Susceptibility of Methyl 3-Amino-1H-pyrazole-5-carboxylate to Acylation

Abstract In the search for a new method of synthesis of hybrid peptides with aminopyrazole carboxylic acid, we tried to force selective acylation at the aromatic amino group instead of at the ring nitrogen atom with fairly gentle acylating agents. The acylating agents used were acid anhydrides: acetic anhydride, tert-butyl pyrocarbonate, and 2-(2-methoxyethoxy)ethoxyacetic acid/dicyclohexylcarbodiimide. We succceded in acylation at this amino group with almost none at the ring nitrogen atom. Sometimes, however, acylation in small quantities at the ring nitrogen atom was observed as a by-product. Addition reactions of heterocycles. VI. Reactions of 1,2-dimethylpyrrole and 1-methyl-2-carbomethoxypyrrole with nitrilimines

Addition reactions of 1,2-dimethylpyrrole and 1-methyl-2-carbomethoxypyrrole with C-acetyl-N-phenylnitrilimine, have been investigated. 1,2-Dimethylpyrrole gives three different types of adducts: i.e. bis-cycloadducts (Vc) and (VIc), spirocycloadduct (IX), and non cyclic bis-adduct (XII). On the other hand, 1-methyl-2-carbomethoxypyrrole gives the bis-cycloadduct (VIb) only. Compound XII arises probably through a double 1,3-addition reaction, whereas the formation of cycloadducts Vc, VIc, and IX depends on the substituents present at C2 of the pyrrole ring and consequentially on the intermediary occurence of mono-cycloadduct (IIIc), its methylenic tautomer VII, VIc, and XL The behaviour of …

ChemInform Abstract: Organocatalytic Enantioselective Synthesis of Pyrazoles Bearing a Quaternary Stereocenter.

An efficient one-pot asymmetric synthesis of pyrazoles bearing a chiral quaternary stereocenter has been developed. Quinine-derived thiourea catalyzed the enantioselective addition of pyrazolones to isatin-derived ketimines, providing the corresponding acetylated pyrazoles after in situ treatment with Ac2 O/Et3 N. The corresponding pyrazoles were afforded in high yields and excellent enantioselectivities.

Thermal decomposition of the diazonium sulfate derived from N-methyl-(1-phenyl-3-methylpyrazol-5-yl)-2-aminobenzamide afforded products formulated as 1-phenyl-3-methyl[2]benzopyrano[4,3-c]pyrazol-5-one (yield 10%), 1,4-dimethyl-3-phenylpyrazolo[3,4-c]isoquinolin-5-one (yield 10%), N-methyl-(1-phenyl-3-methylpyrazol-5-yl)-2-hydroxybenzamide (yield 8%) and 4′-hydroxy-2,3′-dimethyl-1′-phenylspiro[isoindoline-1,5′-[2]-pyrazolin]-3-one (yield 20%). The fragmentation reactions induced by electron impact of eighteen title compounds has been investigated with the aid of low beam energy spectra (14 eV, nom. value), metastable ion detection, high resolution measurements and labelling experiments. The loss of the 4-carbonyl oxygen together with the 3-substituent, which constitutes a characteristic fragmentation route of 3-aryl and 3-heteroaromatic substituted-4(3H)-quinazolinones, is again observed, but the presence of a carboxyethyl group at the 4′-position of the pyrazole ring is responsible of an anomalous loss of 47 daltons from the molecular ion. Unequivocal determination of isomeric products of reaction between 3-methyl-1-phenyl-2-pyrazoline-4,5-dione and aromatic 1,2-diamines

Abstract Regioselectivity of condensation of 3-methyl-1-phenyl-2-pyrazoline-4,5-dione with aromatic 1,2-diamines is dependent on substituent present. Isomeric 3-methyl-1-phenyl-1H-pyrazolo-[3,4-b]-quinoxaline products are distinguished by comparison of their 2D z-gradient selected 1H, 15N HMBC (Heteronuclear Multiple Bond Correlation) spectra. Multiplicity of H5 signal, which is recognizable by the cross-peak for CH3(3)-N4 and H5-N4 interactions, indicates substitution in position 6 or 7. The applied method is expected to be useful for structure determinations in other positional isomers.

Asymmetric Synthesis of Five-Membered Spiropyrazolones via N-Heterocyclic Carbene (NHC)-Catalyzed [3+2] Annulations

A new synthetic strategy for the asymmetric synthesis of five-membered spiropyrazolones via N-heterocyclic carbene-catalyzed [3+2] annulations employing enals and unsaturated pyrazolones as substrates has been developed. The new protocol allows the flexible variation of all four substituents of the pharmaceutically important spiropyrazolones in moderate to very good yields and in most cases with excellent diastereoselectivities and good to excellent enantioselectivities.

About the Reaction of „Pyrazolone Blue” with Diazomethane The reaction of “pyrazolone blue” with diazomethane was investigated. A pyridazinone derivative 3, a heterocyclic spiro-cyclopropyl product 4 and a compound 7, which contains three pyrazolone and two diazomethane entities, are formed. 3 reacts with an excess of diazomethane to the methoxy derivative 6. The spiro-cyclopropyl compound 4 is not stable and isomerizes to the corresponding 4,4′-methylidene-bispyrazolone 5. The structures of the products were established by spectroscopic methods and X-ray analysis.
