Data from: Transcriptomic data from panarthropods shed new light on the evolution of insulator binding proteins in insects

2017

Background Body plan development in multi-cellular organisms is largely determined by homeotic genes. Expression of homeotic genes, in turn, is partially regulated by insulator binding proteins (IBPs). While only a few enhancer blocking IBPs have been identified in vertebrates, the common fruit fly Drosophila melanogaster harbors at least twelve different enhancer blocking IBPs. We screened recently compiled insect transcriptomes from the 1KITE project and genomic and transcriptomic data from public databases, aiming to trace the origin of IBPs in insects and other arthropods. Results Our study shows that the last common ancestor of insects (Hexapoda) already possessed a substantial number …

Data from: Not only for egg yolk - functional and evolutionary insights from expression, selection and structural analyses of Formica ant vitellogeni…

2015

Vitellogenin, a storage protein, has been extensively studied for its egg-yolk precursor role, and it has been suggested to be fundamentally involved in caste differences in social insects. More than one vitellogenin copy has been reported in several oviparous species, including ants. However, the number and function of different vitellogenins, their phylogenetic relatedness and their role in reproductive queens and non-reproductive workers has been studied in few species only. We studied caste biased expression of vitellogenins in seven Formica ant species. Only one copy of conventional vitellogenin was identified in Formica species, and three vitellogenin homologues, derived from ancient …

FTIR studies of the high pressure dissociation of insulin and alpha-synuclein amyloids

2012

Amyloid fibrils are highly ordered aggregates whose formation occurs during the development of several serious disorders, like Althzeimer's and Parkinson's diseases. Even if most biophysical investigations of fibril formation have attempted to elucidate the structural events that occur during amyloid fibril assembly, relatively little is known about the thermodynamics of the aggregated protein state, and the kinetic mechanisms of its formation. Fibrillation is an irreversible process and a key challenge for the field is the development of therapeutic strategies able to inhibit or reverse the aggregation. The coupling of conventional Fourier Transform Infrared Spectroscopy (FTIR) and high Pr…

Shed vesicles are involved in the release of some leaderless proteins.

2006

Most proteins destined for secretion in the extracellular matrix are characterized by the presence of N-terminal signal peptides which direct their translocation into the endoplasmic reticulum, they are subsequently transferred to the Golgi apparatus and then secreted in the extracellular space. A growing number of secreted proteins, are being identified which, however, lack signal peptides allowing their entrance into the endoplasmic reticulum. They include the inflammatory cytokine interleukin 1b, galactins, macrophage migration inhibitory factor (MIF), acid and basic fibroblast growth factors (FGF-1, FGF-2) and Sphingosine kinase1(SphK-1). These proteins are secreted from the cell by unc…

Atomic mean square displacements in proteins by Molecular Dynamics: a case for analysis of variance

2004

Information on protein internal motions is usually obtained through the analysis of atomic mean-square displacements, which are a measure of variability of the atomic positions distribution functions. We report a statistical approach to analyze molecular dynamics data on these displacements that is based on probability distribution functions. Using a technique inspired by the analysis of variance, we compute unbiased, reliable mean-square displacements of the atoms and analyze them statistically. We applied this procedure to characterize protein thermostability by comparing the results for a thermophilic enzyme and a mesophilic homolog. In agreement with previous experimental observations, …
