Detailed characterization of human Mycobacterium tuberculosis specific HLA-E restricted CD8+TÂ cells
2018
HLA-E presented antigens are interesting targets for vaccination given HLA-Esâ essentially monomorphic nature. We have shown previously that Mycobacterium tuberculosis (Mtb) peptides are presented by HLA-E to CD8+effector TÂ cells, but the precise phenotype and functional capacity of these cells remains poorly characterized. We have developed and utilized in this study a new protocol combining HLA-E tetramer with intracellular staining for cytokines, transcription factors and cytotoxic molecules to characterize these cells in depth. We confirm in this study the significantly increased ex vivo frequency of Mtb-peptide/HLA-E-TM+CD8+TÂ cells in the circulation of patients with active tubercu…
The influence of the Covid-19 pandemic on Czech-Polish cross-border cooperation: From debordering to re-bordering?
2021
Abstract The COVID-19 pandemic brought many changes to social behaviours in Europe. One of its major consequences was the temporary closure of borders, which was introduced as a measure to prevent the uncontrolled pandemic spreading and involved internal Schengen borders. This has had a major impact in the way in which cross-border cooperation has been conducted in Europe, including the Czech-Polish borderland, as it dramatically restrained all flows across borders. In this paper, we evaluate the impact of the pandemic on five roles of cross-border cooperation: 1) as a multi-level governance form; 2) as a regional development tool; 3) as a para-diplomacy form; 4) as a post-conflict reconcil…
Kaitējuma atlīdzināšanas problemātika sauszemes transportlīdzekļu īpašnieku civiltiesiskās atbildības obligātajā apdrošināšanā
2015
Pētījuma mērķis ir parādīt šī brīža aktuālākās problēmas kaitējumu atlīdzināšanā sauszemes transportlīdzekļu tiesīgo lietotāju obligātajā civiltiesiskajā apdrošināšanā. Darbā tiek konstatētas problēmas spēkā esošo tiesību normu pielietojumā reālās situācijās, konstatējot transportlīdzekļu tiesīgo lietotāju atbildības pakāpi un atlīdzinot radīto kaitējumu trešajām personām. Tāpat tiek analizētas Latvijas tiesību normu savstarpējās pretrunas, kā arī to atbilstība Eiropas Savienības tiesību normām un Eiropas Savienības Tiesu nolēmumiem. Ņemot vērā Eiropas Savienības tiesību avotu ietekmi uz Latvijas tiesību normām, mēģināts sniegt iespējamos risinājumus, kas izpaužas gan interpretācijā, gan iz…
Thoughts on What Chemists Can Contribute to Fighting SARS-CoV-2 - A Short Note on Hand Sanitizers, Drug Candidates and Outreach.
2020
Abstract The SARS‐CoV‐2 outbreak causing the respiratory disease COVID‐19 has left many chemists in academia without an obvious option to contribute to fighting the pandemic. Some of our recent experiences indicate that there are ways to overcome this dilemma. A three‐pronged approach is proposed.
Geographical and temporal structures of Legionella pneumophila sequence types in comunitat valenciana (Spain), 1998 to 2013
2015
ABSTRACT Legionella pneumophila is an accidental human pathogen associated with aerosol formation in water-related sources. High recombination rates make Legionella populations genetically diverse, and nearly 2,000 different sequence types (STs) have been described to date for this environmental pathogen. The spatial distribution of STs is extremely heterogeneous, with some variants being present worldwide and others being detected at only a local scale. Similarly, some STs have been associated with disease outbreaks, such as ST578 or ST23. Spain is among the European countries with the highest incidences of reported legionellosis cases, and specifically, Comunitat Valenciana (CV) is the se…
Impact of biocide treatments on the bacterial communities of the Lascaux Cave.
2009
The Lascaux Cave contains a remarkable set of paintings from the Upper Palaeolithic. Shortly after discovery in 1940, the cave was modified for public viewing and, in 2001, was invaded by a Fusarium solani species complex. Benzalkonium chloride was used from 2001 to 2004 to eliminate the fungal outbreak. In this study, we carried out a sampling in most of the cave halls and galleries. Sequence analysis and isolation methods detected that the most abundant genera of bacteria were Ralstonia and Pseudomonas. We suggest that, as a result of years of benzalkonium chloride treatments, the indigenous microbial community has been replaced by microbial populations selected by biocide application.
Molecular relationship among Salmonella dublin isolates identified at the Center for Enterobacteriaceae of Palermo during the years 1971-85.
1987
SUMMARYA molecular epidemiological study was carried out on 60Salmonella dublinisolates identified at the Southern Italy Enterobacteriaceae Center between 1971 and 1985. These included 23 isolates from children with diarrhoea in Palermo obtained during 1984.All isolates from the outbreak of gastroenteritis in children were resistant to chloramphenicol and streptomycin and harboured two plasmids of 50 MDa and 3 MDa molecular weight, whereas the majority of the isolates identified before 1984 were susceptible to these antibiotics and carried only a 50 MDa molecular weight plasmid. FourS. dublinstrains successively identified from cattle (Palermo, Foggia, Portici) and from a child (Palermo) we…
rRNA probing of chromosomal DNA of epidemic and sporadic isolates of Salmonella enterica subsp. Enterica serovar kottbus from Northern and Southern I…
1990
Fifty-two strains of Salmonella enterica subsp. enterica serovar Kottbus, identified at the Centres of Enterobacteriaceae of Northern and Southern Italy, were investigated by molecular genetic methods. Thirteen isolates were recovered during two food-poisoning outbreaks that occurred in May 1987 in Lombardy. The rDNA gene restriction patterns, obtained by probing endonuclease cleaved chromosomal DNA with photobiotin labeled Escherichia coli rRNA, revealed some heterogeneity among strains isolated from Southern Italy, whereas Northern Italy isolates exhibited virtually identical banding patterns.
rDNA fingerprinting as a tool in epidemiological analysis of Salmonella typhi infections
1991
SUMMARYCharacterization of 169 strainsof Salmonella typhiof phage types C1, C4, D1and D9isolated in 1975–88 was carried out by rDNA gene restriction pattern analysis. Twenty-four isolates had been recovered during four large waterbone outbreaks in the last 20 years in Sicily; 145 strains, isolated from apparently sporadic cases of infection in Southern Italy in the same period of time, were also examined.Application of rRNA–DNA hybridization technique after digestion of chromosomal DNA withClaI showed the identity of patterns of the epidemic strains of phage types C1and D1, confirming attribution of the outbreaks to single bacterial clones. Patterns of the two available strains of lysotype …
Ongoing spread of colistin-resistant Klebsiella pneumoniae in different wards of an acute general hospital, Italy, June to December 2011.
2012
We describe polyclonal spread of colistin-resistant Klebsiella pneumoniae in an acute general hospital in Italy. Between June and December 2011, 58 colistin-resistant K. pneumoniae isolates were recovered from 28 patients admitted to different wards, but mainly in the intensive care units. All isolates were tested for drug susceptibility and the presence of beta-lactamase (bla) genes. Clonality was investigated by repetitive extragenic palindromic (rep)-PCR and multilocus sequence typing (MLST). Fifty-two isolates had minimum inhibitory concentrations (MICs) for colistin of 6-128 mg/L, carried blaKPC3 and were attributed to sequence type ST258. The remaining six isolates were susceptible to…
