Bypassing hazard of housekeeping genes: their evaluation in rat granule neurons treated with cerebrospinal fluid of multiple sclerosis subjects

2015

Gene expression studies employing real-time PCR has become an intrinsic part of biomedical research. Appropriate normalization of target gene transcript(s) based on stably expressed housekeeping genes is crucial in individual experimental conditions to obtain accurate results. In multiple sclerosis (MS), several gene expression studies have been undertaken, however, the suitability of housekeeping genes to express stably in this disease is not yet explored. Recent research suggests that their expression level may vary under different experimental conditions. Hence it is indispensible to evaluate their expression stability to accurately normalize target gene transcripts. The present study ai…

Quantitative Detection of the nosZ Gene, Encoding Nitrous Oxide Reductase, and Comparison of the Abundances of 16S rRNA, narG , nirK , and nosZ Genes…

2006

ABSTRACT Nitrous oxide (N 2 O) is an important greenhouse gas in the troposphere controlling ozone concentration in the stratosphere through nitric oxide production. In order to quantify bacteria capable of N 2 O reduction, we developed a SYBR green quantitative real-time PCR assay targeting the nosZ gene encoding the catalytic subunit of the nitrous oxide reductase. Two independent sets of nosZ primers flanking the nosZ fragment previously used in diversity studies were designed and tested (K. Kloos, A. Mergel, C. Rösch, and H. Bothe, Aust. J. Plant Physiol. 28:991-998, 2001). The utility of these real-time PCR assays was demonstrated by quantifying the nosZ gene present in six different …

The Conserved Foot Domain of RNA Pol II Associates with Proteins Involved in Transcriptional Initiation and/or Early Elongation

2011

RNA polymerase (pol) II establishes many protein-protein interactions with transcriptional regulators to coordinate different steps of transcription. Although some of these interactions have been well described, little is known about the existence of RNA pol II regions involved in contact with transcriptional regulators. We hypothesize that conserved regions on the surface of RNA pol II contact transcriptional regulators. We identified such an RNA pol II conserved region that includes the majority of the >foot> domain and identified interactions of this region with Mvp1, a protein required for sorting proteins to the vacuole, and Spo14, a phospholipase D. Deletion of MVP1 and SPO14 affects …

Molecular biology approaches utilized in preimplantation genetics: real-time PCR, microarrays, next-generation sequencing, karyomapping, and others

2020

Abstract Over the past few decades the development of new molecular technologies has revolutionized diagnosis in the reproductive medicine field, with the evaluation of the two main factors involved in a successful pregnancy: the embryo and the endometrium. The detection of genetically abnormal embryos, as well as the identification of an optimum endometrium using transcriptomics have become a priority in assisted reproductive treatments to increase pregnancy rates. This chapter provides an overview of the molecular techniques currently employed in assisted reproduction for embryo evaluation such as preimplantation genetic testing karyotyping, fluorescence in situ hybridization, polymerase …

The Near Real Time Ionospheric Model of Latvia

2015

A highly accurate ionosphere model is necessary to enable a fast and reliable coordinate determination with GNSS in real time. It is a partially ionized atmospheric region ranging up to 1,000 km height, affected by spatial variations, space weather, seasonal and solar cycle dependence. New approaches and algorithms of modelling techniques are sought to provide better solutions in the territory of Latvia. Ionospheric TEC value has large differences in Western Latvia and Eastern Latvia. Actual ionospheric map should be calculated and delivered to the surveyors near real time and published on the WEB. Delivering actual map to rover GNSS devices in a field will provide the surveyors with ionosp…

Establishment of a quantitative RT-pCR for detection of vascular cell adhesion molecule-1 transcripts in endothelial cells after stimulation with adv…

1998

Advanced glycation endproducts (AGE) are supposed to increase endothelial expression of adhesion molecules like vascular cell adhesion molecule-1 (VCAM-1) by inducing an intracellular stress with subsequent activation of nuclear transcription factor NF-kappa-B. Quantitative analysis of VCAM-1-transcription has not been demonstrated concerning this topic. Thus, the aim of this study was to establish quantitative reverse transcription polymerase chain reaction (RT-PCR) assays using a spacer gene in order to measure the amounts of specific mRNA for VCAM-1 in human umbilical vein endothelial cells (HUVEC) which were stimulated with AGE-albumin (AGE-BSA). A recombinant RNA-standard was synthesiz…

ARQ Aware Scheduling for the IEEE 802.16 Base Station

2008

The IEEE 802.16 technology defines the ARQ mechanism that enables a connection to resend data at the MAC level if an error is detected. In this paper, we analyze the ARQ aware scheduling for the 802.16 base station. In particular, we consider how the BS scheduler can account for the ARQ block size, absence of the ARQ block rearrangement, and the ARQ transmission window. We propose a set of constraints that can be applied to any base station scheduler algorithm. To test them, we run a number of simulation scenarios. The simulations results confirm that the ARQ aware scheduling can improve the overall performance.

Design of a Generic 3-D Scene Generator for Passive Optical Missions and Its Implementation for the ESA’s FLEX/Sentinel-3 Tandem Mission

2018

During the design phase of a satellite mission, end-to-end mission performance simulator (E2ES) tools allow scientists and engineers evaluating the mission concept, consolidating system technical requirements and analyzing the suitability of the implemented technical solutions and data processing algorithms. The generation of synthetic scenes is one of the core parts of an E2ES, providing scenes (ground truth) as would be observed by satellite instruments and used as reference against simulated retrieved mission products. An appropriate generation of the scene also allows assessing the performance of the ground data processing chain replacing real instrument data before the mission is in or…

Extracellular cyclic GMP and its derivatives GMP and guanosine protect from oxidative glutamate toxicity.

2013

Cell death in response to oxidative stress plays a role in a variety of neurodegenerative diseases and can be studied in detail in the neuronal cell line HT22, where extracellular glutamate causes glutathione depletion by inhibition of the glutamate/cystine antiporter system xc(-), elevation of reactive oxygen species and eventually programmed cell death caused by cytotoxic calcium influx. Using this paradigm, we screened 54 putative extracellular peptide or small molecule ligands for effects on cell death and identified extracellular cyclic guanosine monophosphate (cGMP) as a protective substance. Extracellular cGMP was protective, whereas the cell-permeable cGMP analog 8-pCPT-cGMP or the …

Fully pipelined real time hardware solution for High Efficiency Video Coding (HEVC) intra prediction

2016

International audience; A fully pipelined hardware accelerator for the High Efficiency Video Coding (HEVC) intra prediction is presented in this paper in order to reduce the computation complexity coming with this module and to accelerate the concerned calculations. Two reconfigurable structures are developed in this paper, the first one concerns angular modes and is identified as Processing Element for Angular (PEA) modes, the other is made in order to handle with the Planar mode and is identified as Processing Element for the Planar (PEP) mode. Each structure is repeated in five paths, that our architecture composed of, working in parallel way. This architecture supports all intra predict…
