Lung fixation for the preservation of air spaces.

2004

The procedures for the fixation of entire lungs of small rodents are presented together with various techniques used to verify the structural integrity of the lung tissue. To achieve this, the lungs were dissected out from rats and mice killed by ether overdose. The specimens were rinsed with isotonic saline and fixation solution under low vacuum conditions. After fixation, they were dried using alcohol and stored in a noncollapsed state (i.e., state of inhalation). Light and scanning electron microscopy as well as magnetic resonance imaging using hyperpolarized 3He were employed to verify the intact state on interalveolar septa and walls of smaller bronchi as well as accessibility of the a…

CD34+ progenitor to endothelial cell transition in post-pneumonectomy angiogenesis.

2012

In many species, pneumonectomy triggers compensatory lung growth that results in an increase not only in lung volume, but also in alveolar number. Whether the associated alveolar angiogenesis involves the contribution of blood-borne progenitor cells is unknown. To identify and characterize blood-borne progenitor cells contributing to lung growth after pneumonectomy in mice, we studied wild-type and wild-type/green fluorescence protein (GFP) parabiotic mice after left pneumonectomy. Within 21 days of pneumonectomy, a 3.2-fold increase occurred in the number of lung endothelial cells. This increase in total endothelial cells was temporally associated with a 7.3-fold increase in the number of …

Exceptional LAS Requests in Eurotransplant: Analysis of an 8-year Effort to Improve Lung Allocation for Precarious Patients

2020

PURPOSE: Following introduction of the lung allocation score (LAS) in 2011, Eurotransplant member centers can apply for an exceptional LAS (eLAS) if the calculated LAS insufficiently reflects the perceived transplant benefit for a patient, specifically in case of primary pulmonary hypertension group 1 and 4; combined lung+non-renal transplantation; rare diseases; or extracorporeal support. Each eLAS proposal is evaluated by a LAS Review Board, consisting of ≥3 lung transplant experts, which subsequently declines or approves the eLAS request in consensus of ≥3 votes. In case of a lower than accepted score, predefined business rules to assign LAS percentiles are used. METHODS: A retrospective…

Outcomes of Donor-Recipient Gender Mismatched Lung Transplantation in the Eurotransplant Area

2019

Purpose An effect of donor-recipient gender mismatching on long-term survival outcomes following organ transplantation has been postulated but remains controversial. We aimed to investigate the influence of gender mismatches on long-term survival after lung transplantation in the Eurotransplant area. Methods In this retrospective, multicentre study in the Eurotransplant area, all lung transplants performed from DBD lungs in the period from 01.01.2000 until 31.12.2015 were included. A total of 7688 lung transplant recipients (≥ 14 years old) for long-term survival and the effect of gender mismatches (Kaplan-Meier, multivariate regression). Results More men (4079; 52 %) were transplanted comp…

Pulmonary endarterectomy in chronic thromboembolic pulmonary hypertension.

2018

Management and outcome of patients with operable chronic thromboembolic pulmonary hypertension (CTEPH) who underwent pulmonary endarterectomy (PEA) at a large German referral center were investigated. In Germany, 394 PEAs were performed in 2014 and 2015 with an in-hospital mortality rate of 5.8%. Of these, 253 patients (64.2%) were treated at the Kerckhoff Clinic, Bad Nauheim, and 237 (93.7%; median age, 62 years [interquartile range [IQR], 52-72 years]; 46.0% female) were included in the present analysis. On referral, 52 patients (22.0%) were treated with pulmonary arterial hypertension-specific drugs and 95 (40.4%) were treated with non-vitamin K-dependent oral anticoagulants, and 14 (5.9%)…

Conclusions: Our findings indicate that unsupervised rehabilitation may represent an alternative in the management of post lung transplant patients.
