Invasive Observation by Atomic Force Microscope of a Langmuir-Blodgett Monolayer of Gramicidin

The properties of gramicidin, a linear antibiotic polypeptide of 15 amino acids, have been studied at the air-water interface. Analysis of the pressure-area isotherm is not able to conclude about the conformational behavior of gramicidin in the monolayer. Langmuir-Blodgett deposition of gramicidin layers onto a mica substrate has been developed for atomic force microscopy (AFM) observations. At high pressure of deposition, the gramicidin monolayer is composed of dimers perpendicular to the surface. The possibility of removing the half upper part of this dimer monolayer with the AFM tip is more in favor of a structure of single-stranded helical dimers.

Synthesis of new, UV-photoactive dansyl derivatives for flow cytometric studies on bile acid uptake.

Four new fluorescent derivatives of cholic acid have been synthesized; they incorporate a dansyl moiety at 3 alpha-, 3 beta-, 7 alpha- or 7 beta- positions. These cholic acid analogs are UV photoactive and also exhibit green fluorescence. In addition, they have been demonstrated to be suitable for studying the kinetics of bile acid transport by flow cytometry.

Spectroscopic study of the loading of cationic porphyrins by carbon nanohorns as high capacity carriers of photoactive molecules to cells

Carbon nanomaterials are attractive candidates for drug delivery due to their high surface area, ease of functionalisation and biocompatibility. This work describes the spectroscopic monitoring of the loading capacity of oxidised carbon nanohorns for two cationic porphyrins. Addition of the COOH functionalised carbon nanohorns to both meso-tetra(4-N-methylpyridyl) free base (H2TMPyP4) and platinum (PtTMPyP4) porphyrin in aqueous solution results in hypochromism of the Soret band and quenching of the porphyrin emission. DHEA-Bodipy–a functional fluorescent DHEA analog for live cell imaging

International audience; The androgen dehydroepiandrosterone (DHEA) has been reported to protect neuronal cells against dysfunction and apoptosis. Several signaling pathways involved in these effects have been described but little is known about the intracellular trafficking of DHEA. We describe design, synthesis and characterization of DHEA-Bodipy, a novel fluorescent DHEA analog. XAFS Studies of High-Valent Porphyrinato Manganèse Complexes with Ozone

We produce strong evidence of the interaction of ozone with a series of Manganese (III) porphyrinato complexes to form a well defined reactive intermediate. In the latter species, the Manganese atom which is formally in a tetravalent (IV) state, is (at least) hexacoordinated with the metal unambiguously lying in the basal plane of the four pyrrolic nitrogens of the porphyrin macrocycle. Substitution of Bromine or Chlorine axial ligands by ozonids is most likely to happen. This conclusion is also supported by XANES and EXAFS spectra recorded at the Chlorine K-edge which suggest that ozone may even form with chlorine a complex anion.

Carbonyl reductase 1 is a predominant doxorubicin reductase in the human liver.

A first step in the enzymatic disposition of the antineoplastic drug doxorubicin (DOX) is the reduction to doxorubicinol (DOX-OL). Because DOX-OL is less antineoplastic but more cardiotoxic than the parent compound, the individual rate of this reaction may affect the antitumor effect and the risk of DOX-induced heart failure. Using purified enzymes and human tissues we determined enzymes generating DOX-OL and interindividual differences in their activities. Human tissues express at least two DOX-reducing enzymes. The in vitro metabolic activation of dibenz[a,h]anthracene, catalyzed by by rat liver microsomes and examined by 32P-postlabelling.

DNA has been incubated in vitro with dibenz[a,h]anthracene (DB[a,H]A) and the related 5,6-diol and 3,4-diol in the presence of 3-methylcholanthrene- or Aroclor 1254-induced rat liver microsomes. After incubation, the DNA was extracted and examined for the presence of aromatic adducts using the nuclease P1 modification of the 32P-postlabelling technique. The maps of PEI-cellulose plates and autoradiography showed that 92% of the radioactivity contained in DB[a,h]A-DNA adduct spots is derived from the related 3,4-diol and that about 50% of the adducts may be formed following the conversion of this diol to the bay-region anti- and syn-3,4-diol 1,2-oxides.
