Anaerobic Reduction of Glycerol to Propanediol-1.3 by Lactobacillus brevis and Lactobacillus buchneri

Summary Three strains of Lactobacillus brevis and one strain of Lactobacillus buchneri grew very poorly on glucose. Good growth was observed on glucose plus glycerol; while glucose was fermented to acetate or ethanol, lactate and CO 2 , glycerol was dehydrated to 3-hydroxypropanal and subsequently reduced to propanediol-1.3. Cell extracts of L. brevis and L. buchneri grown on glucose plus glycerol contained a B 12 -dependent glycerol dehydratase and a propanediol-1.3 dehydrogenase. Glycerol was not metabolized when used as the only substrate. Fructose as sole carbon source was partially reduced to mannitol. Tritium labelled cyclohexene oxide, trans-stilbene oxide and phenanthrene 9,10-oxide were prepared with specific activities of 0.7 - 1.1 mCi per mmole starting with monoor diketo compounds. Tritium was introduced by reducing the ketone precursors with tritiated complex metal hydrides. The resulting alcohols were transformed to the epoxides by methods described for the unlabelled compounds. The syntheses require only two or three steps and yield cyclohexene oxide, trans-stilbene oxide and phenanthrene 9,10-oxide, important substrates for the study of epoxide hydratase and glutathione S-transferases in high radiochemical purity.

Benzo(a)pyrene is metabolized to two major groups of mutagenically reactive metabolites: Monofunctional epoxides and dihydrodiol-epoxides. Various monooxygenase forms catalyze the various pathways at very different rates. In metabolic situations where the contribution by dihydrodiol-epoxides is small, epoxide hydratase represents a very efficient protective system. However, in situations where the mutagenic effect is predominately due to dihydrodiol-epoxide, the effect of epoxide hydratase is complicated and weak. SUMMARY Primary sclerosing cholangitis (PSC) is a chronic cholestatic liver disease of unknown aetiology. Recent studies have shown that genetic factors and both cellular and humoral immunological abnormalities are important in the pathogenesis of PSC. The most prominent autoantibodies in PSC are anti-neutrophil cytoplasmic antibodies (ANCA). The autoepitopes of ANCA in PSC are not well defined. The deficiency of the molybdenum cofactor (Moco) is an autosomal recessive disease, which leads to the loss of activity of all molybdoenzymes in humans with sulfite oxidase being the essential protein. Moco deficiency generally results in death in early childhood. Moco is a sulfur-containing cofactor synthesized in the cytosol with the sulfur being provided by a sulfur relay system composed of the L-cysteine desulfurase NFS1, MOCS3, and MOCS2A.
