Spatial analysis of plant metabolism: Sucrose imaging within Vicia faba cotyledons reveals specific developmental patterns

During legume embryogenesis the differentiation of the cotyledons proceeds gradually in a wave-like manner. The process is metabolically and genetically controlled and regulated by sugars. In order to perform a spatial and temporal analysis of the sugar distribution pattern a new method was developed to specifically measure sucrose directly in tissues via bioluminescence and single photon counting. This enabled a quantitative sucrose imaging with a resolution close to the single cell level. Exopolysaccharide produced by Weissella confusa: Chemical characterisation, rheology and bioactivity

Abstract The purpose of this study was to characterise the production of exopolysaccharide (EPS) by indigenous lactic acid bacteria (LAB) isolated from traditional Algerian dairy products and to evaluate their possible use in agri-foods. Among the collection of isolated strains, the strain Weissella confusa (W4) was selected for its ability to produce EPS once exposed to a sucrose culture medium. Studies on sea urchin oocytes. II. Synthesis of RNA during oogenesis.

Abstract Isolated oocytes of the sea urchin Paracentrotus lividus actively incorporate 3H-uridine into RNA. Labeled RNA was analysed by sucrose gradient and acrylamide gel electrophoresis following cell fractionation. Much of the radioactivity is incorporated at the nucleolar level in the form of rRNA precursors. The kinetics of maturation of these latter suggests that this occurs at a slower rate than during embryogenesis. Other non-nucleolar RNA classes are also actively labelled and retained in the nucleus for many hours. These results are confirmed by an autoradiographic investigation.

The triplet state of positronium, o-Ps, is used as a probe to characterize a starch-20% w/w sucrose matrix as a function of temperature (T). A two-step decomposition (of sucrose, and then starch) starts at 440 K as shown by a decrease in the o-Ps intensity (I(3)) and lifetime (τ(3)), the latter also disclosing the occurrence of a glass transition. Upon sucrose decomposition, the matrix acquires properties (reduced size and density of nanoholes) that are different from those of pure starch. A model is successfully established, describing the variations of both I(3) and τ(3) with T and yields a glass transition temperature, T(g) = (446 ± 2) K, in spite of the concomitant sucrose decomposition…

Echovirus 1 Endocytosis into Caveosomes Requires Lipid Rafts, Dynamin II, and Signaling EventsV⃞

Binding of echovirus 1 (EV1, a nonenveloped RNA virus) to the α2β1 integrin on the cell surface is followed by endocytic internalization of the virus together with the receptor. Here, video-enhanced live microscopy revealed the rapid uptake of fluorescently labeled EV1 into mobile, intracellular structures, positive for green fluorescent protein-tagged caveolin-1. Partial colocalization of EV1 with SV40 (SV40) and cholera toxin, known to traffic via caveosomes, demonstrated that the vesicles were caveosomes. Fructooligosaccharide and levan producing activity of Zymomonas mobilis extracellular levansucrase

Abstract The present work was devoted to investigations of the fructooligosaccharide (FOS) and levan forming activity of ethanol producing bacteria Zymomonas mobilis and their extracellular levansucrase. After cell separation the culture liquid was treated with ethanol to precipitate levan together with extracellular levansucrase. Levan–levansucrase sediment can be used as biocatalyst for fructooligosaccharide (FOS) production in sucrose syrup and levan sediment as soluble fibre source. Two Methods for Highlighting the Influence of Sugars Type on Fermentation Processes Produced by Staphylococcus Carnosus and Lactobacillus Curvatus

Purpose of this paper is to establish the influence of different types of sugars on fermentation processes found in the meat industry. In the analysis used a mixture of bacteria (Lactobacillus curvatus and Staphylococcus carnosus ), and the types of sugars were used lactose, lactulose, sucrose and two types of glucose syrup. To highlight the speed with which the fermentation process takes place, the variation in pH for the environment and the amount of energy released during biotechnological processes was measured.

In peach (Prunus persica [L.] Batsch.), sorbitol and sucrose are the two main forms of photosynthetic and translocated carbon and may have different functions depending on the organ of utilization and its developmental stage. The role and interaction of sorbitol and sucrose metabolism was studied in mature leaves (source) and shoot tips (sinks) of'Nemaguard' peach under drought stress. Influence of Microspore Development on Pollen Protoplast Isolation in Digitalis obscura

Summary Protocols for the enzymatic isolation of Digitalis obscura pollen protoplasts are described. The developmental stage of the microspores was a critical factor for successful isolation since protoplasts were obtained only from tetrads. The highest yield of viable protoplasts was achieved with an enzymatic mixture containing helicase, cellulase and pectolyase in 0.5 M sucrose. In different culture techniques employed, cell wall regeneration took place only when protoplasts were plated in agarose drops.

Hormone directed sucrose transport during fruit set induced by gibberellins in Pisum sativum

A new system has been developed to study hormone-directed transport in intact plants during parthenocarpic fruit set induced by gibberellins. Gibberellic acid (GA3) and gibberellin A1 (GA1) applied to unpollinated ovaries of pea (Pisum sativum L. cv. Alaska) promoted sucrose transport from the leaf to the site of hormone application. In vivo experiments showed an early (30 min) accumulation of [14C]-sucrose in ovaries of pea stimulated by gibberellins. This activation of sucrose transport appears to be mediated by gibberellins (GA1, GA3), increasing both loading of phloem with sucrose in the leaf (source) and sucrose unloading in the ovary (sink).
