Anaerobic Reduction of Glycerol to Propanediol-1.3 by Lactobacillus brevis and Lactobacillus buchneri

1984

Summary Three strains of Lactobacillus brevis and one strain of Lactobacillus buchneri grew very poorly on glucose. Good growth was observed on glucose plus glycerol; while glucose was fermented to acetate or ethanol, lactate and CO 2 , glycerol was dehydrated to 3-hydroxypropanal and subsequently reduced to propanediol-1.3. Cell extracts of L. brevis and L. buchneri grown on glucose plus glycerol contained a B 12 -dependent glycerol dehydratase and a propanediol-1.3 dehydrogenase. Glycerol was not metabolized when used as the only substrate. Fructose as sole carbon source was partially reduced to mannitol. The joint fermentation of fructose and glycerol yielded propanediol-1.3 from glycero…

Radioactively labelled epoxides part II. (1) tritium labelled cyclohexene oxide, transstilbene oxide and phenanthrene 9,10-oxide

1980

Tritium labelled cyclohexene oxide, trans-stilbene oxide and phenanthrene 9,10-oxide were prepared with specific activities of 0.7 - 1.1 mCi per mmole starting with monoor diketo compounds. Tritium was introduced by reducing the ketone precursors with tritiated complex metal hydrides. The resulting alcohols were transformed to the epoxides by methods described for the unlabelled compounds. The syntheses require only two or three steps and yield cyclohexene oxide, trans-stilbene oxide and phenanthrene 9,10-oxide, important substrates for the study of epoxide hydratase and glutathione S-transferases in high radiochemical purity.

Are Heme-Dependent Enzymes Always Using a Redox Mechanism? A Theoretical Study of the Kemp Elimination Catalyzed by a Promiscuous Aldoxime Dehydratase

2020

The design of biocatalysts is a goal to improve the rate, selectivity and environmental friendship of chemical processes in biotechnology. In this regard, the use of computational techniques has provided valuable assistance in the design of enzymes with remarkable catalytic activity. In this paper, hybrid QM/MM simulations have allowed getting an insight into the mechanism of a promiscuous aldoxime dehydratase (OxdA) for the Kemp elimination. We first demonstrate that, based on the use of linear response approximation (LRA) methods, the lowest energy electronic state of the benzisoxazole placed in the active sit of OxdA corresponds to a singlet state, being the triplet and the quintet state…

Dihydrodiol Dehydrogenase: An Important Enzyme in Dihydrodiol-Epoxide Pathway — Mediated Benzo(A)Pyrene Mutagenicity

1978

Benzo(a)pyrene is metabolized to two major groups of mutagenically reactive metabolites: Monofunctional epoxides and dihydrodiol-epoxides. Various monooxygenase forms catalyze the various pathways at very different rates. In metabolic situations where the contribution by dihydrodiol-epoxides is small, epoxide hydratase represents a very efficient protective system. However, in situations where the mutagenic effect is predominately due to dihydrodiol-epoxide, the effect of epoxide hydratase is complicated and weak. We have now obtained evidence that a dihydrodiol dehydrogenase represents an efficient protective system in the latter situation. The enyzme was purified to homogeneity and the pu…

Enzymes as Regulators of Toxic Reactions by Electrophilic Metabolites

1979

Conversion of many compounds which are not electrophilically reactive as such to metabolites responsible for cytotoxic, mutagenic and/or carcinogenic effects is catalyzed by mammalian enzymes. Many reactive agents, whether metabolites or parent compounds, are also subject to inactivation by mammalian enzymes.

Identification and characterization of autoantibodies against catalase and α-enolase in patients with primary sclerosing cholangitis

1998

SUMMARY Primary sclerosing cholangitis (PSC) is a chronic cholestatic liver disease of unknown aetiology. Recent studies have shown that genetic factors and both cellular and humoral immunological abnormalities are important in the pathogenesis of PSC. The most prominent autoantibodies in PSC are anti-neutrophil cytoplasmic antibodies (ANCA). The autoepitopes of ANCA in PSC are not well defined. The aim of this study was to identify corresponding ANCA autoantigens in patients with PSC. A biochemical approach with enrichment and partial purification of soluble neutrophil proteins, detection of autoantibodies by Western blot and partial amino acid sequencing were used. Two new autoantigen/aut…

Analysis of the Cellular Roles of MOCS3 Identifies a MOCS3-Independent Localization of NFS1 at the Tips of the Centrosome

2019

The deficiency of the molybdenum cofactor (Moco) is an autosomal recessive disease, which leads to the loss of activity of all molybdoenzymes in humans with sulfite oxidase being the essential protein. Moco deficiency generally results in death in early childhood. Moco is a sulfur-containing cofactor synthesized in the cytosol with the sulfur being provided by a sulfur relay system composed of the L-cysteine desulfurase NFS1, MOCS3, and MOCS2A. Human MOCS3 is a dual-function protein that was shown to play an important role in Moco biosynthesis and in the mcm(5)s(2) U thio modifications of nucleosides in cytosolic tRNAs for Lys, Gln, and Glu. In this study, we constructed a homozygous MOCS3 …
