Hypothalamic S-Nitrosylation Contributes to the Counter-Regulatory Response Impairment following Recurrent Hypoglycemia

Recurrent hypoglycemia (RH) impairs the counter-regulatory response (CRR) which restores euglycemia. During hypoglycemia, ventromedial hypothalamus (VMH) production of nitric oxide (NO) and activation of its receptor soluble guanylyl cyclase (sGC) are critical for the CRR. Hypoglycemia also increases brain reactive oxygen species (ROS) production. NO production in the presence of ROS causes protein S-nitrosylation. Centralizēti reģistrēto bioloģisko un biolīdzīgo medikamentu reimatoīdo slimību ārstēšanai pieejamība Latvijā

Bioloģiskajiem medikamentiem ir nozīmīga loma dažādu slimību terapijās, savukārt to biolīdzīgie analogi palīdz padarīt bioloģisko terapiju lētāku un pieejamāku pacientiem. Viena no slimībām, kur tiek izmantota bioloģiskā terapija, ir reimatoīdais artrīts (RA). Ja šī slimība progresē, tad nākotnē tā var novest pie nopietniem veselības traucējumiem vai pat invaliditātes, kas var radīt finansiālo slogu gan valstij, gan pacientam. Pētījumam veikšana tika izvēlētas kvantitatīvās datu ieguves metodes. Tika analizēti centralizēti reģistrēti 19 bioloģiskie un 36 biolīdzīgie medikamenti. Darbā aprakstīta triju mezglu bojājumpiecietīgas virtuālo serveru izmitināšanas vides izveide, kas sevī iekļauj koplietojamas augstas pieejamības disku apakšsistēmas izveidi un šī kompleksa bojājumpiecietības un ātrdarbības aspekti. Darba mērķis ir rast risinājumu bojājumpiecietīgas virtuālo serveru izmitināšanas vides izveidei, izmantojot trīs fiziskus serverus, kas atbilst iestādes norādītajām prasībām. Rezultātā izveidota strādājoša vide, kur vienas komponentes kļūme nerada risinājuma nodrošināto servisu zaudlaiku, kā arī noskaidrotas Ceph un augstas pieejamības virtualizācijas klastera izmantošanas nianses, ātrdarbība un risinājumi dažādu kļūmju situācijās.

Impact of uncharged and charged stabilizers on in vitro drug performances of clarithromycin nanocrystals

The purpose of this study was to evaluate the effect of charge on the in vitro drug performances of clarithromycin nanocrystals. To prepare different charges of nanocrystals, media milling was employed with the use of different stabilizing systems. The uncharged nanocrystals were prepared from poloxamer 407. The negatively and positively charged nanocrystals were stabilized using a combination of poloxamer 407 with sodium lauryl sulfate (SLS) and cetyltrimethylammonium bromide (CTAB), respectively. Formulation strategy towards minimizing viscosity mediated negative food effect on disintegration and dissolution of immediate release tablets.

Food induced viscosity can delay disintegration and subsequent release of API from solid dosage form which may lead to severe reduction in the bioavailability of BCS type III compounds. Formulations of such tablets need to be optimized in view of this postprandial viscosity factor. In this study, three super disintegrants, croscarmellose sodium (CCS), cross-linked polyvinylpolypyrrolidone (CPD), and sodium starch glycolate (SSG) were assessed for their efficiency under simulated fed state.
