LS104, a non-ATP-competitive small-molecule inhibitor of JAK2, is potently inducing apoptosis in JAK2V617F-positive cells
2008
Abstract The activating JAK2V617F mutation has been described in the majority of patients with BCR-ABL-negative myeloproliferative disorders (MPD). In this report, we characterize the small-molecule LS104 as a novel non-ATP-competitive JAK2 inhibitor: Treatment of JAK2V617F-positive cells with LS104 resulted in dose-dependent induction of apoptosis and inhibition of JAK2 autophosphorylation and of downstream targets. Activation of these targets by JAK2 was confirmed in experiments using small interfering RNA. LS104 inhibited JAK2 kinase activity in vitro. This effect was not reversible using elevated ATP concentrations, whereas variation of the kinase substrate peptide led to modulation of …
An automated and self-cleaning nano liquid chromatography mass spectrometry platform featuring an open tubular multi-hole crystal fiber solid phase e…
2017
An open tubular (OT) sample preparation/separation platform was developed. A multi-channel polymer layer open tubular (mPLOT) solid phase extraction (SPE) column was prepared by wall-coating the 126 channels (8μm inner diameter (ID) each) of a crystal fiber capillary with an organic polymer, namely poly(styrene-co-octadecene-co-divinylbenzene) (PS-OD-DVB). The mPLOT SPE was coupled on-line with a 10μm×2m poly(styrene-co-divinylbenzene) (PS-DVB) OT liquid chromatography column with nanospray mass spectrometry (OTLC-MS). Compared to using monolithic/particle-packed SPEs, mPLOT-SPE-OTLC allowed both fast loading and sufficient refocusing on the OT analytical column of small model compounds (su…
Influence of oscillating main flow on separation efficiency in asymmetrical flow field-flow fractionation.
2021
The steadily rising interest in the investigation of interactions between nanomaterials and biological media has also led to an increasing interest in asymmetrical flow field-flow fractionation (AF-FFF). The biggest strength of AF-FFF is the possibility to alter the flow profiles to suit a specific separation problem. In this paper, the influence of an oscillating main flow on the separation efficiency of AF-FFF is investigated. Such oscillations can e.g. be caused by the main pump To investigate the influence of such flow conditions on the separation efficiency in AF-FFF systematically, different oscillation profiles were applied and their influence on the elution profile and the retention…
Enantio- and diastereocontrol in intermolecular cyclopropanation reaction of styrene catalyzed by dirhodium(II) complexes with bulky ortho-metalated …
2004
Enantiomerically pure dirhodium(II) complexes with ortho-metalated p-substituted aryl phosphines have been shown to be enantio- and diastereoselective in the cyclopropanation of styrene by ethyl diazoacetate. Enantioselectivities up to 91% and diastereoselectivities up to 90% are observed for ethyl cis-2-phenylcyclopropanecarboxylate. Abstract LiChrolut EN [poly(styrene-divinylbenzene), PSDVB], Carbograph (graphitized carbon black, GCB), Isolute CN (cyanopropylsilica), Isolute C 2 (ethylsilica), and LiChrospher RP18 (octadecylsilica) were studied for on-line solid-phase extraction of twelve nitrogen containing pesticides from water. Determination (UV 210 nm) was performed with a Spherisorb C 8 analytical column and an acetonitrile-water gradient. The mean recoveries from 50 ml ranged from 83% for RP18 to 44% for GCB, and decreased in the following order: RP18, CN, LiChrolut EN, C 2 , and GCB. GCB showed poor recoveries due to incomplete desorption with the acetonitrile-water gradient used, and memory effects were detecte…
Electrophoretic flow behaviour and mobility of colloidal fluids and crystals.
2007
We report on measurements of the electrophoretic mobility mu of charged colloidal spheres in the deionized state, where the suspensions show fluid or crystalline order. In the fluid state, parabolic flow profiles are observed due to electro-osmotic solvent flow. In the crystalline state, complex flow profiles occur due to additional crystal cohesion. The mobility mu then may inferred from the flow velocity averaged over the complete cell cross section as performed in our home built super-heterodyne Doppler velocimeter. For two particle species of 68 and 122 nm diameter we measured mu as a function of particle concentration. Starting from a plateau value at low concentration, mu decreases ap…
Time-dependent effects of polystyrene nanoparticles in brine shrimp Artemia franciscana at physiological, biochemical and molecular levels
2019
Micro- (<5 mm) and nanoplastics (<1 μm) are emerging threats for marine ecosystems worldwide. Brine shrimp Artemia is recognized as a suitable model among planktonic species for studying the impact of polystyrene nanoparticles (PS NPs) through short and long-term bioassays. Our study aims to evaluate the time-dependent effects of cationic amino-modified PS-NH (50 nm) in A. franciscana after short- (48 h) and long-term exposure (14 days). For this purpose, nauplii were exposed to a concentration range of PS-NH (0.1, 1, 3 and 10 μg/mL) in natural sea water (NSW), and physiological, biochemical and molecular responses were investigated. Short-term exposure to PS-NH caused a decrease in nauplii…
Antibodies against homogeneous epoxide hydratase provide evidence for a single enzyme hydrating styrene oxide and benz(a)pyrene 4,5-oxide
1976
THE microsomal enzyme epoxide hydratase (EC 4.2.1.63) is potentially important in the inactivation of metabolically produced epoxides which may be responsible for the mutagenic and/or carcinogenic properties of polycyclic hydrocarbons (for reviews see refs 1–3). Reports4,5 suggest that the enzyme plays a dual role in (a) producing proximate carcinogens which, after biotransformation to carcinogens by microsomal mono-oxygenase(s) are (b) inactivated by epoxide hydratase. As this enzyme can be induced6–8, activated9–10 and inhibited9–13 it should be useful in studies of the mechanism of chemical carcinogenesis: some inverse correlations have been reported between susceptibility to carcinogene…
Influence of the level of cytosolic epoxide hydrolase on the induction of sister chromatid exchanges by trans-beta-ethylstyrene 7,8-oxide in human ly…
1991
Abstract trans -β-Ethylstyrene 7,8-oxide, a substrate of cytosolic epoxide hydrolase, and 4-fluorochalcone oxide, an inhibitor of this enzyme, were investigated on induction of sister chromatid exchanges (SCE) in human lymphocytes. Both epoxides enhanced the frequency of SCE. 4-Fluorochalcone oxide at low concentration (2.5μM) inhibited cytosolic epoxide hydrolase activity towards trans -β-ethylstyrene 7,8-oxide in lymphocytes by 74% and had no effect on glutathione transferase activity using this substrate. At this concentration it did not induce SCE itself, but it potentiated the effect of trans -β-ethylstyrene 7,8-oxide several fold. In lymphocytes from different subjects, the number of …
Polystyrene nanoparticles in the presence of (ethylene oxide)13(propylene oxide)30(ethylene oxide)13, N,N-dimethyloctylamine-N-oxide and their mixtur…
2008
Polystyrene nanoparticles were synthesized by emulsion polymerization of styrene. They were functionalized using the conventional surfactant N,N-dimethyloctylamine-N-oxide (ODAO), the tri-block copolymer (ethylene oxide)13(propylene oxide)30(ethylene oxide)13 (L64) and their mixtures. To this purpose, dynamic light scattering and calorimetric experiments were carried out and provided information consistent to each other. The L64 adsorption is Langmuir-type in the copolymer dilute regime and generates complex structures at larger concentrations. In the region where ODAO is in the unimeric state, the adsorption process is cooperative leading to hemi-micelle formation at the polystyrene nanopa…
