Convenient Synthesis of 3-Cinnamoyl-2-styrylchromones: Reinvestigation of the Baker-Venkataraman Rearrangement

An efficient and straightforward, one-pot sequence gives access to highly functionalized 3-cinnamoyl-2-styrylchromones in excellent yields. The low solubility of the target molecules allows convenient isolation. The formation of an α,α-dicinnamoylated acetophenone, as a consequence of a two-fold Baker-Venkataraman sequence, has to be anticipated.

Sulfur-incorporating cyclotriveratrylene analogues: the synthesis of cyclotrithioguaiacylene.

Cyclotriguaiacylene 1 is the universal precursor of cryptophanes, and represents an important intermediate for the preparation of functionalized cavitands of the cyclotriveratrylene family. Its thio analogue (cyclotrithioguaiacylene 3) was synthesized by two different routes, involving either the Newman-Kwart or the Pummerer rearrangement. The latter, performed starting from a trisulfoxide precursor, produced a purer compound in higher overall yield.

Transamidation reactions of 2-(2-sulfonylguanidino)acetamides

Abstract The reactivity of a series of sulfonylguanidinoacetamides 2A–E towards amines is reported. Guanidinoacetamides 2A–C, containing the arylsulfonylimino moiety, undergo a facile transamidation to give substituted carboxamides 4A–C, through the imidazolidinone intermediate 3. Acetamide 2D, having a methanesulfonylimino substituent, affords the imidazolidinone 3D and no transamidated carboxamides 4 are detected. In the case of guanidinoacetamide 2E, with a p-nitrobenzenesulfonylimino substituent, a Smiles rearrangement was observed.

1,2,4-Oxadiazole rearrangements involving an NNC side-chain sequence.

The thermal rearrangement of N-1,2,4-oxadiazol-3-yl-hydrazones into 1,2,4-triazole derivatives is reported. This represents the first example of a three-atom side-chain rearrangement involving an NNC sequence linked at the C(3) of the oxadiazole. The reactions carried out under solvent-free conditions produced good to high yields of the final products.

This chemical class concerns natural products, agrochemistry, molecular materials, biology and pharmacy, although it is certainly in coordination chemistry and in asymmetric catalysis using chiral transition metal complexes that P-chirogenic phosphorus compounds are the most used. Abstract The effect of β-cyclodextrin (β-CD) on the reactivity in the base-catalyzed pathway for the rearrangement in water of some ( Z )-hydrazones of 3-benzoyl-1,2,4-oxadiazoles ( 1b – f ) into the relevant triazoles ( 2b – f ) was investigated, finding different behavior as a function of the proton concentration. ESIMS and 1 H NMR data evidence the formation of host – guest complexes. The whole of the experimental and calculated (MM2) data enabled us to draw some intriguing conclusions concerning the influence of the structures of the substrates and the nature of the formed host – guest complexes on the real site of the reaction.

One-pot synthesis of fluorinated 2-amino-pyrimidine-N-oxides. Competing pathways in the four-atom side-chain rearrangements of 1,2,4-oxadiazoles

Abstract Trifluoromethylated 2-amino-pyrimidine N-oxides have been synthesized by reaction of the 3-amino-5-methyl-1,2,4-oxadiazole with trifluoromethyl-β-diketones in the presence of perchloric acid, followed by hydrolysis. In this ring-to-ring transformation an initial formation of (unisolated) 1,2,4-oxadiazole-pyrimidinium salts, and subsequent ring-opening at the oxadiazole moiety occurs. Isolation of 2-(hydroxyamino)-pyrimidine from the reaction mixture evidenced the presence of a competing pathway where the N(4) nitrogen of the oxadiazole is involved in the formation of a regioisomeric pyrimidinium salt. The effect of the trifluoromethyl group on the product distribution is discussed.…

Interplay of acidity and ionic liquid structure on the outcome of a heterocyclic rearrangement reaction

The study of suitable probe reactions is a powerful tool to investigate the properties of nonconventional solvents such ionic liquids (ILs). In particular, we studied the acid-catalyzed mononuclear rearrangement of heterocycles (MHR) of the Z-phenylhydrazone of 5-amino-3-benzoyl-1,2,4-oxadiazole into the relevant 1,2,3-triazole, in solution of ILs by means of kinetic measurements. We chose as solvents six ILs differing both in the cation and anion, in the presence of five carboxylic and sulfonic acids as catalysts.
