Influence of double infections on the induction of thymidine kinase by UV-irradiated herpes simplex virus types 1 and 2 and pseudorabies virus

Simultaneous infection of primary rabbit kidney cells with HSV type 1 TK+ and a TK- strain results in a mutual influence of both viruses on the induction of thymidine kinase (TK). TK+ virus has an enhancing and TK- virus a depressing effect on TK induction by a superinfecting TK+ virus. The enzyme induction depends on the ratio of multiplicities of both viruses. The mutual influence on TK induction depends further on the time of addition of the superinfecting virus: the effect of the second virus can still be observed when given 6 hours after primary infection. Connu par des sources littéraires antiques tardives, le castrum de Dijon a laissé quelques traces dans l’urbanisme contemporain. Les cartes anciennes, l’étude des carnets de Louis-Bénigne Baudot, ainsi qu’un réexamen de visu des restes conservés permettent aujourd’hui de renforcer notre connaissance de l’enceinte tardive. L’enquête de terrain a ainsi permis de retrouver une inscription funéraire, inédite, gravée sur un « pyramidion », forme typique des monuments lingons. The thymidine kinase-complex isolated from herpes simplex virus type 1 and type 2 (HSV-1 and HSV-2) is associated with the following enzyme activities: ATP:dThd (dCyd) deoxypyrimidine kinase, ATP:dTMP thymidylate kinase, ADP:dThd- and AMP:dThd5′-phosphotransferase. In kinetic experiments it is shown that ara-AMP inhibits AMP:dThd- and ADP:dThd phosphotransferase activity, while acyclo-GMP impairs ADP:dThd phosphotransferase reaction only; the inhibition was found to be non-compertitive. The functional subunit ATP:dThd kinase was not affected by either compound.

Water-soluble polysaccharides from Opuntia stricta Haw. fruit peels: Recovery, identification and evaluation of their antioxidant activities

Abstract Opuntia stricta Haw. is considered as one of the most common cactus plant growing in Tunisia. Extracting valuable compounds from its fruit peel, considered as by-product, is drawing more and more attention, making it on the verge of commercialization. Water-soluble polysaccharides were extracted from Opuntia stricta Haw. peels, and their chemical composition assessed using thin layer chromatography. The antioxidant activities of the extracted polysaccharides were assessed using 2,2-diphenyl-1-picrylhydrazyl free radical scavenging activity, total antioxidant activity and reducing power capacity. Hemicellulose hydrolysis catalysed by solid acids

Depolymerising hemicellulose into platform sugar molecules is a key step in developing the concept of an integrated biorefinery. This reaction is traditionally catalysed by either enzymes or homogeneous mineral acids. We compared various solid catalysts for hemicellulose hydrolysis, running reactions in water, under neutral pH and relatively mild temperature and pressure (120°C and 10 bar) conditions. Sulphonated resins are highly active, but they leach out sulphonic groups. Sulphonated silicas are less active, but more stable. Biopolymer interactions affect the functional properties of edible films based on agar, cassava starch and arabinoxylan blends

Abstract Edible films based on the binary combination of agar (AG), cassava starch (CAS) and arabinoxylan (AX) were studied with regard to their microstructure, moisture barrier and mechanical properties. Though the films appear macroscopically homogeneous, electron microscopy observations reveal a phase separation and dispersion, respectively, in AG–CAS and AG–AX blend film structures, whereas the structure of CAS–AX blend films seems homogeneous. Inhibition of herpesvirus DNA synthesis by 9-beta-D-arabinofuranosyladenine in cellular and cell-free systems.

9-beta-D-Arabinofuranosyladenine 5'-triphosphate (ara-ATP) is an inhibitor both of DNA polymerase-alpha and -beta from noninfected rabbit kidney cells and of the DNA-dependent DNA polymerase induced by herpes simplex virus Type 1 (strain IES). The studies were performed with partially purified enzymes, and each of the different polymerase preparations contained only one DNA-dependent DNA polymerase species. These enzymes were inhibited in a competitive manner. The HSV-induced DNA-dependent DNA polymerase was 39-fold more sensitive to ara-ATP than was cellular DNA polymerase-beta and 116-fold more sensitive than cellular DNA polymerase-alpha. Arabinosylamine in Asymmetric Syntheses of Chiral Piperidine Alkaloids.

The stereodifferentiating potential of arabinosyl aldimines was utilized in stereoselective syntheses of 2-substituted dehydropiperidinones and their further transformation to 2,6-cis-substituted piperidinones. The absolute configuration was proven by X-ray analysis and by the synthesis of the enantiomerically pure alkaloid (+)-dihydropinidine. The presented method offers the possibility to synthesize piperidine derivatives enantiomeric to those obtained by the application of the corresponding galactosylamine auxiliary.
