Development and Characterization of Diamondback Moth Resistance to Transgenic Broccoli Expressing High Levels of Cry1C
2000
ABSTRACT A field-collected colony of the diamondback moth, Plutella xylostella , had 31-fold resistance to Cry1C protoxin of Bacillus thuringiensis . After 24 generations of selection with Cry1C protoxin and transgenic broccoli expressing a Cry1C protein, the resistance that developed was high enough that neonates of the resistant strain could complete their entire life cycle on transgenic broccoli expressing high levels of Cry1C. After 26 generations of selection, the resistance ratios of this strain to Cry1C protoxin were 12,400- and 63,100-fold, respectively, for the neonates and second instars by a leaf dip assay. The resistance remained stable until generation 38 (G38) under continuous…
In Vivo and In Vitro Binding of Vip3Aa to Spodoptera frugiperda Midgut and Characterization of Binding Sites by 125 I Radiolabeling
2014
ABSTRACT Bacillus thuringiensis vegetative insecticidal proteins (Vip3A) have been recently introduced in important crops as a strategy to delay the emerging resistance to the existing Cry toxins. The mode of action of Vip3A proteins has been studied in Spodoptera frugiperda with the aim of characterizing their binding to the insect midgut. Immunofluorescence histological localization of Vip3Aa in the midgut of intoxicated larvae showed that Vip3Aa bound to the brush border membrane along the entire apical surface. The presence of fluorescence in the cytoplasm of epithelial cells seems to suggest internalization of Vip3Aa or a fragment of it. Successful radiolabeling and optimization of the…
Les Républicains au Congrès
2015

C57BL/6-specific conditions for efficient in utero electroporation of the central nervous system.
2014
Abstract Background In utero electroporation is a fast an efficient tool to specifically address gene expression in the murine central nervous system. This technique was originally established in ICR/CD-1 outbred mice. Neuroanatomical differences between the different mouse strains and variations in gestation length require the optimization of the conditions for each strain to avoid severe complications. Furthermore the relevant position information is currently only scarcely standardized and not always easy to transfer to C57BL/6 mice. New method In this study we present an improved method for in utero electroporation of C57BL/6 including a detailed atlas that allows for specific and effic…
Effect of two novel bio-based rejuvenators on the performance of 50% RAP mixes - a statistical study on the complex modulus of asphalt binders and as…
2019
An experimental study was conducted to evaluate the effectiveness of two bio-additives as rejuvenators on the properties of asphalt mixtures containing 50% RAP and their binder constituents containing 37% RAP binder. Before mixing, the rejuvenators were blended with fresh bitumen and the extracted and recovered RAP bitumen, and changes in the rheological properties of the binders were assessed using performance grading (PG) criteria. The results showed that both rejuvenators could improve the low-temperature performance of the aged RAP binder and restore its low-temperature properties. Master curves for the unaged, RTFO-aged, and PAV aged blends were constructed using both the Christensen-A…
T-cell receptor transfer into human T cells with ecotropic retroviral vectors
2014
Adoptive T-cell transfer for cancer immunotherapy requires genetic modification of T cells with recombinant T-cell receptors (TCRs). Amphotropic retroviral vectors (RVs) used for TCR transduction for this purpose are considered safe in principle. Despite this, TCR-coding and packaging vectors could theoretically recombine to produce replication competent vectors (RCVs), and transduced T-cell preparations must be proven free of RCV. To eliminate the need for RCV testing, we transduced human T cells with ecotropic RVs so potential RCV would be non-infectious for human cells. We show that transfection of synthetic messenger RNA encoding murine cationic amino-acid transporter 1 (mCAT-1), the re…
Endothelin: an endothelium-derived vasoactive peptide and its possible role in the pathogenesis of cerebral vasospasm
1991
The contractile response to endothelin has been examined in cerebral arteries from rats subjected to a prior subarachnoid haemorrhage (SAH) and compared with saline injected controls. Endothelin elicited strong concentration-dependent contraction of rat basilar artery segments. The response was slow in onset and long lasting. The endothelin-induced contraction was much stronger in the SAH compared to control animals. Our findings suggest a role of the peptide in the pathophysiology of cerebral vasospasm.
Full-scale validation of bio-recycled asphalt mixtures for road pavements
2019
Abstract Recycling of asphalt has become a well-established practice in many countries, however the road pavement industry remains a bulk consumer of extracted raw materials. Novel solutions that find root in circular economy concepts and life-cycle approaches are needed in order to enable optimisation of infrastructure resource efficiency, starting from the design stage and spanning the whole value chain in the construction sector. Itis within this framework that the present study presents a full-scale validation of asphalt mixtures specifically designed to ensure durability of flexible road pavements and at the same time enabling the reuse of reclaimed asphalt pavement (RAP) through the i…
