Endothelial cells stimulate osteogenic differentiation of mesenchymal stem cells on calcium phosphate scaffolds

The interaction of mesenchymal stem cells (MSCs) with endothelium in vivo is significant for regenerative processes in organisms. To design concepts for tissue engineering for bone regeneration based on this interaction, the osteogenic differentiation of human bone marrow-derived MSCs in a co-culture with human dermal microvascular endothelial cells (HDMECs) was studied. Evidence for essential primary amino groups in a bacterial coupling factor F1ATPase.

Abstract We have found that the binding of pyridoxal-5′-phosphate to 6 primary amino groups leads to the inactivation of the enzyme. A preferential reaction of pyridoxal-5′-phosphate with the α-subunits of this enzyme can be demonstrated. The reactivity of the amino groups is influenced by various effectors. In the presence of ATP the inhibition of the ATPase activity is noncompetitive.

Identification and structural characterization of O-beta-ribosyl-(1"----2')-adenosine-5"-phosphate in yeast methionine initiator tRNA.

We report in this paper on the complete structure determination of the modified nucleotide A*, now called Ar(p), that was previously identified in yeast methionine initiator tRNA as an isomeric form of O-ribosyl-adenosine bearing an additional phosphoryl-monoester group on its ribose2 moiety. By applying immunologic, electrophoretic and enzymatic methods, extracts of different raft types of Culex pipiens were analysed. Rafts of the crosses Pa x Pa and Ha x Ha contained four common antigens, while unfertilized rafts of Pa and Ha (no antisera were prepared against them) and rafts of the crosses Og x Og, Og x Pa, and Pa x Og shared three common antigens with the remaining raft extracts. Interrelationships between Growth Yield, ATPase and Adenylate Kinase Activities inZymomonas mobilis

The presence of cytoplasmic and membrane-bound adenylate kinase (EC 2.7,4.3) as well as inorganic pyrophosphatase (EC 3.6.1.1) was detected in Zymomonas mobilis ATCC 29191. An increase in the molar growth yield (Y X/S ) of Z. mobilis under aerobic growth conditions appeared to be in proportion to a reduction of membrane-bound adenylate kinase (mAK) and ATPase activities and to an increase in cytoplasmic adenylate kinase (AK) activity. Lysosome-like particles in Geotrichum candidum : A cytochemical study

Lysosome-like particles were localized in Geotrichum candidum by means of the technique for the detection of acid phosphatase of PEARSE and staining with the vital dyes Brilliant cresyl blue and Neutral red. Pretreatments of the cells with procedures injuring the lysosome membrane resulted in shortening of the incubation time (2 min instead of 30 min). After exposure to 1% Triton X-100 the cells showed uniform strong staining due to the release of the enzyme into the cytoplasm.

Immobilization of enzymes by multilayer microcapsules

A method is described to obtain a new type of microcapsules for immobilization of enzymes or living cells. The wall of these capsules consists of several layers of poly(ethyleneimine) and poly(acrylic acid). The idea is that diverging properties of the whole assembly can be better controlled when the membrane is built up by several consecutive steps, each being optimized with respect to a special property, for example, permeability or mechanical strength. Kinetics of alkaline phosphatase from pig kidney. Influence of complexing agents on stability and activity

Metal ion-complexing agents, like KCN, EDTA etc., inactivate alkaline phosphatase of pig kidney. This inactivation is reversible at low concentrations of the complexing agents and irreversible at high concentrations. The reversible inhibition is probably due to removal of Zn2+ ions from the active site, where they are necessary for catalytic action, whereas the irreversible inhibition results from the removal of Zn2+ ions necessary for preservation of the structure. The inactivation is pseudo-first order.
