Purification and characterization of an intracellular family 3 β-glucosidase from Lactobacillus sanfranciscensis CB1

Lactobacillus sanfranciscensis CB1 uses cellobiose and other β-glucosides (methyl-β-glucoside, arbutin, amygdalin and salicin) as carbon sources. A hexameric ca. 288 kDa β-glucosidase was purified to homogeneity from Lact. sanfranciscensis CB1 by four chromatographic steps. The enzyme was optimally active at pH 7.5 and 40°C. It had a pI of ca. 4.38 and a D55°C value of ca. 27 sec. Almost total inhibition was found with sulfhydryl-modifying agents and divalent cations such as Cu2+, Co2+, Hg2+, Ni2+ and Fe2+ at a concentration of 2 mM. Maģistra darba ietvaros tiek izpētīta bezpalīdzības stāvokļa izpratne krimināltiesībās balstoties uz teoriju, normatīvo regulējumu un tiesu praksi, vēsturisko jēdziena izpratni, kā arī salīdzinot Latvijas regulējumu ar citām valstīm. Darba ietvaros tika analizētas vairākas grāmatas, periodika un likumi, kā arī veikta prokuroru aptauja. Rezultātā bezpalīdzības stāvoklis tika izpētīts un izdarīti secinājumi, kuri tiek atspoguļoti maģistra darba kopsavilkumā.
Pulsed Electric Field Processing Optimization of Ascorbic Acid in a Mango and Papaya Beverage Sweetened with Stevia Rebaudiana

During the processing of liquid foods, ascorbic acid may be altered, thus reducing beneficial health effects. For this reason, the degradation kinetics of ascorbic acid were determined in a fruit juice mixture (papaya and mango) sweetened with Stevia rebaudiana (SR) infusion after treatment by pulsed electric field (PEF) processing. The variable ranges were 20-40 kV/cm (electric field strength, E) during 100-360 μs (time treatment, t). The degradation equation was AA (mg/100 mL) = 26.842 - 0.101·E - 0.003·t- 10.371·%Stevia + 2.865·%Stevia2, demonstrating the use of PEF as an alternative to pasteurization treatments. In recent years, an increase in the consumer interest has been observed for craft beers, which are unpasteurized and unfiltered. Thus, the use of probiotic starters remaining in the craft beers could significantly increase health benefits. Here, the first study on the use of the probiotic Sacharomyces boulardii (Sb) yeast as single starter for craft brewing at 20 L scale is presented. The probiotic biomass growth was studied in bioreactor batch culture and modelled by the logistic and Gompertz equations. Finally, the probiotic biovolume of Sb was estimated from microscopy images. Training programs for career practitioners continue to increase in both quantity and quality. For example, international opportunities for those interested in career development facilitator training have increased substantially during the past decade. In this chapter we provide highlights of these developments as they have occurred throughout the world. The chapter details the substantial variability that exists regarding the training career practitioners receive. Finally, we make recommendations regarding minimal training standards and the need for more uniform language within our profession. peerReviewed
