Quantitative and Qualitative in vivo Analysis of Metal Residuals in the Saliva of Subjects with and without Fixed Orthodontic Appliances
2020
Aim of the Work: The aim of this work was to evaluate Ni e Cr ions in the saliva of subjects with and without fixed orthodontic appliances. Materials and Methods: This was a prospective, non-randomized study of 100 patients, so selected: 50 have undergone fixed orthodontic appliance at least from 24 months; others 50 subjects as control-group, without any fixed and/or removable orthodontic appliance It was conducted according to the Declaration of the World Medical Association of Helsinki. The two selected groups were compatible for age, sex, absence of restoration in amalgam or other metals in the oral cavity. The saliva of all the subjects was collected with standardized sterile methods. …
Super-resolution binding activated localization microscopy through reversible change of DNA conformation
2018
ABSTRACT Methods of super-resolving light microscopy (SRM) have found an exponentially growing range of applications in cell biology, including nuclear structure analyses. Recent developments have proven that Single Molecule Localization Microscopy (SMLM), a type of SRM, is particularly useful for enhanced spatial analysis of the cell nucleus due to its highest resolving capability combined with very specific fluorescent labeling. In this commentary we offer a brief review of the latest methodological development in the field of SMLM of chromatin designated DNA Structure Fluctuation Assisted Binding Activated Localization Microscopy (abbreviated as fBALM) as well as its potential future app…
Fast UPLC/PDA determination of squalene in Sicilian P.D.O. pistachio from Bronte: Optimization of oil extraction method and analytical characterizati…
2017
Abstract A fast reversed-phase UPLC method was developed for squalene determination in Sicilian pistachio samples that entry in the European register of the products with P.D.O. In the present study the SPE procedure was optimized for the squalene extraction prior to the UPLC/PDA analysis. The precision of the full analytical procedure was satisfactory and the mean recoveries were 92.8 ± 0.3% and 96.6 ± 0.1% for 25 and 50 mg L−1 level of addition, respectively. Selected chromatographic conditions allowed a very fast squalene determination; in fact it was well separated in ∼0.54 min with good resolution. Squalene was detected in all the pistachio samples analyzed and the levels ranged from 5…
On the Antibacterial Activity of Azacarboxylate Ligands: Lowered Metal Ion Affinities for Bis-amide Derivatives of EDTA do not mean Reduced Activity.
2018
EDTA is widely used as an inhibitor of bacterial growth, affecting the uptake and control of metal ions by microorganisms. We describe the synthesis and characterisation of two symmetrical bis-amide derivatives of EDTA, featuring glycyl or pyridyl substituents: AmGly2 and AmPy2 . Metal ion affinities (logK) have been evaluated for a range of metals (Mg2+ , Ca2+ , Fe3+ , Mn2+ , Zn2+ ), revealing less avid binding compared to EDTA. The solid-state structures of AmGly2 and of its Mg2+ complex have been determined crystallographically. The latter shows an unusual 7-coordinate, capped octahedral Mg2+ centre. The antibacterial activities of the two ligands and of EDTA have been evaluated against …
Metal Ions and Metal Complexes in Alzheimer's Disease.
2015
Background: Alzheimer’s disease (AD) is the most common form of dementia that seriously affects daily life. Even if AD pathogenesis is still subject of debate, it is generally accepted that cerebral cortex plaques formed by aggregated amyloid-β (Aβ) peptides can be considered a characteristic pathological hallmark. It is well known that metal ions play an important role in the aggregation process of Aβ. Methods: This review focuses on the anti-Aβ aggregation activity of chelating ligands as well as on the use of metal complexes as diagnostic probes and as potential drugs. Conclusion: While chelating agents, such as curcumin or flavonoid derivatives, are currently used to capture metal ions …
Monitoring few molecular binding events in scalable confined aqueous compartments by raster image correlation spectroscopy (CADRICS)
2016
The assembly of scalable liquid compartments for binding assays in array formats constitutes a topic of fundamental importance in life sciences. This challenge can be addressed by mimicking the structure of cellular compartments with biological native conditions. Here, inkjet printing is employed to develop up to hundreds of picoliter aqueous droplet arrays stabilized by oil-confinement with mild surfactants (Tween-20). The aqueous environments constitute specialized compartments in which biomolecules may exploit their function and a wide range of molecular interactions can be quantitatively investigated. Raster Image Correlation Spectroscopy (RICS) is employed to monitor in each compartmen…
Correlation between octopaminergic signalling and foraging task specialisation in honeybees
2020
Regulation of pollen and nectar foraging in honeybees is linked to differences in the sensitivity to the reward. Octopamine (OA) participates in the processing of reward-related information in the bee brain, being a candidate to mediate and modulate the division of labour among pollen and nectar foragers. Here we tested the hypothesis that OA affects the resource preferences of foragers. We first investigated whether oral administration of OA is involved in the transition from nectar to pollen foraging. We quantified the percentage of OA-treated bees that switched from a sucrose solution to a pollen feeder when the sugar concentration was decreased experimentally. We also evaluated if feedi…
Characterizing microstructural tissue properties in multiple sclerosis with diffusion MRI at 7 T and 3 T: The impact of the experimental design
2019
The recent introduction of advanced magnetic resonance (MR) imaging techniques to characterize focal and global degeneration in multiple sclerosis (MS), like the Composite Hindered and Restricted Model of Diffusion, or CHARMED, diffusional kurtosis imaging (DKI) and Neurite Orientation Dispersion and Density Imaging (NODDI) made available new tools to image axonal pathology non-invasively in vivo. These methods already showed greater sensitivity and specificity compared to conventional diffusion tensor-based metrics (e.g., fractional anisotropy), overcoming some of its limitations. While previous studies uncovered global and focal axonal degeneration in MS patients compared to healthy contr…
A Single-sensor High-resolution Panoramic Optical Mapping Configuration for Simultaneous Non-overlapped Complete Atrial and Ventricular Parametric Im…
2017
Nowadays optical mapping (OM) is the primary method for imaging electrophysiologically relevant parameters from the outer surface of Langendorff-perfused hearts. This technique has become essential for comprehensively understanding mechanisms of cardiac propagation during physiological activation, arrhythmia, and therapeutic antiarrhythmic interventions in translational hearts. Panoramic whole heart optical mapping systems, using either multiple cameras, plane mirrors or a combination of both, have been developed to overcome intrinsic visualization limitations to traditional single-sensor designs. However current panoramic OM systems are financially challenging for physiology and engineerin…
Dual-mode holographic microscopy imaging platform
2018
We report on a novel layout capable of dual-mode imaging in real time with different magnifications and resolution capabilities in lensless microscopy. The concept is based on wavelength multiplexing for providing two illuminations with different wavefront curvatures: one is collimated, allowing a large field of view (FOV) with a poor resolution limit, and the other is divergent, to achieve a better resolution limit (micron range) over a small FOV. Moreover, our recently reported concept of MISHELF microscopy [M. Sanz, J. Á. Picazo-Bueno, L. Granero, J. García and V. Micó, Sci. Rep., 2017, 7, 43291] is applied to the divergent illumination case, improving the image quality by noise averagin…
