A FRET-based assay for characterization of alternative splicing events using peptide nucleic acid fluorescence in situ hybridization

We describe a quantitative method for detecting RNA alternative splicing variants that combines in situ hybridization of fluorescently labeled peptide nucleic acid (PNA) probes with confocal microscopy Förster resonance energy transfer (FRET). The use of PNA probes complementary to sequences flanking a given splice junction allows to specifically quantify, within the cell, the RNA isoform generating such splice junction by FRET measure. Plasma from Ciona intestinalis, Phallusia mamillata and Ascidia malaca possess hemagglutinin for a variety of erythrocytes. Results obtained by physical and chemical treatments suggest that hemagglutinin for Phallusia mamillata and Ascidia malaca may be a protein or a protein-like substance.

The ontogenetic development and pattern of main digestive enzyme activities (proteases, carbohydrases and lipases) were investigated in farmed sharpsnout seabream Diplodus puntazzo larvae and juveniles, during the passage from the larval stage to adult (from 21 to 277 days after hatching - DAH). Larvae showed exponential growth, thus confirming that Rotifers and Artemia nauplii are suitable preys for early feeding of D. puntazzo larvae. Pepsin activity was low in the larvae, but it increased afterwards in relation with stomach development. Pasaulē un Latvijā novēro augstus mirstības rādītājus kuņģa vēža gadījumā sakarā ar pārsvarā asimptomātisko slimības norisi un vēlīno diagnostiku. Invazīvas skrīninga metodes, kā endoskopija ar biopsiju veikšanu, ir efektīvas tikai augsta riska reģionos. Līdz ar to nepieciešams izstrādāt jaunas, efektīvas neinvazīvas skrīninga metodes. Visplašāk tiek pētīti pepsinogēnu testi, kas identificē pacientus ar kuņģa gļotādas atrofiju. Darba mērķis bija noteikt pepsinogēnu līmeņus kuņģa vēža pacientiem, tā izvērtējot šo biomarķieru efektivitāti kuņģa vēža pacientu identifikācijai. Samples of Hizikia fusiforme edible seaweed, a commercially available dried food with high concentrations of total arsenic (t-As) and inorganic arsenic (i-As), both raw and cooked (boiling at 100 °C, 20 min), were selected for the bioaccessibility study. Cooking caused a significant reduction in the concentrations of t-As (30–43%) and i-As (46–50%), despite which the i-As contents in the cooked product were high (42.7–44.6 µg g−1 seaweed). Ievads. Neskatoties uz to, ka Kazahstānā ir augsta kuņģa vēža incidence, ir ierobežojami dati, kas informētu par H. pylori un kuņģa atrofiskā gastrīta izplatību šajā valstī. Darba mērķis. Noteikt un izanalizēt H. pylori infekcijas un atrofiskā gastrīta izplatību asimptomātiskiem 40 – 64 gadus jauniem Kazahstānas iedzīvotājiem. Materiāli un metodes. Dalībniekiem noteica anti-H. pylori IgG ≥10 U/mL un PGI, PGII ar lateksa-aglutinācijas metodi. Tika analizētas divas PG robežvērtības: PGI ≤ 70 ng/mL un PGI/II ≤ 3 un PGI ≤ 30 ng/mL un PGI/II ≤ 2. Biomarķieru metode kuņģa gļotādas atrofijas noteikšanai

Promocijas darba „Biomarķieru metode kuņģa gļotādas atrofijas noteikšanai” anotācija latviešu valodā Pētījumā tika veikta kuņģa biomarķieru metodes validācija, salīdzinot metodes rezultātus ar standartizēti veiktiem morfoloģijas izmeklējumu rezultātiem. Iegūtie dati parāda labu korelāciju starp PGI/II attiecību un kuņģa korpusa daļas atrofiju (p<0.0001) ar jutību (83.3%), kopējo precizitāti (86.7%). Biomarķieru samazinājums PGI un PGI/II tika noteikts atbilstoši paaugstinoties OLGA gastrīta stadijām. Purpose: The GISTAR Pilot study is part of a large multi-centre randomised trial conducted in Latvia with the main aim to evaluate preventive strategies to decrease mortality from gastric cancer in high-risk areas, especially in the Baltic States and Eastern Europe. Participants: A total of 3,447 apparently healthy, asymptomatic participants (40-64 y/o) were recruited between 2013 and 2015 for the GISTAR pilot study. Abstract Pretreatment with zinc acexamate (25–100 mg kg−1 i.p.) inhibited acid and pepsinogen secretion in the pylorus-ligated rat. Zinc acexamate (5–50 mg kg−1 p.o.) also inhibited the increases in acid secretion induced by carbachol (10 μg kg−1) and 2-deoxy-D-glucose (200 mg kg−1) in the perfused stomach of the anaesthetized rat. A delayed antisecretory effect was observed with this drug on histamine induced responses. High concentrations of zinc acexamate (10−5-10−2 M) did not modify the in-vitro activity of pepsin.
