The Stress-Inducible Protein DRR1 Exerts Distinct Effects on Actin Dynamics.

2018

Cytoskeletal dynamics are pivotal to memory, learning, and stress physiology, and thus psychiatric diseases. Downregulated in renal cell carcinoma 1 (DRR1) protein was characterized as the link between stress, actin dynamics, neuronal function, and cognition. To elucidate the underlying molecular mechanisms, we undertook a domain analysis of DRR1 and probed the effects on actin binding, polymerization, and bundling, as well as on actin-dependent cellular processes. Methods: DRR1 domains were cloned and expressed as recombinant proteins to perform in vitro analysis of actin dynamics (binding, bundling, polymerization, and nucleation). Cellular actin-dependent processes were analyzed in trans…

Seasonal and spatial dynamics of enteric viruses in wastewater and in riverine and estuarine receiving waters

2018

International audience; Enteric viruses represent a global public health threat and are implicated in numerous foodborne andwaterborne disease outbreaks. Nonetheless, relatively little is known of their fate and stability in the environment. In this studywe used carefully validatedmethods tomonitor enteric viruses, namely adenovirus (AdV), JC polyomavirus (JCV), noroviruses (NoVs), sapovirus (SaV) and hepatitis A and E viruses (HAV and HEV) fromwastewater source to beaches and shellfish beds. Wastewater influent and effluent, surface water, sediment and shellfish samples were collected in the Conwy catchment (North Wales, UK) once a month for one year. High concentrations of AdV and JCV wer…

Multi-approach metabolomics analysis and artificial simplified phytocomplexes reveal cultivar-dependent synergy between polyphenols and ascorbic acid…

2017

Fruits of the sweet cherry (Prunus avium L.) accumulate a range of antioxidants that can help to prevent cardiovascular disease, inflammation and cancer. We tested the in vitro antioxidant activity of 18 sweet cherry cultivars collected from 12 farms in the protected geographical indication region of Marostica (Vicenza, Italy) during two growing seasons. Multiple targeted and untargeted metabolomics approaches (NMR, LC-MS, HPLC-DAD, HPLC-UV) as well as artificial simplified phytocomplexes representing the cultivars Sandra Tardiva, Sandra and Grace Star were then used to determine whether the total antioxidant activity reflected the additive effects of each compound or resulted from synergis…

Transplantation of ACE2- Mesenchymal Stem Cells Improves the Outcome of Patients with COVID-19 Pneumonia

2020

A coronavirus (HCoV-19) has caused the novel coronavirus disease (COVID-19) outbreak in Wuhan, China. Preventing and reversing the cytokine storm may be the key to save the patients with severe COVID-19 pneumonia. Mesenchymal stem cells (MSCs) have been shown to possess a comprehensive powerful immunomodulatory function. This study aims to investigate whether MSC transplantation improves the outcome of 7 enrolled patients with COVID-19 pneumonia in Beijing YouAn Hospital, China, from Jan 23, 2020 to Feb 16, 2020. The clinical outcomes, as well as changes of inflammatory and immune function levels and adverse effects of 7 enrolled patients were assessed for 14 days after MSC injection. MSCs …

Validity and reliability of the CatWalk system as a static and dynamic gait analysis tool for the assessment of functional nerve recovery in small an…

2017

Introduction: A range of behavioral testing paradigms have been developed for the research of central and peripheral nerve injuries with the help of small animal models. Following any nerve repair strategy, improved functional outcome may be the most important evidence of axon regeneration. A novel automated gait analysis system, the CatWalk™, can measure dynamic as well as static gait patterns of small animals. Of most interest in detecting functional recovery are in particular dynamic gait parameters, coordination measures, and the intensity of the animals paw prints. This article is designed to lead to a more efficient choice of CatWalk parameters in future studies concerning the functio…

Supporting recovery from brain injury

2018

The beauty and intricacy of the human brain is unfortunately also mirrored by its vulnerability. Damage to the brain is typically permanent. Because cells of the adult brain, apart from rare exceptions, no longer divide, there is essentially no regrowth of damaged brain tissue. Acquired brain injury in the majority of cases occurs directly through traumatic events such as an accident involving a blow to the head or indirectly through interruption of the blood supply, namely a stroke. Brain injury is a major burden, with an estimated 1.7 million people in the United States suffering a traumatic brain injury and nearly 800,000 Americans suffering a stroke each year ( 1 , 2 ). However, current…

Elements of biomechanics correlated to the body mass index in knee osteoarthritis

2020

Introduction. Osteoarthritis is considered a complex biopsychosocial condition, with consequences for the patient, the family, society and public health. Age is considered the main risk factor in the occurrence of osteoarthritis. Studies show that 2 out of 3 obese individuals associate knee osteoarthritis whereas the incidence of the latter increases at the same time with increasing body mass index. Obesity contributes to the initiation of the process of osteoarthritis by the mechanical overload of the joint, causing cartilage damage with increasing horizontal cracks and the increased incidence of the osteophites in the knee. Purpose. We conducted a study in patients with knee osteoarthriti…

The quality of life - an indicator for assessing the recovery program in patients diagnosed with degenerative disorders

2020

Introduction. Osteoarthritis is a chronic joint disease, a progressive non-inflammatory arthropathy-type. Globally, around 1.7 billion people are affected by musculoskeletal and rheumatic diseases, which indicates an increase in the last 20 years of around 45%. Material and method. The objectives of the treatment of patients diagnosed with osteoarthritis were: decreasing the pain, increasing joint mobility, muscle strength, trophic and muscle tone, increasing the quality of life and reintegration into the social and family environment. Demographic data and the functional status were assessed using the VAS scale, the WOMAC scale, the QOL scale. Results and discussions. The pain, the most imp…

Droplet microfluidics is a technology that enables the production and manipulation of small volumes. In biosciences, the most popular application of this technology is Droplet Digital™ PCR (ddPCR™), where parallel nanoliter-scale PCR assays are used to provide a high sensitivity and specificity for DNA detection. However, the recovery of PCR products for downstream applications such as sequencing can be challenging due to the droplets' stability. Here we compared five methods for disrupting the droplets to recover DNA. We found that rapid freezing in liquid nitrogen results in a clear phase separation and recovery of up to 70% of the DNA content. Liquid nitrogen freezing can thus offer a s…
