La pression partielle de l'oxygene et la «clearance» de l'hydrogene ont ete mesurees dans des regions circonscrites. Les deux valeurs sont enregistrees par la meme microelectrode de platine (diametre de la pointe 10μ). Cette methode nouvelle est appliquee dans des experiences faites sur le cerveau de Rats narcotises et on est capable d'evaluer les inhomogenites locales de l'apport d'oxygene dependant du flux du sang.

A two-circuit apparatus for the perfusion of the isolated rat brain.

A perfusion apparatus for the isolated rat brain is described which has two recirculating systems running synchronously. Instantaneous transfer from one circulating system to the other is possible by use of a 2-way stopcock, enabling different media to be perfused through the brain without interrupting the perfusion. The feasibility of the system is demonstrated experimentally.

The effect of β-mercaptoethanol on lactate dehydrogenase from rat brain

Abstract Catalytic studies on lactate dehydrogenase from rat brain in the presence of β-mercaptoethanol have been performed. It has been found that pyruvate reduction and lactate dehydrogenation were differently affected by the addition of the sulfhydryl compound to the reaction mixture. The effect of β-mercaptoethanol on the catalyzed reaction has been shown to be dependent on the hydrogen ion concentration. When lactate dehydrogenation is measured, β-mercaptoethanol causes a net enhancement of the reaction rate at the more acidic pH values.

The uptake of promazine, chlorpromazine, desmethylpromazine, and desmethylchlorpromazine into the isolated rat brain was determined in relation to their lipophilic character and their binding to albumin and erythroeytes in the perfusion media. The desmethylmetabolites showed about the same degree of binding as their parent compounds promazine and chlorpromazine. On the other hand the partition coefficients of promazine and chlorpromazine were about 10 times higher than those of their desmethylmetabolites. Post-transcriptional regulation of mRNA metabolism is involved in processes as different as cell fate specification in development and cell response to a large variety of environmental cues. Regulation of all steps of RNA metabolism depends on RNA-binding proteins (RBPs). By using a T1 RNase protection assay, we previously identified three H1° RNA-binding factors (p40, p70 and p110), highly expressed in the rat brain. Thyroid hormones induce sumoylation of the cold shock domain-containing protein PIPPin in developing rat brain and in cultured neurons.

We previously identified a cold shock domain (CSD)-containing protein (PIPPin), expressed at high level in brain cells. PIPPin has the potential to undergo different post-translational modifications and might be a good candidate to regulate the synthesis of specific proteins in response to extracellular stimuli. Here we report the effects of thyroid hormone (T3) on PIPPin expression in developing rat brain. We found that a significant difference among euthyroid- and hypothyroid- newborn rats concerns sumoylation of nuclear PIPPin, that is abolished by hypothyroidism.
