Electrical and mechanical activity of mammalian heart muscle fibres treated with papaverine

The action of papaverine on electrical and mechanical activity was investigated in ventricular and atrial heart muscle fibres from guinea-pigs and cats. 1. Papaverine (10−5 M–5×10−5M) had positive, negative or no inotropic effects in ventricular preparations; positive inotropic effects were not observed after pretreatment of the animals with reserpine. In atrial preparations, papaverine (2×10−5M) had a positive inotropic effect that was independent of endogenously stored catecholamines. 2. Influence of calcium on the effects of okadaic acid and its interaction with caffeine and theophylline in rat myometrium.

The effects of okadaic acid (OA), a monocarboxylic acid produced by marine dinoflagellates belonging to the genera Dinophysis and Prorocentrum, and their interactions with theophylline and caffeine were studied on the rat-isolated uterus in a calcium-containing medium and a calcium-free medium in the presence of 10−3 M EGTA. Okadaic acid (5 × 10−6 to 5 × 10−5 M) induced a concentration-dependent contraction of the rat-isolated uterus corresponding, with 5 × 10−5M, to 142.3±6.1% (n = 7) of the contraction induced by oxytocin 10−6 M. Penile Abflußvenenokklusion: Vergleich von erektiler Funktion und Kavernosometrie vor und nach perkutanen Eingriffen

After percutaneous transpenile or retrograde venous occlusion for the treatment of a cavernous leak, the clinical results were correlated with cavernosometric flow measurements in 27 patients. In 15 patients, venous occlusion led to a reduction of the maintenance flow: in patients who showed improvement there was an average reduction of 30 ml/min, in those without improvement or deterioration it averaged 0.3 or 13 ml/min respectively. In 7 patients there was increased flow (up to +21 ml/min). Multiple actions of glaucine on cyclic nucleotide phosphodiesterases, α1-adrenoceptor and benzothiazepine binding site at the calcium channel

1. In the present study, the properties of glaucine (an aporphine structurally related to papaverine) were compared with those of papaverine, diltiazem, nifedipine and prazosin. Hypertension and Impotence

In a group of 472 impotent patients who were evaluated with pharmacologic duplex sonography, 117 patient (24.8%) had a history of hypertension, 26 of them (22.2%) for more than 10 years. Objective data about the changes in pulsation, diameter and blood flow velocity of the penile arteries after papaverine injection and the resulting erectile response allowed indirect assessment of the penile venous function. Varying degrees of impaired arterial function were diagnosed in 85% of the patients. The duration of hypertension had less deteriorating effects on the penile arterial system than second risk factors such as diabetes mellitus (n = 31) or smoking (n = 26). Phosphodiesterase 10A in the Rat Pineal Gland: Localization, Daily and Seasonal Regulation of Expression and Influence on Signal Transduction

The cyclic nucleotide phosphodiesterase 10A (PDE10A) is highly expressed in striatal spiny projection neurons and represents a therapeutic target for the treatment of psychotic symptoms. As reported previously [J Biol Chem 2009; 284:7606–7622], in this study PDE10A was seen to be additionally expressed in the pineal gland where the levels of PDE10A transcript display daily changes. As with the transcript, the amount of PDE10A protein was found to be under daily and seasonal regulation. The effect of S-(+)-boldine on the α1-adrenoceptor of the guinea-pig aorta

1. The cardiovascular activity of S-(+)-boldine, an aporphine alkaloid structurally related to papaverine, was determined. The work includes functional studies on guinea-pig isolated aorta contracted with noradrenaline, caffeine, KCl or Ca2+, and on guinea-pig trachea contracted with acetylcholine or histamine. 2. Effects of papaverine on human isolated bladder muscle

Papaverine is a non-specific smooth muscle relaxant and is thought to act at a site beyond the receptor sites on the cell membrane. In this study the relaxing properties of papaverine were tested in isolated muscle strips from the human bladder dome. In carbachol-induced contractions papaverine, even in high concentrations of 10(-4) mol/l had virtually no effects on peak tension generation, whereas the fading was accelerated and the steady state tension at 30 min. was reduced by about 54%. 1. Noradrenaline 1 microM induced a contractile response in rat isolated aorta in the presence or in the absence of extracellular Ca2+ with depletion of intracellular Ca2+ stores. Thereafter, during incubation in the presence of Ca2+, an increase in the resting tone was observed. Such a contractile response did not occur after exposure to caffeine or 5-hydroxytryptamine. 2. Papaverine decreases the efflux of42K in guinea-pig atrial heart muscle

The effects of papaverine on resting potential and efflux of42K were investigated in guinea-pig left atria. Papaverine significantly reduced the potassium efflux in beating preparations. In resting preparations, the efflux of potassium was only slightly affected. However, the resting potential was significantly reduced by papaverine by about 5 mV.
