The binding of lipophilic drugs to albumin and erythrocytes and its importance for the uptake of the drugs into the isolated rat liver were investigated. Promazine and chlorpromazine were taken as lipophilic drugs, which were compared in some experiments with 3,3′-dimethyl-4,4′-diacetyldiphenylbisguanylhydrazone (BG 60), a much more hydrophilic drug.

Bestimmung von Glykolyse-Metaboliten in isolierten Zellkernen

Isolated rat liver nuclei have been analyzed for the intranuclear concentration of 8 glycolytic metabolites, and in addition for the 3 adenine nucleotides, and for malate. All these substrates are to be found in nuclei in a range of concentration comparable to that in the whole tissue. The consequences of these findings with respect to the definite establishment of glycolysis in isolated nuclei, as well as for the intranuclear formation of energy-rich phosphate bonds, are discussed.

Experiments on the metyrapone reducing microsomal enzyme system.

The formation of reduced metyrapone [2-methyl-1.2-bis-(3-pyridyl)-1-propanol] from metyrapone [2-methyl-1.2-bis-(3-pyridyl)-1-propanone] in the liver has been studied. Reduced metyrapone appeared as the main metabolite of metyrapone in the isolated perfused rat liver. Experiments on the intracellular distribution of the metyrapone reducing activity revealed that metyrapone reductase is mainly localized in the microsomal fraction. In the 105,000 × g supernatant only a little activity was found.

Glutamic dehydrogenase of rat liver mitochondria is inhibited by phenylbutazone at a final concentration of 2,5 mg-% (0,81 · 10−4 M). However, glutamic dehydrogenase in heart muscle homogenate and the cristalline enzyme from liver are not inhibited by phenylbutazone up to 10 mg-% (3,24 · 10−4 M), but they are inhibited by a metabolite arising during incubation of liver mitochondria with phenylbutazone.

Effect of nutritional imbalances on cytochrome P-450 isozymes in rat liver

Male Sprague-Dawley rats were fed for six weeks either a control diet containing 22% casein (C) and 5% fat (F) or a low-protein diet (6% C, 5% F) or high-lipid diet (30% C, 30% F). A group of rats received a control diet containing 50 ppm of Phenoclor DP6. Three major forms of cytochrome P-450, UT 50, BP 3a and MC 2 were purified from livers of DP6-fed rats and only two forms, UT 50 and PB 3a, were purified from control and dietary groups. The amino acid composition and the catalytic activities towards all substrates tested were only significantly modified in the purified UT 50 P-450 isozyme from rats fed the low-protein diet. We have recently shown that the uptake of amino acids by rat lactating mammary gland is 15 mmoles/day (1). This effect is achieved by an increase in food intake during lactation and probably by changes in amino acid metabolism in other tissues. Rat liver removes 75% of the total amino acids derived from a protein meal (2); therefore plays a key role in the regulation of plasma amino acid concentration.
