Extracellular matrix regulation of PTHrP and PTH/PTHrP receptor in a human breast cancer cell line

1999

AbstractIt was previously reported that 8701-BC breast cancer cells express the gene for parathyroid hormone-related peptide (PTHrP) and its cognate receptor (PTHrP-R), and release immunoreactive PTHrP in the extracellular medium; it was also found that PTHrP, in turn, exerts a role on the proliferative and invasive behavior in vitro of the same cell line. On the other hand, evidence has been produced that adhesion of 8701-BC cells onto different collagen substrates influences in various ways a number of phenotypic expressions, such as cell growth, motility, invasion of reconstituted basement membrane and production of lytic enzymes of the extracellular matrix (ECM). In light of these previ…

Endocannabinoid Role in Synaptic Plasticity and Learning

2009

Endocannabinoids have recently emerged as versatile modulators of synaptic transmission and can act as retrograde neurotransmitters. As they cannot be stored in synaptic vesicles, endocannabinoid signaling is believed to start ‘on-demand,’ via a stimulus-dependent synthesis from membranous precursors at the postsynaptic site. After synthesis, endocannabinoids bind presynaptically to cannabinoid type 1 (CB1) receptors, leading to a short- or long-term suppression of neurotransmitter release. CB1 receptors are present in a plethora of different synaptic connections in the brain. Electrophysiological and behavioral analyses of mutant mice lacking CB1 receptors and of pharmacologically treated …

Nicotinic receptor agonists as neuroprotective/neurotrophic drugs. Progress in molecular mechanisms

2007

In the present work we reviewed recent advances concerning neuroprotective/neurotrophic effects of acute or chronic nicotine exposure, and the signalling pathways mediating these effects, including mechanisms implicated in nicotine addiction and nAChR desensitization. Experimental and clinical data largely indicate long-lasting effects of nicotine and nicotinic agonists that imply a neuroprotective/neurotrophic role of nAChR activation, involving mainly alpha 7 and alpha 4 beta 2 nAChR subtypes, as evidenced using selective nAChR agonists. Compounds interacting with neuronal nAChRs have the potential to be neuroprotective and treatment with nAChR agonists elicits long-lasting neurotrophic e…

Impact of pharmacological and environmental manipulations on neuronal structural plasticity and dynamics

2017

The corticotrophin-releasing factor/urocortin system regulates white fat browning in mice through paracrine mechanisms.

2015

Objectives:\ud The corticotrophin-releasing factor (CRF)/urocortin system is expressed in the adipose tissue of mammals, but its functional role in this tissue remains unknown.\ud \ud Methods:\ud Pharmacological manipulation of the activity of CRF receptors, CRF1 and CRF2, was performed in 3T3L1 white pre-adipocytes and T37i brown pre-adipocytes during in vitro differentiation. The expression of genes of the CRF/urocortin system and of markers of white and brown adipocytes was evaluated along with mitochondrial biogenesis and cellular oxygen consumption. Metabolic evaluation of corticosterone-deficient or supplemented Crhr1-null (Crhr1−/−) mice and their wild-type controls was performed alo…

Système olfactif et neurobiologie

2006

Sex steroid hormone receptors, their ligands, and nuclear and non-nuclear pathways

2015

The ability of a cell to respond to a particular hormone depends on the presence of specific receptors for those hormones. Once the hormone has bound to its receptor, and following structural and biochemical modifications to the receptor, it separates from cytoplasmic chaperone proteins, thereby exposing the nuclear localization sequences that result in the activation of the receptor and initiation of the biological actions of the hormone on the target cell. In addition, recent work has demonstrated new pathways of steroid signaling through orphan and cell surface receptors that contribute to more rapid, “non-nuclear” or non-transcriptional effects of steroid hormones, often involving G-pro…

Sigma 1 receptora ietekme uz peļu mezenhimālo stromas šūnu osteodiferenciāciju

2021

A novel intrinsically disordered outer membrane lipoprotein ofAggregatibacter actinomycetemcomitansbinds various cytokines and plays a role in biofil…

2017

Intrinsically disordered proteins (IDPs) do not have a well-defined and stable 3-dimensional fold. Some IDPs can function as either transient or permanent binders of other proteins and may interact with an array of ligands by adopting different conformations. A novel outer membrane lipoprotein, bacterial interleukin receptor I (BilRI) of the opportunistic oral pathogen Aggregatibacter actinomycetemcomitans binds a key gatekeeper proinflammatory cytokine interleukin (IL)-1b. Because the amino acid sequence of the novel lipoprotein resembles that of fibrinogen binder A of Haemophilus ducreyi, BilRI could have the potential to bind other proteins, such as host matrix proteins. However, from th…
