AuX3-Mediated Selective Head-to-Head Dimerization of Difluoropropargyl Amides

2009

A dimerization of difluoropropargyl amides by reaction with gold(III) halides is described. A reductive elimination of a divinylgold species can be invoked to rationalize its formation. Initial studies of the unusual reactivity of these 1,4-dihalo-1,3-dienes have been performed.

New indications for the potential involvement of C–F-bonds in hydrogen bonding

2006

Abstract Solid state structures of a selection of 2-fluoro-2-phenylcyclopropane derivatives were examined by X-ray crystallography in order to identify short intermolecular contacts of C–F groups to H–X moieties (X=C, N). Particularly, several cis -configured fluorinated phenylcyclopropane derivatives showed extremely close intermolecular contacts. The shortest of such C–H⋯F–C-distances (2.17 A, C–F–H angle 162°) was found in (1 S ,2 R )-(2-fluoro-2-phenylcyclopropyl)methyl N -(4-bromophenyl)carbamate ( 8 ) and the closest N–H⋯F–C-interaction (2.01 A, C–F–H angle 167°) was found in (±)- cis -2-fluoro-2-phenylcyclopropyl carboxamide ( 4 ). Comparison of the structures of several of the fluor…

AICA-ribosiduria due to ATIC deficiency: Delineation of the phenotype with three novel cases, and long-term update on the first case.

2020

5-Amino-4-imidazolecarboxamide-ribosiduria (AICA)-ribosiduria is an exceedingly rare autosomal recessive condition resulting from the disruption of the bifunctional purine biosynthesis protein PURH (ATIC), which catalyzes the last two steps of de novo purine synthesis. It is characterized biochemically by the accumulation of AICA-riboside in urine. AICA-ribosiduria had been reported in only one individual, 15 years ago. In this article, we report three novel cases of AICA-ribosiduria from two independent families, with two novel pathogenic variants in ATIC. We also provide a clinical update on the first patient. Based on the phenotypic features shared by these four patients, we define AICA-…

Synthesis and antiproliferative activity of 3-amino-N-phenyl-1H-indazole-1-carboxamides

2007

Abstract A series of new 3-amino-N-phenyl-1H-indazole-1-carboxamides 10 have been prepared from commercially available phenyl isocyanate precursors 8 and 3-aminoindazole 9. Some of the synthesized compounds were evaluated for their in vitro antineoplastic activity against 60 human cell lines derived from seven clinically isolated cancer types (lung, colon, melanoma, renal, ovarian, brain, and leukemia) according to the NCI standard protocol. The test results indicated that 3-amino-1H-indazole-1-carboxamides 10 were endowed with an interesting antiproliferative activity. The most active compounds of this series, 10d,e, were able to inhibit cell growth of many neoplastic cell lines at concent…

Synthesis of substituted 3-amino-N-phenyl-1H-indazole-1-carboxamides endowed with antiproliferative activity

2010

Abstract Several new N-phenyl-1H-indazole-1-carboxamides 1c–h and 4l,m were prepared by reacting phenyl isocyanate derivatives 3a,b with 3-amino-1H-indazole derivatives 2c,e,g or 1H-indazole 2l respectively. Chemical transformations of compounds 1a,b and 1g,h gave 3-acetamido-N-phenyl-1H-indazole-1-carboxamide derivatives 5a,b, and 3,5-diamino-N-phenyl-1H-indazole-1-carboxamide derivatives 4i, j respectively. Finally, 3,5-diacetamido-N-phenyl-1H-indazole-1-carboxamide derivatives 6a,b were prepared by acetylation of 4i, j. Some of synthesized compounds were evaluated for their in vitro antiproliferative activity against the full NCI tumor cell lines panel derived from nine clinically isolat…

The Halide Binding Behavior of 2-Carbamoyl-7-ureido-1H-indoles: Conformational Aspects

2009

Indole-based anion receptors with an carboxamide unit in 2- and an urea in 7-position were prepared and found to bind halides (as well as acetate and nitrate) in chloroform solutions at room temperature. Investigations of the binding behaviour show that the receptor is selective for chloride. Surprisingly, the truncated receptor 3 without the 2-carbamoyl substituent shows the highest affinity for Cl–. Thorough 1H, 13C and 15N NMR investigations indicate different binding modes for acetate, nitrate and halides to the receptor 2. The observation of a major conformational change of this receptor during the binding of the halide ions leads to an understanding of the relative binding affinities …

Studies on the synthesis of pentacyclic strychnos indole alkaloids. photocyclization of n-chloroacetyl-1,2,3,4,5,6-hexahydro-1,5-methanoazocino[4,3-b…

1985

Abstract Photocyclization of 2-chloroacetyl-1,2,3,4,5,6-hexahydro-1, 5-methanoazocino[4,3-b]indole ( 5 ) takes place at the indole 4-position to give a 1 ,2 ,3 ,4 , 5 ,6-hexahydro-2 ,11-ethano-1 ,5-methanoazocino [4 , 3-6] indole system. Consequently, the method appears to be unsuitable for constructing the pyrrolidine ring of pentacyclic Strychnos indole alkaloids.

Synthesis, Characterization and Magnetic Properties of New Dinuclear Copper(II) Complexes with N,N′-Disubstituted Dithiooxamides derived from α-Amino…

2005

Four new dinuclear copper(II) complexes have been synthesized and have the general formula [Cu2(L)(H2O)2], where L = GLYDTO [N,N′-bis(carboxymethyl)dithiooxamide], ALADTO [N,N′-bis(carboxyethyl)dithiooxamide], VALDTO [N,N′-bis(1-carboxy-2-methylpropyl)dithiooxamide] and LEUDTO [N,N′-bis(1-carboxy-3-methylbutyl)dithiooxamide]. The complexes were characterized by elemental analysis as well as by IR, electronic and EPR spectroscopy. These techniques provided evidence for the presence of the CuNO2S chromophore. Magnetic susceptibility measurements on all the complexes in the range 4–300 K show the existence of a dominant antiferromagnetic interaction with −J values greater than 300 cm−1. Therma…

Heterocyclic photorearrangements. Photoinduced rearrangement of 3-styryl-1,2,4-oxadiazoles

1990

The photochemical behaviour of 3-styryl-5-phenyl-(5-methyl)-1,2,4-oxadiazoles in methanol at 254 nm has been investigated. A photoinduced rearrangement to the quinoline system has been pointed out and explained as proceeding through an initial photolysis of the ring ON bond, followed by a six membered ring closure reaction involving the styryl moiety.

Sheets of alternating chirality in the structure of a novel iron(III) complex with a cyclic oxamide ligand

1998

An organic–metalloorganic analogue of the inorganic clay minerals has been obtained from the cyclization of N,N′-bis(ethoxalyl)phenylenediamine in basic medium in the presence of iron(III) ions leading to formation of a cyclic oxamide ligand (L). The combination of intramolecular (covalent) interactions between metal and ligand and intermolecular (noncovalent) ionic interactions creates a novel layered compound with an intriguing crystal structure (shown on the right).
