number of diphenhydramine-dye systems was carried out in order to determine the most suitable precipitate for the turbidimetric determination of diphenhydramine using flow injection (FI). The reagent selected was Bromophenol Blue. The chemical and FI variables were optimised. The calibration graph was linear over the concentration range 50-230 p.p.m. of diphenhydramine hydrochloride. A number of interfering substances were also investigated.

Probing the electronic state of a single coronene molecule by the emission from proximate fluorophores.

We measured electronic transitions of the 2D graphene-type molecule hexa-peri-hexabenzocoronene (HBC) at the single-molecule level. The large intersystem crossing rate and long triplet state lifetime in the range of seconds are prohibitive for direct single-molecule observation. By covalently coupling fluorescent acceptor molecules (perylenecarboximide, PMI) to HBC, efficient singlet energy transfer gives rise to strong PMI fluorescence. Confocal single-molecule fluorescence microscopy with two excitation colours matching the HBC and PMI transition frequencies, respectively, was conducted. New Austalides from the Sponge-Associated Fungus Aspergillus sp.

Chromatographic separation of a crude extract obtained from the fungus Aspergillus sp., isolated from the Mediterranean sponge Tethya aurantium, yielded five new meroterpenoid metabolites, austalides M–Q (1–5), together with nine known compounds (6–13). The structures of the new compounds were unambiguously elucidated on the basis of extensive 1D and 2D NMR methods and by mass spectral analysis. Express RGB mapping of three to five skin chromophores

Skin melanin, oxy- and deoxy-hemoglobin were snapshot-mapped under simultaneous 448-532-659 nm laser illumination by a smartphone RGB camera. Experimental prototypes for double-snapshot RGB mapping of four (melanin, bilirubin, oxy- and deoxy-hemoglobin) and five (melanin, bilirubin, lipids, oxy- and deoxy-hemoglobin) skin chromophores with reduced laser speckle artefacts have been developed and tested. A set of 405-448-532-659 nm lasers were used for four chromophores mapping, and a set of 405-448-532-659-842 nm lasers for five chromophores mapping. Clinical tests confirmed functionality of the developed devices.

Photocatalytic coalescence of functionalized gold nanoparticles.

A novel strategy for the synthesis of chromophore-functionalized AuNPs with a narrow size distribution is reported. It consists of increasing the size of preprepared NPs by means of a fast (second scale) and clean (light and an organic photocatalyst) method. The results agree with thiolate ligand liberation from the NP surface promoted by photogenerated radicals. This lets gold cores come together and finally coalesce.
