Genetic lesions associated with Muller's ratchet in an RNA virus

The molecular basis of Muller's ratchet has been investigated using the important animal pathogen foot-and-mouth disease virus (FMDV). Clones from two FMDV populations were subjected to serial plaque transfers (repeated bottleneck events) on host BHK-21 cells. Relative fitness losses were documented in 11 out of 19 clones tested. Small fitness gains were observed in three clones. One viral clone attained an extremely low plating efficiency, suggesting that accumulation of deleterious mutations had driven the virus near extinction. New ground state constants of 12CH335Cl and 12CH337Cl from global polyad analysis

Abstract A global analysis of the infrared spectrum of chloromethane involving the ground state and the 13 vibrational states lying up to 2600 cm −1 was recently achieved using high resolution Fourier transform spectra of pure isotopomers. More than 20 000 transitions (cold and hot bands) for each isotopomer 12 CH 3 35 Cl and 12 CH 3 37 Cl have been assigned and fitted with a standard deviation of about 3 × 10 −4  cm −1 close to the experimental precison. This work incorporates and extends to the fifth polyad, the preliminary study of the lower four polyads published by [J. Mol. Spectrosc. 221 (2003) 199]. High resolution study of the and “hot” bands and ro-vibrational re-analysis of the polyad of the 32SO2 molecule

Abstract The weak “hot” absorption bands, ν 1 + 2 ν 2 − ν 2 and 2 ν 2 + ν 3 − ν 2 , were analysed with high resolution using the Fourier transform interferometer Bruker IFS-120 HR. In order to make possible an analysis of the ν 1 + 2 ν 2 − ν 2 and 2 ν 2 + ν 3 − ν 2 bands, as the first step, we re-analysed considerably the stronger “cold” bands, ν 1 + ν 2 and ν 2 + ν 3 , which are located in the same spectral regions. As the result of analysis we obtained about 2650 and 2050 transitions (1069 and 1001 upper state ro-vibrational energy values) with Jmax. = 78, Kamax. = 27 and Jmax. = 68, Kamax. = 24 for the bands ν 1 + ν 2 and ν 2 + ν 3 , respectively, that is considerably higher than in the …

Abstract The Spherical Top Data System (STDS) program suite developed in Dijon has been extended into two directions. First, the vibrational extrapolation is now possible for any kind of polyad scheme, this one being fully specified in the input parameters of the programs for hamiltonian and transition moment model calculations. Abstract Polyadenylation and splicing of heterogeneous nuclear RNA, two crucial steps in mRNA processing, are apparently enzymically mediated processes. This contribution summarizes the properties and the presumed functions of the known poly(A) catabolic enzymes (endoribonuclease IV and V, 2′,3′-exoribonuclease) as well as those of the pyrimidine-specific endoribonucleases associated with snRNP—hnRNP complexes (endoribonuclease VII, acidic p I 4.1 endoribonuclease and poly(U)-specific U1 snRNP-nuclease).

ISOLATION OF RAT CDNA CLONES CODING FOR THE AUTOANTIGEN SS-B/LA - DETECTION OF SPECIES-SPECIFIC VARIATIONS

Clones of cDNA coding for the autoantigen La (or SS-B) were isolated from a library made from rat liver. A comparison of the rat La cDNA (encoding from nt 38 to 1281 for rat La protein) with the sequences known for human and bovine La protein resulted in the identification of species-specific inserts. The inserts seem to be the result of multiplication of flanking sequences during evolution. In addition to these variations, we observed that rat La cDNAs exhibit non-canonical polyadenylation sites. Noncanonical RNAs from transcripts of the Drosophila muscleblind gene.

It has become increasingly evident that eukaryotic cells produce RNA molecules from coding genes with constitutions other than those of typically spliced mRNA transcripts. Here we describe new cDNAs from the Drosophila melanogaster muscleblind (mbl ) locus that identify two such atypical RNA molecules: RNAs containing an incomplete exon 2 tandem repetition (mblE2E2#) or having exons with a different order compared to the corresponding genomic DNA (mblE2E3#E2#; exon scrambling). The existence of exon duplications and rearrangements in the genomic locus that might explain such cDNAs was ruled out by genomic Southern blotting and in silico analysis of the Drosophila genome sequence.
