4,5,6,7-Tetrahydro-isoxazolo-[4,5-c]-pyridines as a new class of cytotoxic Hsp90 inhibitors.

Hsp90 is considered an interesting therapeutic target for anticancer drug development. Here we describe a new class of 4,5,6,7-tetrahydro-isoxazolo-[4,5-c]-pyridine compounds. A small library of derivatives has been synthesized and investigated. Some reported compounds show interesting properties combining both notable binding to Hsp90 and potent cell growth inhibitory activity. N-5 substitution with a 2,4 resorcinol carboxamide appears crucial for activity. Moreover, a derivative bearing a hydroxamic acid residue bound to C-3 amide portion was found to inhibit both Hsp90 and HDAC6.

Publisher Summary This chapter discusses the mononuclear heterocyclic rearrangements. The chapter discusses the ring conversion of type 1→2 characterized by nucleophilic attack from a Z atom on a sp 2 -nitrogen atom with displacement of an atom. On this basis, the known ring conversions are tabulated. Taking into account the reactivities observed in the rearrangements, results are presented for 1,2,4-oxadiazole, isoxazole, and 1,2,5-oxadiazole. The sequence involving an oxime group in the side chain is discussed. Some organo-Main-Group-IV-element compounds of steroids of the type R3M(COL), R2Sn(COL)2 and (R3M)2(HDCA) (where R = Me or Ph; M = Si, Sn and Pb; HCOL = cholest-5-en-3β-ol; H3DCA = 3α, 12α-dihydroxy-5β-cholan-24-oic acid), have been synthesized. The structures of these compounds have been investigated by vibrational, mass, NMR (1H, 13C, 29Si, 119Sn and 207Pb) and 119Sn Mossbauer spectroscopy. For Me3Sn(COL) both in solution and in the solid state an associated structure with distorted trigonal bipyramidal geometry around tin and three-coordinated oxygen atoms is postulated. Abstract The Borexino detector has convincingly shown its outstanding performance in the in the sub-MeV regime through its unprecedented accomplishments in the solar and geo-neutrinos detection, which make it the ideal tool to unambiguously test the long-standing issue of the existence of a sterile neutrino, as suggested by several anomalies: the outputs of the LSND and Miniboone experiments, the results of the source calibration of the two Gallium solar ν experiments, and the recently hinted reactor anomaly. In a search for retinoic acid (RA) receptor ligands endowed with potent apoptotic activity, a series of novel arotinoids were prepared. Because the stereochemistry of the C9-alkenyl portion of natural 9-cis-RA and the olefinic moiety of the previously synthesized isoxazole retinoid 4 seems to have particular importance for their apoptotic activity, novel retinoid analogues with a restricted or, vice versa, a larger flexibility in this region were designed and prepared. The new compounds were evaluated in vitro for their ability to activate natural retinoid receptors and for their differentiation-inducing activity. High sensitivity of northern pike larvae to UV-B but no UV-photoinduced toxicity of retene

In order to investigate whether increased UV-B radiation is a risk factor, a series of acute laboratory experiments was conducted with larval stages of the northern pike (Esox lucius L.), hatching in Nordic waters in May. Further, a comparative investigation on the acute phototoxicity of retene (7-isopropyl-1-methylphenanthrene), a PAH compound recently revealed to posses UV-B-induced phototoxicity in larval coregonids, was conducted with pike larvae. Leaching of glyphosate and AMPA under two soil management practices in Burgundy vineyards (Vosne-Romanée, 21-France)

Some drinking water reservoirs under the vineyards of Burgundy are contaminated with herbicides. Thus the effectiveness of alternative soil management practices, such as grass cover, for reducing the leaching of glyphosate and its metabolite, AMPA, through soils was studied. The leaching of both molecules was studied in structured soil columns under outdoor conditions for 1 year. The soil was managed under two vineyard soil practices: a chemically treated bare calcosol, and a vegetated calcosol.
