Semiquantitative bioluminescent assay of glutathione

A novel technique has been developed for semiquantitative detection of glutathione (GSH) in small volumes of liquid samples. GSH is detected via enzymatic linkage to the NADP/NADPH + H+ redox system through glutathione reductase. Accumulated NADPH is measured via the bioluminescent FMN oxidoreductase bacterial luciferase reaction. A linear correlation is obtained between bioluminescence intensity of the luciferase reaction and the GSH content of the liquid sample. Possible applications of this procedure are discussed. © 1998 John Wiley & Sons, Ltd.

Synthesis and Photoluminescent Properties of 1,1‘-Binaphthyl-Based Chiral Phenylenevinylene Dendrimers

New chiral, soluble binaphthyl derivatives that incorporate stilbenoid dendrons at the 6,6′-positions have been prepared. The synthesis of the new enantiopure dendrimers was performed in a convergent manner by Horner-Wadsworth-Emmons (HWE) reaction of the appropriately functionalized 1,1′-binaphthyl derivative (R)-1 and the appropriate dendrons (R)2nGn-CHO. Different electroactive units were incorporated in the peripheral positions of the dendrons in order to tune both the optical and electrochemical behavior of these systems. Thioredoxin-related protein of 14 kDa may directly reduce protein cysteinylation motifs

Disulfide stress has been associated with inflammation and characterized by an increase in cystine levels and protein cysteinylation. Furthermore, it was recently discovered that thioredoxin-related protein of 14 kDa (TRP14, encoded by TXNDC17) exhibits efficient cystine reductase activity. The aim of our research was to elucidate if TRP14 is also able to reduce cysteinylated proteins in mammalian cells. Thus, protein cysteinylation was assessed in control and TRP14 knockdown cells in vitro through their pre-treatment with 25 µg/ml cycloheximide for 30 min and incubation with 250 µM biotinylated cysteine for 1 h. Towards Multifunctional Materials Incorporating Elastomers and Reversible Redox-Active Fragments

This paper presents a novel and unique feature of metallacarboranes, consisting of the linkage of this redox electro-active site to a stretchable polymer. This is based on polyTHF, a known and applied material. This hybrid material has the two ends functionalized: one with the aforemen- tioned redox molecule and the other with a terminal OH group, both linked by a molecular spring. ChemInform Abstract: New Strategy for the Stereoselective Synthesis of Fluorinated β-Amino Acids.

Racemic and chiral nonracemic α-substituted and α-unsubstituted β-fluoroalkyl β-amino acid derivatives 6 and 9 have been synthesized in two steps starting from fluorinated imidoyl chlorides 1 and ester enolates. This approach is based on the chemical reduction of previously obtained γ-fluorinated β-enamino esters 4 by using ZnI2/NaBH4 in a nonchelated aprotic medium (dry CH2Cl2) as the reducing agent. A metal-chelated six-membered model has been suggested to explain the stereochemical outcome of the reduction reaction. New ferrocene-containing copolyesters

We have synthesized four new ferrocene monomers (three diols and one diester). The redox potential of these ferrocene derivatives varies between 0 mV and 230 mV due to different degrees of ring alkylation. Amorphous and liquid crystalline copolyesters were prepared with these monomers in the polymer main chain. Cyclovoltammetric measurements show that the redox potential of the ferrocene units is increased by about 40 mV upon polymer formation (esterification). Since the ester group is 4 to 6 σ-bonds away from the ferrocene unit this increase is probably caused by some charge-transfer interaction through space. Redox Materials by the Covalent Entrapment of Redox-Active Dirhodium(II,II) Species in a Siloxane Network

Hydrolysis and polycondensation of the coupling agent (aminopropyl)triethoxysilane (APS), axially coordinated to the redox-active complex [Rh 2 (form) 2 (CH 3 COO) 2 -(APS) 2 ], lead to the insertion of redox-active inorganic microdomains into a siloxane network; the new polymers undergo cyclic redox reactions indicating that dirhodium(II,II) centres retain their redox activity even when incorporated into siloxane networks.

Characterization of poly(N-alkylanilines) by Raman spectroscopy

Abstract Thin films of poly( N -alkylaniline) were synthesized in acidic aqueous solution and in mixtures of aqueous and organic solvents. The polymer films (alkyl = methyl, ethyl, propyl and butyl) were characterized by Raman spectroscopy with the excitation wavelengths of 514.5, 632.8 and 780 nm. The main Raman bands have been characterized for the leucoemeraldine, emeraldine and pernigraniline oxidation states between −0.2 and 0.8 V (vs. Ag|AgCl). This fundamental study shows that the structure of the half-oxidized emeraldine form contains quinoid units, which supports the commonly accepted oxidation and reduction scheme of poly( N -alkylanilines).

Structure and function of ferredoxin-NADP+-oxidoreductase

The redox-enzyme ferredoxin-NADP-oxidoreductase has been shown to be activated by light and inactivated in the dark. This review will summarize recent data concerning the biochemical characterization of the enzyme compared to its in-vivo activation. Further-more the mechanism of this activation process is discussed as a conformational change caused by the light-driven proton gradient.
