Alzheimer’s disease (AD) is one of over 18 different disorders known as tauopathies, characterized by the pathological aggregation and accumulation of tau, a microtubule-associated protein. Tau aggregates are heterogeneous and can be divided into two major groups: large metastable neurofibrillary tangles (NFTs) and oligomers. Recently, it has been shown that tau oligomers are highly toxic in vitro and efficient seeds for the propagation of pathology as compared to NFTs. [EN] The sample preparation procedure for MALDI-TOF MS of polymers is addressed in this study by the application of a statistical Design of Experiments (DoE). Industrial poly (ethylene terephthalate) (PET) was chosen as model polymer. Different experimental settings (levels) for matrixes, analyte/matrix proportions and concentrations of cationization agent were considered. Alzheimer’s Disease (AD) is a multifactorial neurodegenerative dementia with no curative therapeutic options. One of the earliest impairments in AD triggering cognitive decline is the synaptic dysfunction induced by the selective targeting and interruption of the synaptic region by the small oligomeric form of amyloid beta (Aβ). Recently, the co-presence of Aβ oligomers (Aβo) and tau oligomers (tau-o) at the synapses has been suggested to exacerbate this dysfunction. Therefore, the development of therapeutics aimed at protecting the synapses from the toxic binding of both proteins at the synapses can preserve synaptic health and cognitive function.
